Abstract

Colibacillosis is an infectious disease in chickens caused by Avian Pathogenic Escherichia coli (APEC). The ability of APEC to cause disease depends on many pathogenic factors, one of which is the iutA pathogenic gene. This study was purposed to determine the DNA sequence of the APEC iutA gene in Bali and its kinship with the iutA gene from other countries. Two APEC isolates from free-range chicken in Tabanan dan Badung has been used. The isolates have been purified and were available at the Bacteriology Laboratory of Veterinary Medicine Faculty, Udayana University. Chelex 10% was used for DNA isolation. DNA amplification using published DNA primer has been conducted with the polymerase chain reaction (PCR) method. The PCR product was sequenced at Malaysia, First Base Laboratories using Sanger’s Dideoxy Nucleotide Termination method. The iutA gene of both isolates can be analyzed and have 250 bp in length. Both were 100% homologous. A phylogenic test using 58 DNA sequences of iutA gene from Escherichia coli and other bacteria in the world was conducted in MEGA 5.2. All data have 43 polymorphic sites of nucleotide acid and 13 polymorphic sites of amino acid. The iutA gene from Bali is in one group, with the iutA gene from Brazil (KP657535) in 2011, Germany (LT599825) in 2016, and China (CP033635) in 2016. This gene can be used as a pathogenic marker of APEC in Indonesia.

Abstract

Broiler merupakan jenis ayam yang paling cepat berproduksi dan dagingnya memiliki mutu yang tinggi dibandingkan dengan jenis ayam ras lainnya. Penelitian ini bertujuan untuk mengetahuikepekaan bakteri Escherichia coli (E. coli) sebagai penyebab kolibasilosis terhadapsulfametoksazol, ampisilin, dan oksitetrasiklin pada peternakan broiler di Banjar Tangkub, DesaPayangan. Bakteri E. coli diisolasi dari 30 sampel feses broiler penderita diare diuji kepekaanyadengan menggunakan metode Kirby-Bauer dan dianalisis secara deskriptif. Hasil penelitianmenunjukkan bahwa sebanyak 25 isolat resisten (83,33%), satu isolat intermediet dan sisanyaempat isolat sensitif terhadap antibiotika oksitetrasiklin. Sedangkan terhadap antibiotika ampisilin,sejumlah 24 isolat (86,67%) menunjukkan resisten dan sisanya yakni empat isolat sensitif terhadapantibiotika tersebut. Terhadap sulfametoksazol, kuman E coli yang diisolasi menunjukkan 29 isolatresisten (96,67%) dan satu isolat sensitif. Berdasarkan hasil penelitian dapat disimpulkan bahwapenggunaan sulfametoksazol, ampisilin dan oksitetrasiklin sebaiknya dihindari.

Abstract

This research was conducted for the detection of Staphylococcus sp. in the respiratory tract of pigs. Samples differentiated by sex (male pigs and female pigs). A total of 100 nasal swab samples were collected aseptically from pigs showing symptoms of respiratory disease, from pig farms in the area of Badung and Tabanan. All samples were cultured on to Blood Agar for 18-24h at 37°C. Suspected colonies were then Gram staining, primary tests (tested for catalase and oxidase) and series of biochemical tests. Statistical analysis (Chi-square) was performed to explore the differences in positive samples numbers of Staphylococcus sp. isolated between male and female pigs. Of the 100 samples, seven positive samples of Staphylococcus sp. (six of the boars and one from the female pig). Statistically, the number of boars infected with Staphylococcus sp. higher (P<0.01) than with female pigs Staphylococcus species isolated were: Staphylococcus aureus, Staphylococcus epidermidis, and Staphylococcus saprophyticus, respectively.

Abstract

An observational study in order to determine the distribution Enterobacter spin Bali cattle at different geographical area (highland and lowland) and according to the animal maturity (calves, heifers, adult) have been undertaken. Isolation and identification of bacteria based on the method of Carter and Cole with slightly modification and the antibiotic sensitivity test were determined using the disc diffusion method by Kirby and Bauer. The Enterobacter sp. was isolated from 16 (13.3%) out of the 120 nasal swab samples,where 8 (6.7%), 6(5.0%), and 2 (1.7%) was isolated from calf, heifers, and adults, respectively. Meanwhile, 6 (5.0%) Enterobacter sp. was isolated from the highland and the remains (8.3%) from the lowland. Antibiograms showed that 13 of the 16 isolates (81.2%) weresensitive to sulfamethoxazole and the remains (18.8%) intermediate. While 12 (75.0%), 1 (6.2%), and 3 (18.8%) isolates weresensitive, intermediate, and resistant to ampicillin, respectively. To oxytetracycline 8 (50.0%) isolates were sensitive, 4 (25.0%) isolates were intermediate, and 4 (25.0%) isolates were resistant. Generally, the Enterobacter sp isolates were sensitive to all the antibiotics tested.

