Abstract

Sugarcane (Saccharum officinarum) is a high-value economical crops, only grows in tropical climates of Java and parts of Sumatra. Ratoon stunting disease (RSD) is a vascular disease in sugarcane which its visual symptoms is difficult to detect. The disease has spread across the sugarcane plantations in Indonesia with the percentage of attacks 10-100%. The use of buds chips treatment of seedling plant is an act of reducing pathogen development. The research effect of hot water treatment / HWT (0, 30, 60, and 90 min) and dose application of Trichoderma sp. (0, 25, and 50 g) using 864 varieties, is designed with a completely randomized factorial design. The results showed that Trichoderma sp. able to infect the roots and stems of seedling age 3 months, part of plant tissue increase of protein and proline content, decrease of glucose content so that increase resistance and health in the growth of sugarcane seedling before planting in the land.Tanaman tebu (Saccharum officinarum) adalah tanaman yang bernilai ekonomi tinggi, hanya tumbuh di daerah beriklim tropis terutama di Jawa dan sebagian Sumatra. Penyakit ratoon stunting disease (RSD) merupakan penyakit pembuluh pada tebu yang sulit dideteksi gejala visualnya. Penyakit tersebut telah tersebar di seluruh pertanaman tebu di Indonesia dengan presentase serangan mencapai 10-100%. Penggunaan bahan tanam benih asal mata tunas (bud chip) dapat mengurangi perkembangan patogen. Penelitian pengaruh waktu perendaman air panas (0, 30, 60, dan 90 menit) dan aplikasi dosis jamur Trichoderma sp (0, 25, dan 50 g) terhadap pertumbuhan benih tebu klon 864, dirancang dengan menggunakan rancangan acak lengkap faktorial. Hasil penelitian menunjukkan bahwa jamur Trichoderma sp mampu menginfeksi bagian akar dan batang benih umur 3 bulan, pada bagian jaringan tanaman terjadi peningkatan kadar protein dan prolin serta penurunan kadar glukosa sehingga meningkatkan ketahanan dan kesehatan pada pertumbuhan awal benih tebu sebelum ditanam di lahan.

Abstract

One of the main problems in the waste management in Indonesia is the treatment of leachate, which mostly dumped to the river This research is aimed to obtain information of histological alteration in gills and liver of C. carpio L. exposed to leachate. Measurements on the water quality parameters comprised water temperature, pH, and dissolved oxygen (DO). This research was conducted by exposing leachate to C. carpio for 96 hours. The concentration of leachate were 0 ppm, 80 ppm, and 100 ppm. Histological preparation were made on the gills and liver using 10% fixative Neutral Buffered Formalin and Ehrlich Hematoxylin-Eosin staining with qualitative observation descriptive analyses for discussion. The result showed that increasing water temperature is directly proportional to the leachate concentration in the aquaria, while the value of pH and DO inversely proportional to the leachate concentration. Damages on the gills with 80 ppm leachate concentrasion were identified as follows: fusion of secondary gill filaments and hyperplasia of epithelial cell, along with karyorrhexis and hydropic degeneration on the liver. Damages on the gills of fishes exposed to leachate with 100 ppm concentrasion were identified as follows: fusion of secondary gill filaments, hyperplasia of epithelial cell, congestion, and edema along with karyorrhexis, hydropic degeneration and melanomacrophage centre (MMC) found on the liver. The results of this study can be used as an overview of the impact of an environmental pollution by leachate as indicated from histological damage to the gills and liver of C. carpio, thus contribute significan information to aquaculture sector and endorse better waste management

