Abstract

The purpose of this study was to examine the effect of fish silages, bakasang as an immunostimulant in eel (Anguilla marmorata) feed against Aeromonas hydrophila infection. Three hundred elvers with the size of + 10 cm taken from Freshwater Aquaculture Institute (BBAT) Tatelu. The elvers were placed into 4 glass aquarium, each equipped with aerator and 1 for stock. Prior to the study, the eels were acclimatized for two weeks in order to adapt to new environmental condition. Two sets of researches were done: feeding the eels with additional bakasang with different doses for 6 weeks, which then further were challenged with Aeromonas hydrophila by immersion method. This study used a completely randomized design (CRD) with 4 treatments A (0 mL /kg diet), B (50 mL/kg diet), C (100 mL/kg diet), D (150 mL/kg diet). The growth data was taken by measuring the weight and length of the eels at the end of the period of feeding treatment while the mortality data of the eels were observed for 10 days after challenge test.  The results of analysis of variance showed that the resistance of the eels in treatment B (50 mL/kg diet), C (100 mL/kg diet) and D (150 mL/kg diet) significantly different compared with treatment A.  However, among the treatments B, C and D, there were no significant differences.  Furthermore, the best eel growth was observed in treatment B with an absolute growth of 1.14 g, followed by treatment of D with an absolute growth of 1.04 g. In treatment A the absolute growth was only 0.82 g while treatment C was 0. 88 g.  This suggests that the addition of bakasang in eels feed can promote the growth of eels as well as resistance against pathogen infection, thus served as immunostimulant. Abstract

Probiotic from carp intestines and its effects on increasing growth performance and feed efficiency had been investigated. Fish used were carp fingerlings weighing 2.33 g  in average. The fish were taken from the Tatelu Freshwater Aquaculture Fisheries Center.  Probiotics used were isolated from the intestines of carp. Before running the experiment, the fish were acclimatized for 10 days under laboratory conditions. During acclimatization the fish were fed commercial pellets without the addition of probiotics. The dose of feeding was 5% /bw/day, 2 times a day at 09:00 am and 16:00 pm. Furthermore, fish are stocked in 15 aquariums each measuring 60x40x40cm3 with a density of 20 fish/aquarium. Each aquarium was equipped with aeration and water pumps for recirculation. Uneaten feed and feces were removed through siphoning. Water exchange was done every 2-3 days.  After acclimatization, the fish were fed with pellet supplemented with probiotic at  concentration of A: without addition of probiotic; B: 1 x 106 cfu /mL; C: 1 x 107 cfu / mL; D: 1 x 108 cfu / mL; E: 1 x 109 cfu/mL. The fish were fed twice a day with feeding dose of 5% /bw/ day, for 21 days. Data collected consisted of daily growth rates, growth rates, feed efficiency and feed conversion ratio. The results of research showed that the addition of probiotics in feed had a very significant effect on growth, feed efficiency, and feed conversion ratio (P <0.01) with the best results achieved in administering probiotics as much as 1 x 108 cfu / mL. In conclusion, probiotic bacteria originating from the intestines of carp can be used to improve the growth performance of the fish itself.

Abstract

This study aimed to identify the type of pathogenic bacteria that attack tilapia (Oreochromis niloticus) cultivated in Dimembe District, North Minahasa Regency in 2019. The benefit of this research were to obtain information about the types of bacteria in tilapia (Oreochromis niloticus), so prevention efforts can be made or treatment of the disease. Tilapia as a research sample obtained from existing cultivation sites in the District of North Minahasa Dimembe, North Sulawesi Province. From each location 5-10 individuals were taken with a size of 5-12 cm. The samples were then taken to the laboratory for bacterial isolation. The media for growing bacteria was TSA (Tryptic Soya Agar). The results obtained were adjusted to the Bacterial Fish Pathogens identification book. The data obtained were analyzed descriptively through the presentation of tables and figures. Research results found the bacteria that infected tilapia in North Minahasa Regency including Aeromonas sp, Plesiomonas sp, Flavobacterium sp and Enterobacter sp. Regular and continuous monitoring of fish diseases is needed to monitor the spread of fish diseases, especially quarantine fish diseases in North Minahasa Regency.

