Abstract

Tiga senyawa turunan kalkon telah berhasil disintesis dari suatu keton aromatik dan aldehida aromatik melaluireaksi kondensasi Claisen-Schmidt dalam suasana basa. Turunan kalkon tersebut adalah 2’-hidroksi kalkon,(2Z,4Z)-1-(2-hidroksifenil)-5-fenilpenta-5-2-4-dien-1-on dan 2’-hidroksi-4-dimetilamino kalkon. Senyawa yangdihasilkan relatif murni, dilihat dari uji KLT yang menunjukkan satu noda dan titik leleh ketiganya berada padarange kecil. Elusidasi struktur didasarkan pada data spektra UV, IR, 1H-NMR dan 13C-NMR. Uji antimikroba terhadapketiga senyawa, hanya satu senyawa yang memberikan hasil positif yaitu senyawa 2’-hidroksi-4-dimetilaminokalkon pada konsentrasi 60 ìg terhadap bakteri B.subtilis.

Abstract

A bacterial lipase producer was isolated from garbage dump soil and was identified its genus. Lipase was extractedaccording to production time optimized, purified using ammonium sulfate fractionation and gel chromatograph.Determination of enzyme characteristic studied were influence of pH, temperature, various metals to lipaseactivity. The result of this research shows that the genus of isolated bacteria which produced lipase wasAcinetobacter sp., the lipase optimum production time is about 18 hours with the activity is about 115 unit/mL. Thehighest activity of lipase fractionation using ammonium sulfate is about 45% and the highest activity of purifyingwith filtration gel chromatograph column using Sephadex G-150 at 24 th fraction. Lipase from crude extract andpurifying product at this fraction has optimum pH 6 and optimum temperature is about 40 oC. Lipase to be classifiedas metalloenzyme that shows with decreasing the activity after added the EDTA. Metals ion, such as Cu 2+ and Zn2+were inhibited the lipase activity. Ca 2+ ion could increase lipase crude extract activity but inhibited the activity oflipase purifying product. Hg2+ ion could increase the activity of lipase purifying product.

Abstract

The effect of addition cytokinins and modification of sucrose concentration on growth and alkaloid canthinoneproduction in cell suspension cultures of Eurycoma longifilia Jack were studied. The additions of cytokines, BAand kinetin, show effect on the production of biomass and alkaloid in cell suspension of E. longifilia Jack. Theoptimum totals of two-alkaloids were obtained on addition 4.44 μM BAP and without kinetin, respectively. Theaddition of 4.44 μM BA (6-benzyladenine) into TAM medium stimulated increased total of 9-hydroxycanthine-6-one,but decreased total of 9-methoxycanthin-6-one. While the addition of 2.32 - 9.29 μM kinetin (6-furfurylaminopurine)into TAM medium decreased total of two alkaloids (from 0.582 mg to 0.461 - 0.257 mg per 25 ml medium). Whensucrose concentration in TAM medium was increased from 3% to 5%, production of biomass would increase from0.374 g to 0.585 g dry weight per 25 ml medium. While total of two-alkaloids increase from 0.328 mg to 0.441 mgper 25 ml medium when concentration of sucrose in TAM medium was increased from 3% to 4% sucrose.

Abstract

Two resveratrol dimers, (-)-ampelopsin F dan (-)-laevifonol were isolated from aceton extract of the stem bark ofShorea parvifolia. The structures of these compounds were determined based on their spectroscopic data includingspectroscopy ultra violet (UV), infra red (IR), nuclear magnetic resonance (1H-NMR, 13C-NMR) and also were comparedto the reported data.

Abstract

erbal products in powder or capsule forms made of sarang semut plant have been widely distributed in market.This plant is believed to treat several diseases, such as cancer, gout, liver, stroke, heart, hemorrhoid, back pain,allergy, and as tonicum to increase sexual desire. Information on the plants was still limited on plant distribution,ecology, ethnobotany, and taxonomy, without research on toxicity and clinical aspect. Considering that sarangsemut products has been distributed at market and limited scientific publication of the plant, hence acute toxicitytest of sarang semut extract should be conducted. The results showed that dose of 375 mg/kg bw caused liverdegeneration on day-5 and normal on day-19. Whereas dose of 3750 mg/kg bw caused cell necrosis up to day-12,degeneration still apparent on day-26.

Abstract

Urinary Tract Infection (UTI) is caused by a variety of microorganism, but bacteries are more often. Antibacterial isthe empiric treatment for UTI. The antibacterial resistance pattern of UTI can be changed by the place and time.Urine culture test and sensitivity test will help us to choose the effective treatment. The purpose of this researchis to find out the pattern of UTI in Pekanbaru. Results of this research showed that UTI bacterial resistancy mostlyfound in Gram negative bacateria with penicillin and 1 st cephalosporin antibiotic.