Abstract

Creative thinking skills are important for long-term success. Empowering creative thinking skills which can be trained through problem-solving activities in learning, thereby promoting high cognitive engagement. This study aimed to test the effectiveness of the research and development product of Problem-Based Learning module of environmental changes in empowering students’ creative thinking skills. The research method used was the Quasi-experiment with nonequivalent control group design. The sample of the research was the tenth-graders of a Senior High School in Karanganyar district, Central Java. The sampling was cluster sampling by involving two classes, namely the Grade X-MIA 1 with 28 students and the X-MIA 4 with 28 students. The qualitative and quantitative-descriptive approaches were used to analyze the data. The result shows the mean of creative thinking skills score of experiment group is higher than the control group. This developed module could empower students’ creative thinking skills by problem-solving activities as needed for Indonesian educational goal . The study concludes that development module was constructed based on Problem-based Learning activities in the module could stimulate students solved and identified the problems with their ideas and developed their creative thinking.

Abstract

Vitamin A merupakan vitamin yang berperan sebagai imunostimulan. Penelitian ini bertujuan untuk mengetahui apakah pemberian vitamin A dapat menurunkan parasitemia Plasmodium berghei pada mencit strain Swiss. Penelitian ini menggunakan 24 ekor mencit strain Swiss jantan berumur 6-8 minggu dengan berat badan 20-30 gram/ekor. Penelitian dilaksanakan secara eksperimental dengan menggunakan time series design. Dalam penelitian ini dilakukan perlakuan berupa pemberian vitamin A dengan 3 variasi dosis yaitu 0 IU/g BB, 35 IU/g BB dan 70 IU/g BB serta kelompok kontrol negatif dengan masing- masing kelompok terdiri dari 6 ekor mencit. Vitamin A diberikan 1 jam sebelum penginfeksian dan mencit dirawat sampai mencit pada kelompok kontrol negatif mati. Sediaan apus darah tipis dibuat 2 hari sekali dan parasitemia dihitung dengan pengecatan Giemsa. Data parasitemia dianalisis dengan ANOVA.Untuk hasil yang signifikan maka dilanjutkan dengan uji lanjut Post hoc pada taraf kesalahan 1%. Hasil uji ANOVA untuk kelompok perlakuan B, C dan D diperoleh nilai p <0,001 pada masing-masing kelompok perlakuan. Hal tersebut menyatakan bahwa adanya perbedaan yang signifikan pada perlakuan yang diberikan. Begitu juga untuk uji lanjut Post hoc yang telah dilakukan diperoleh nilai p < 0,001. Dapat disimpulkan bahwa pemberian vitamin A berpengaruh terhadap penurunan parasitemia Plasmodium berghei pada mencit strain Swiss.Vitamin A is a vitamin that acts as an immunostimulant. This research aims to determine whether the administration of vitamin A can reduce parasitaemia of Plasmodium berghei in Switzerland strain mice. This research used 24 mice Switzerland strain mice 6-8 weeks old weighing 20-30 grams/tail. Research conducted experimentally by using time series design. In this research, the provision of vitamin A treatment with 3 doses of variation is 0 IU/g BW, 35 IU/g BW and 70 IU/g BW as well as the negative control group, with each group consisting of 6 mice. Vitamin A was given 1 hour before infection then treated mice to mice in the negative control group died. Thin blood smears were made 2 days and parasitaemia was calculated by Giemsa staining. Parasitaemia data were analyzed with ANOVA. Significant results then followed by a further test Post hoc at 1% error level. ANOVA test results for the treatment group B, C and D obtained value of p <0.001 for each treatment group. It is claimed that there are significant differences in the treatment given. Likewise for further Post hoc testing that has been done obtained value of p <0.001. It can be concluded that the administration of vitamin A affects the reduction in parasitaemia of Plasmodium berghei in Switzerland strain mice.