Abstract

The research aimed to isolate probiotic bacteria from catfish intestine and assign the concentration of probiotic bacteria which effectively increase the activity of phagocytosis and total leukocytes of carp fingerlings. The fish was taken from Balai Pengembangan dan Pembinaan Pembudidayaan Ikan (BP3I) Tateli. Fish (average initial weight of 2.27 g) were acclimated for one week in 15 aquaria with a density of 25 fish/aquarium. One week after acclimatization, fish were fed with food added with probiotics with different concentrations, namely 1x106, 1x107, 1x108, 1x109 cfu/mL for four weeks. The dose of feed was 5% /body weight/day and feeding time was at 08.00 and 17.00. Data collected were phagocytosis and total leukocyte count. The results showed that supplementation of probiotic significantly increased phagocytic activity and total leukocytes (p<0.01). The best phagocytic activity and total leukocytes were achived in fish fed diet added with probiotics 1x108 cfu/mL. It can be concluded that supplementation of  probiotic onto feed can increase phagocytic activity and total leukocytes of carp.

Abstract

The objective of this study was to determine the minimum concentration of extract in inhibiting motile motile aeromonad septicaemia. The material used was the stems and leaves of the plant P. grandiflora as much as 10 g. The crushed simplicia was then macerated with aquades with a ratio of 1:10,  then filtered with filter paper and, put in an Erlenmeyer, and evaporated in an incubator at temperature of 30°C. The sensitivity test was performed by placing a paper disc containing extract on the agar medium that had been scratched with bacterial colonies. Clear zone formed around the paper disc was compared with standard classification table for clarifying the inhibition level.  The results showed that stem and leaf extract had inhibition zone against Aeromonas hydrophila bacteria that caused motile aeromonad septicaemia. The best result was obtain at concentration of extract 3.25 mg / L) with an average diameter of 22.67 mm and for leaf extract had a moderate to strong zone for all treatments. These proved that stem and leaf extract of P. grandiflora plant had antibacterial activity and can be used as control of MAS disease caused by A. hydrophila bacteria. Abstract

The aim of this research was to study the effect of Binahong extract on the growth of Nile tilapia. A number of 225 fish measuring 10-12 cm in length with an average weight of 15 g was obtained from Fishery Development Center and Training, Tateli. The fish were put  in oxygenated plastic bags and transported to Laboratory of Aquaculture Technology. The fish was kept in aquarium equipped with an aerator with a density of 15 fish per aquaria. The test material used was the stem of Binahong plant stem, macerated with alcohol 70% for 3 days. The extract was then filtered with whatman filter paper and extract was concentrated using a Rotary Evaporator. The extract was further diluted to obtain  concentrations of 12.5, 25, 50, and 100 mg / mL Binahong  extract as treatment was given to the fish after mixed first into the pellet. The fish was fed with treatment feed for 30 days. Control feed was not supplemented with binahong extract. Growth data was measured at day 30 after treatment. The results showed that the extract of binahong had significant effect on fish growth (p <0.01) with the best result achieved at concentration of 100 mg/mL. In conclusion, supplementation of  binahong extract into fish feed could improve growth of  fish in which the best growth was obtained in fish fed diet containing 100 mg/mL of binahong extract.Keywords: Medicinal plants, tilapia, Anredera cordifolia, growth, maceration

Abstract

This study aimed to evaluate the effect of probiotic on growth and survival of tilapia (Oreochromis niloticus) against Streptococcus agalactiae infection. The test materials used were commercial probiotic bacteria and Streptococcus agalactiae. The probiotic bacteria were administered by mixing into the feed with a dose of 0 mL (without probiotics), 10 mL / kg of feed, 15 mL / kg of feed and 20 mL / kg of feed.  Feed was given 3 times a day at 08.00, 12.00 and 16.00 for 21 days. After that, the challenge test was done with Streptococcus  agalactiae bacteria by cohabitation. Bacteria with a density of 107 cells / mL were mixed into the water and allowed for 2 hours. The result showed that B treatment (10 mL / kg of feed) gave the best result with 93,33 percent of  survival rate. Statistical analysis showed that the survival of fish in treatments B, C and D was significantly different from treatment A (P <0.1). However, there was no significant difference between treatments B, C, and D. It was also found that the addition of probiotic in feed did not affect the growth of Nile tilapia (Orechromis niloticus). Keywords : Probiotic bacteria,  nile tilapia,  Streptococcus  agalactiae, growth,  infection