Abstract

This study aim was to determine the total bacteriacountthat contaminated broiler farming water in Mengesta Village, Penebel District, Tabanan Regency. The water sample was collected from the source of the spring, water reservoir and chicken drinking pot. Each sample was taken as many as 100 ml and placed in the erlenmeyer glass. Water sampling was repeated 9 times infour-day sampling period. The data obtained were tested by analysis of variance and continued with least significant difference test. The results showed that total bacteria in chicken drinking pot(389,78 CFU / ml) was higher than in springs (247,89 CFU / ml), and shelter (171,33 CFU / ml). Its concluded that bacterial contamination occurred along the water flow.

Abstract

The aims of this study is to determine the effect of maturity level and location of the farms to the number of coliform bacteria in bali cattle feces in Nusa Penida. 24 samples of bali cattle’s feces were used in this research. This study used a Randomized Complete Block Design (RCBD) of 2x3 factorial consist of two location of altitude plateu (lowland and highland) and three maturity level (calves, heifers, and adult). The sample was grown on Nutrient Agar using the casting method. The obtained data were tested by analysis of variance followed by Least Significant Difference (LSD) Test. The results showed that the number of coliform bacteria in bali cattle was significantly different at maturity level and location of the farm. Number of coliform bacteria in bali cattle of the lowland was (138.66 x 107CFU/g), which is higher than bali cattle on the high land (102 x 107CFU/g). Test results of LSD Test showed the number of coliform cattle calf is 43,25 x 107 CFU/g is lower compare to heifers 124 x 10­­7 CFU/g and adults 193,75 x 10­­7 CFU/g. The number of coliform bacteria in feces of heifers cattle is lower than the number of coliform bacteria in feces of adult cattle.

Abstract

This study aims were to determine the number of Coliform and Escherichia coli bacteria in drinking water of broiler chickens farm in the Mangesta Village, Penebel Sub-District, Tabanan. This study used a randomized block design from three sampling sites which were Beji (the water springs), water reservoir, and drinking place livestock, and had nine times repetition in each group. The number of Coliform and Escherichia coli bacteria was calculated by the method of spread. The data result from Coliform and Escherichia coli bacteria were analyzed using Analysis of Variance, followed by Duncan's Multiple Range Test. The research number of Coliform bacteria at Beji (water springs), water reservoir, and the drinking plate were 19768 x 105 CFU/100 ml, 11222 x 105 CFU/100 ml, 30957 x 105 CFU/100 ml and the number of Escherichia coli were 9233 x 105 CFU/100 ml, 3457 x 105 CFU/100 ml, and 19657 x 105 CFU/100 ml. Statistically, the number of Escherichia coli and Coliform in drinking place was significantly higher than the Beji (water springs) and water reservoir.

Abstract

This study aims were to determine the sensitivity pattern of Escherichia coli bacteria isolatedfrom the feces of birds chirping with diarrhea against antibiotic sulfamethoxazole, ampicillin, andoksitetrasiklin in Satria and Sanglah bird market.The results showed that all 30 isolates from birdfaeces were examined, sulfamethoxazole sensitive 3.3%, intermediate 13.3%, and resistant 83.3%.In oxytetracycline 3.3% sensitive, 40% intermediate, and 56.7% resistant . While sensitive to ampicillin 13.3%, 23.3% intermediate, and 63.3% resistant.

Abstract

A case study on morbidity, mortality and case fatality rate of swine colibasillosis was done. A totalof 244 pigs aged 1 day to 4 weeks that were collected from semi-intensive pig farms was used inthis study. All of the young pigs were obtained from Sudimara Village, in Tabanan District, Bali.The observation of the disease was evaluated in 30 days. Confirmation of Colibasillosis infectionwas based on the diarrhea clinical sign, and then confirmed with microbiological test using EMBA.The percentage of morbidity, mortality and case fatality rate were calculated based on the formulaof Roberson (2008). Results showed the morbidity, mortality and case fatality rate were 8.6%,2.05%, and 23.8% respectively. These results indicating that morbidity, mortality and case fatalityrate in young pigs that were rising in semi-intensive farm were relatively low. It seemed that thelow level of morbidity, mortality, and case fatality rate were influenced by the pig rising systemwhere the feed, captive, and hygiene are much better than the traditional pig farming system.

Abstract

The study aim was to determine the resistance of Escherichia coli to antibiotics that isolated in laying hens. A total of 100 samples of chicken feces were taken from Tabanan, Bangli and Karangasem districts. E. coli were isolated and identified by Carter (1990) method, followed by a bacterial susceptibility test with agar diffusion method from Kirby Bauer against antibiotics namely oxytetracycline (30 µg), ampicillin (10 µg), streptomycin (10 µg), sulfamethoxazole (30 µg), chloramphenicol (30 µg), and kanamycin (30 µg). The obtained data compared to the next sensitivity standard of the bacteria that classified as sensitive, intermediate, and resistant. The results showed that E. coli from chicken feces are sensitive to kanamycin, whereas the other antibiotics are intermediate or resistant. The increase in resistance is in line to the age of the chicken.