Abstract

Tanaman daun dewa [Gynura procumbens (Lour) Merr.] berguna untuk menurunkan kadar kolesterol darah, mengobati diabetes, tumor, dan sebagai obat anti kanker. Daun dewa mengandung senyawa metabolit sekunder yaitu flavonoid, saponin, dan minyak atsiri. Oleh karena itu, perlu adanya kultur in vitro yang diharapkan mampu meningkatkan jumlah metabolit sekunder, maka perlu dilakukan penelitian tentang induksi kalusdengan penambahan kombinasi 2,4-Dichlorophenoxyacetic acid dan kinetin yang tepat dapat digunakan untuk menginduksi kalus daun dewa. Tujuan penelitian ini untuk mengkaji pengaruh konsentrasi 2,4-D dan Kinetin serta interaksi keduanya terhadap induksi kalus daun dewa. Analisis menggunakan anava dua arah dan uji lanjut Duncan. Parameter yang diamati adalah parameter persentase eksplan yang hidup, persentase berkalus, berat basah, kering kalus, serta tekstur dan warna kalus . Dari Uji Jarak Berganda Duncan diperoleh hasil tertinggi pada konsentrasi 2,4-Dichlorophenoxyacetic acid 0.5 ppm dan konsentrasi kinetin 1 ppm sebesar 33.33% pada parameter persentase berkalus.Daun dewa [Gynura procumbens (Lour). Merr.] plant can be used to lower the blood cholesterol levels, to treat diabetes and tumors, and may be used as an anti-cancer drug. Daun dewa plant produces of secondary metabolites such as flavonoids, saponins, and essential oils. An in vitro culture is a necessity to increase the quantity of secondary metabolites using callus induction with the addition of a combination of 2,4-Dichlorophenoxyacetic acid and kinetin to induce daun dewa callus. The research will ecamine the influence of 2,4-Dichlorophenoxyacetic acid and Kinetin in various concentrations and their interaction on callus induction of daun dewa. Data were analyzed using advanced two-way anova and Duncan test. The test showed that the highest yeield was obtained from the combination of 0.5 ppm 2,4-D and 1 ppm kinetin, where 33.33% of plants had callus.

Abstract

The purpose of this study was to identify the selection criteria to obtain a superior mutant derived from the wheat plants of such varieties as Dewata, Selayar and Alibey, adaptive in medium land. The analysis of agronomic growth characters showed a significantly effect on a growth percentage of the initial growth (8 mutants), flowering time (1 mutant), panicle stem length (15 mutants), number of panicles (7 mutants), the number of grains per panicle (8 mutants), grain weight observed (8 mutants), grain weight per genotype (6 mutants), leaf area (2 mutants) and leaf greenness (5 mutants). The effects on the characters of ripe time, harvest, panicle length and plant height were not significant. The mutants of Dewata, Selayar and Alibey could be selected based on the characters of panicle stem length, number of grains per panicle and grain weight per observation because these characters generated more mutants than the other characters. The correlation analysis between the characters of growth and yield components of wheat mutants showed that the number of grains per panicle was positively correlated with the grain weight observed, while the length of panicle stem was positively correlated with grain weight per genotype, number of panicles and leaf area. Abstract

Feronia limonia (L.) Swingle better known as wood apple is an identity plant of Rembang Regency, Central Java Province, Indonesia. However, the population is very limited. In order to identify the conservation efforts, this ethnobotany study was aimed to determine traditional knowledge on the existence, economic value, utility, breeding technique and conservation of wood apple. The survey was conducted on a total of 102 local people from 10 sub-districts which were determined by proportionally stratified random sampling. Data were collected by questionnaires and individual interviews and then were analyzed descriptively. The results showed that all of respondents knew the figure of wood apple, but only 79.50% understood its status as an identity plant. Most of respondents (96%) stated that the fruits have low economic value, 80.39% respondents have utilized ripe fruit as ingredients of beverages and stems as firewood. This utilization was only equal to 29.72% compared to the potential usability based on research results. It was concluded that the traditional knowledge about the existence, economic value, and breeding technique were good and on the contrary, the knowledge about the plants usability was bad. The results will form a basic knowledge for in-depth studies to provide a government policy of Rembang Regency in order to develop an efficient strategy of management and conservation of wood apple as plant identity.