Abstract

The aimed of the study was to isolate and select the probiotic Lactobacillus from carp intestine potential for use in aquaculture. Carp as a source of probiotics with a size of about 200 g was  taken from the Center for Fish Hatchery and Disease Control (BP3I), Tateli. After aseptic surgery, the intestine was separated and cleaned, weighed as much as 1 g, put in a mortar and crushed until smooth. The scoured results were put into a tube and added 9 ml NaCl to get a ratio of 1:10. The tube that already contains intestinal scouring was then centrifuged at a speed of 1000 RPM for 10 minutes. The centrifugation process was carried out 2 times to get the supernatant that has been cleared of debris. The supernatant was removed and diluted to obtain a concentration of 10-2  to 10-3. The solution was then spread on MRS (De Man Rogosa and Sharpe Agar) media and incubated at 28oC in the Incubator for 24-48 hours. The bacteria that grew in culture fmedia was reisolated several times to get a single pure colony.. The pure colonies were propagated in the same media to be further identified through gram staining, biochemical, proteolytic, amylolytic, cellulolytic, antibiotic resistance, antagonistic, pathogenicity tests. The results of the isolation obtained the characteristics of a colony of milky white color, round and convex with long cells, gram-positive and confirmed based on biochemical tests as Lactobaccilus sp. The bacterium also had proteolytic, amylolytic, cellulolytic, antagonistic, antibiotic-sensitive activity that were characterized by the formation of clear zones in the area around probiotic discs, and did not cause death either as a disease or as a result of poisoning. Thus it can be concluded that Lactobaccilus sp. isolated from carp intestines can be used as probiotics for cultivation.

Abstract

The aim of this research was to count the total leukocytes and phagocytosis activity of goldfish seeds treated with probiotics. The test fish used were goldfish seeds measuring 5-8 cm. 500 fish will be taken from the Tateli Fish Cultivation and Development Center (BP3I). This study used a completely randomized design (CRD) with 5 treatments, where each treatment had 3 replications. The treatments used were probiotic bacteria from the intestines of goldfish with different concentrations. The treatments tested consisted of: A: without the addition of probiotics, B: addition of probiotics 1x106 cfu / mL, C: addition of probiotics 1x107 cfu / mL, D: addition of probiotics 1x108 cfu / mL, E: addition of probiotics 1x109 cfu / mL. Each concentration of bacteria that has been prepared is then sprayed on the commercial feed evenly using a sprayer and then dried to room temperature. After drying the feed was coated with 2% egg yolk and dried again at room temperature. Fish are stocked in an aquarium measuring 60x40x40cm3 with a density of 20 fish / aquarium. Each aquarium is equipped with an aerator and a water pump for recirculation. The fish were then given feed that had been added with probiotics at the same dose and frequency of administration as in the acclimatization process. The treatment is given for 6 weeks. At the end of the treatment feeding, fish samples were taken from each experimental unit to measure immune parameters, namely total leukocytes and phagocytic activity. Based on the results of research using the probiotic Lactobacillus sp. which originated from the intestines of goldfish and added to the feed had a significant effect on increasing total leukocytes and phagocytosis activity and the optimal concenration was  1x107 - 1x108 cfu / mL with a duration of 6 weeks of administration

Abstract

The purposes of research were to examine the effect of extract of Acalypha indica leaf on the growth of Nile tilapia and determine the dose of leaf extract that could enhance the growth of tilapia optimally. The fish used were Nile tilapia taken from the Tatelu Center for Freshwater Aquaculture with an average weight of 3.79±0.50 g. The medicinal plant A. indica was collected from around the Faculty of Fisheries and Marine Science complex and from the North Bolaang Mongondow area. After being washed, the leaves were dried and then ground to obtain leaf powder. Leaf flour was then extracted by maceration method with 95% alcohol solvent. After being concentrated, the extract was then used as a treatment with different concentrations, including A: 0 g extract/kg feed, B:10 g extract/kg feed, C:20 g extract/kg feed, D:30 g extract/kg feed and E: 40g extract/kg feed. The data collected consisted of absolute growth, daily growth and relative growth. The results showed that the extract of A. indica leaf added to the feed had a significantly effect on the growth of tilapia indicated by an increase in absolute growth, daily growth, and relative growth (p<0.05). The best fish growth was achieved in fish fed with feed with the addition of 20 g extract/kg feed. In conclusion, leaf extract of A. indica can increase the growth of Nile tilapia
