Abstract

Streptococcus sp. is one of the primary causes of the Porcine Respiratory Disease Complex (PRDC). This research was conducted to study the bacteria Streptococcus sp. in the pig's respiratory tract sufferers Porcine Respiratory Disease Complex (PRDC) and distribution of the bacteria Streptococcus sp. in pre-weaned and post-weaned pigs. A total of 43 samples were nasal swab taken from pigs that showed PRDC symptoms such as depression, anorexia, dyspnea, the presence of exudates from the nasal, coughing / sneezing, and swelling of the joints. Samples were received from pig farms in Tabanan regency, Badung regency, and Gianyar regency. All samples were planted on sheep media so that blood was continued with the Gram staining test. The suspected colonies were then subjected to a primary test consisting of catalase and oxidase tests as well as biochemical tests with MRPV, TSIA, SIM, coagulase test and sugar test. The results showed that 23 samples (thirteen from pre-weaned pigs and ten from post-weaning pigs) were tested positive for Streptococcus sp. hemolytic ? (20 isolates) and Streptococcus sp. hemolytic ? (3 isolates).

Abstract

This study aims to detect bacterial infectious agents in pigs with complex respiratory tract disease known as porcine respiratory disease complex (PRDC) in some pig herds in Bali and look for the right medication is use to handle the incidence. The research sample comes from Tabanan, Badung and Gianyar totaling 48 Samples were isolated and identified by methods: isolation, the primary test, biochemical test. Test of sensitivity to antibiotics using disc diffusion method according to Kirby-Bauer method. The result showed that bacterial agents isolated from pigs were among others Streptococcus spp. alfa hemolytic non-coagulase (41,6%), Staphylococcus spp. beta hemolytic noncoagulase (41,6%) and Escherichia coli. Staphylococcus spp. beta hemolytic non-coagulase and E. coli were sensitive to streptomycin, kanamycin, doxicyclin, clindamycin and bacitracin. To conclude, Streptococcus spp. alfa hemolytic non-coagulase were sensitive to streptomycin, kanamycin, clindamycin and bacitracin, but resisten to doxicyclin.

Abstract

Research has been conducted to know the number bacterial isolates of Klebsiella sp. from bali cattle in terms of the level of maturity and the breeding location, also its sensitivity to the antibiotic there are ampicillin, sulfamethoxazole, and oxytetracycline. 120 bali cattle's nasal swabs consisting of calves, heifers, and adult cattle which are breeding on highland and lowland has been used as the isolates in this research. The obtained nasal swab was isolated and identified, then proceed with the sensitivity test. Bacterial of Klebsiella sp. has been found in calves were nine isolates (45%), heifers were six isolates (30%), and adult cattle were five isolates (25%). While based on the breeding location in the lowland found 13 isolates (65%) and highland found seven isolates (35%). In the sensitivity test results were obtained for the antibiotic ampicillin all isolates were sensitive (100%). sulfamethoxazole resistant four isolates (20%), intermediate two isolates (10%), and sensitive 14 isolates (70%). Oxytetracycline resistant seven isolates (35%), intermediate two isolates (10%), and sensitive 11 isolates (55%). The results showed that differences in the level of maturity and breeding location of bali cattle can affect the distribution of bacteria Klebsiella sp.

Abstract

The main of this study were characterized of muramidase released protein (MRP) andexctracellular factor (EF) as virulence factor of Streptococcus beta hemolityc of Bali islotesand its patogenecity on mice. The MRP was isolated from the cell walls bacteria withmuramidase (lysozyme) and EF was obtained from supernatant of bacteria precipated with70% ammonium sulphate and then dialysed. These protein were identified by usingsodium-dodecyl sulphate polyacrylamide gel electrophoresis ( SDS-PAGE). Isolates thatwas observed are five consist of three isolates from pigs and two isolates from monkeys.Pathogenecity test using 20 mices divided into four group. Group I inoculated with 0,1 mltodd-hewitt broth steril as negative control, group II inoculated with 0,1 ml inoculum ofbacteria of Streptococcus suis type 2 (strain D282) as positive control, group III inoculatedwith 0,1 ml Streptococcus beta haemolytic isolated from monkey and group IV inoculated with 0,1 ml Streptococcus beta haemolytic isolated from pig. The result of this studyshowed that all isolates were consist of eight protein bands of MRP and one EF of 110 kDamolecular weight. Eight protein MRP were 125 kDa, 76 kDa, 60 kDa, 57 kDa, 48 kDa, 45kDa, 30 kDa, and 28 kDa respectively. Each isolates had two major protein bands of MRP(76 kDa and 45 kDa).The patogenecity test in mice showed that the morbidity andmortality rates were 100% and 60% respectively. The prevalence of meningitis in mice are100%. Clinical sign were observed 30 hour post inoculated (pi) whereas mice found deathstarty 48 pi.