Abstract

Entomopathogenic nematodes Entomopathogenic nematodes (EPN) of the genera Heterorhabditis and Steinernema are commercially used to control pest insect. EPN is widely cultivated through in-vivo and in vitro methods. This research aims to discover the abundance of EPN cultivated in various artificial media. Seven types of media composition were tested in this research: media A (yeast + soybean powder), media B (yeast + chicken liver), media C (yeast + dog food), media D (yolk + soybean powder), media E (yolk + chicken liver), media F (yolk + dog food), and media G (yeast + yolk + dog food). Each media was inoculated in 1.2x103JI/mL. The growth of EPN was observed weekly in 4 weeks. Results showed that EPN could be cultivated using various media; media D, E, F, and G. Abstract

Tujuan penelitian adalah menghasilkan model meksint korefsi dengan pendekatan Jalajah Alam Sekitar pada pembelajaran Struktur Tubuh Hewan sehingga dapat dimanfaatkan sebagai salah satu alternatif kegiatan perkuliahan Struktur Tubuh Hewan. Metode yang digunakan dalam penelitian ini adalah Research& Development dengan tahapan sebagai berikut: planning, design, dan development dengan 2 tahap evaluasi yaitu: uji alpha, uji betha, dan uji lapangan. Hasil uji alpha pada model pembelajaran meksint korefsi oleh ahli pembelajaran biologi memperoleh skor sebesar 3.13 dan hasil uji betha oleh mahasiswa sebagai pengguna memperoleh skor sebesar 3.64. Hasil uji t test antara dua rombel mahasiswa responden uji lapangan diperoleh thitung= 2.94 ? ttabel= 2.39 (? = 0.01; n = 60). Dengan demikian model meksint korefsi dengan pendekatan Jalajah Alam Sekitar layak digunakan sebagai salah satu alternatif pilihan model pembelajaran dalam proses perkuliahan Struktur Tubuh Hewan, sehingga disarankan perlu adanya proses diseminasi model tersebutThe purpose of the study was to develop the meksint korefsi model with Jelajah Alam Sekitar* approach in the Animal Anatomy, for the alternative model in Animal Anatomy lecture. This is a Research and Development, with the following stages: planning, design and development with three stages of evaluation are: alpha testing, beta testing, and field testing. Alpha test results on the learning model meksint korefsi by expert of instructional biologists with scores was 3.13 and beta test results by the students as users gain score of 3.64. Field test results were analyzed by t-test statistical tvalue = 2.94 ? ttable = 2.39 (? = 0:01, n = 60). The result showed that the meksint korefsi model with Jelajah Alam Sekitar* approach was a feasible model in the learning of Animal Anatomy, so it was suggested to disseminate this model.

Abstract

Pak Choy or Bok Choy (Brassica rapa L. var. chinensis) is one of favorite Chinese leafy vegetable for various dishes in Indonesia. In this study, it was used as a plant model to identify the appropriate organic hydroponic nutrient solution for leafy vegetable seedling production. The seed was sown on rock wool slabs submerged with 200 ml of a nutrient solution containing biofertilizer of Beyonic StarTmik@Lob (0, 25, 50, 75, and 100%). commercial hydroponic solution (0, 25, 50, 75, and 100%) and its combination (25, 50, and 75%). The experiments were arranged in a CRD. Meanwhile, the obtained data was analyzed using ANOVA followed by DMRT. The relationship among growth parameters was observed using Pearson correlation analysis. The result of the study showed that the combination of organic and inorganic nutrient (25% Beyonic StarTmik@Lob and 75% commercial hydroponic solution) resulted in the highest seedling growth parameters and leaf indices as well as the perfectly positive correlations among growth parameters. This result indicated that the use of organic nutrient alone was not appropriate for hydroponic seedling production of Pak Choy. Therefore, further study needs to be done to identify the hydroponic solution without inorganic nutrients towards the organic vegetable production.