Abstract

Lais fish of Ompok hypophthalmus is one of high economic fish in Kampar river. It should be protected fromdecreasing of it population estimated due to decreasing of habitat quality and increasing of exploitation. Theobjectives of the research are to study reproduction biology of lais fish as the basic data for conservation. Thisresearch was conducted from January 2007 to January 2008. The results of O. hypophthalmus reproductionaspect show that the smallest female of maturity is 22,9 cm and male is 22,6 cm; the spawning season onSeptember to November; O. hypophthalmus is more appropriate spawning location to oxbow lake that closerelation with tributary; the spawning pattern indicated total spawner fish; the fecundity ranges from 3111 to 11164eggs and the egg diameter ranges 0,41-1,13 mm.

Abstract

This study was conducted to evaluate the potency of Brucea javanica (melur) for controlling two species ofcrucifer pests, i.e. Crocidolomia pavonana and Plutella xylostella. Melur fruits, twigs, and leaves were extracteddirectly with methanol or sequentially with hexane, ethyl acetate, and methanol. The most active extract was thenfractionated by preparative layer chromatography using hexane, mixtures of ethyl acetate and methanol, andmethanol as eluents. The most active fraction was formulated as EC (emulsifiable concentrate) and WP (wettablepowder) formulations, and tested for their toxicity and antifeedant effect against C. pavonana and P. xylostellalarvae. The results showed that methanol extract of melur fruits was more active than that of twigs and leaves.Fractionation of methanol extract of melur fruits yielded an active fraction which was eluted with ethyl acetate-methanol 9:1. EC and WP formulations of melur fruits were active against C. pavonana larvae with LC50 of 0.39%and 0.21%, respectively. The same formulations were also active against P. xylostella larvae with LC50 of 0.31% and0.54%, respectively. In no-choice tests, the antifeedant effect of the EC formulation on C. pavonana larvae (feedinginhibition [FI]: 70.9%-97.5%) was higher than on P. xylostella larvae (FI: 52.2%-83.9%), but the antifeedant effect ofthe WP formulation on the two species was relatively the same. In a choice test, the EC formulation at LC 85completely inhibited feeding by C. pavonana larvae (FI: 100%).

Abstract

Improvement of selected induction culture media on callus induction in anther culture of anthurium and a histologicalstudy on its callus formation were studied at the tissue culture laboratory of the Indonesian Ornamental CropsResearch Institute from February to October 2008. The objectives of the study were to optimize selected media forcallus formation, reveal cell origin of callus derived from anther culture and shoot formation process. Selectedmedia improved in the study were 1) MMS-TBN containing 0,5 mg/l TDZ, 1,0 mg/l BAP and 0,01 mg/l NAA (Winartomedium, WM) and 2) MMS III supplemented with 1,5 mg/l TDZ, 0,75 mg/l BAP and 0,02 mg/l NAA (Winarto andRachmawati medium, WRM). Improvement treatments were carried out by omission and application of 2,4-D in 0.5mg/l and reduction of medium strength of full, half, quarter, one eighth, one sixteenth, and zero strength. Afactorial experiment was arranged using a randomized complete block design with four replications. Results ofthis study indicated that the highest callus induction was clearly established in WRM. The medium stimulatedpotential growth of anther (PGA) up to 81% with 49% of percentage of anther regeneration (PAR) and 2.7 number ofcallus formed per replication (NCF). Significant improvement in callus formation was also recorded by reduction ofmedium strength of WRM to one eighth compared to others. The reduction induced PGA up to 58% with 29% of PARand 1.8 NCF. From histological studies it was well recognized that regenerated callus on half anthers cultured wasoriginated from middle layer cells of anther wall. The morphogenic response of anther wall cells caused primarilyon no androgenesis effect in microspore cells.

Abstract

This research was conducted in the Experimental Field, Faculty of Agriculture, Riau Islamic University and SoilLaboratory, BPTP Riau Province from August to December 2007. The research aim to study the effect of Cu2+, Zn2+,Fe3+ ameliorant addition in peat to N, P, K uptake and rice yield of various varieties. The experimental with CompletelyRandomized Design for Factorial 4 x 4 were used. The first factor was ameliorant addition, consists of four levels(Cu2+; Zn2+; Fe3+) and second factor based on varieties, consists of four levels (PB-42, IR-64, Ciherang, Cisantana).The result of research indicates that: (1) Interactian effect of treatment was significant to N, P, K uptake, productivestem, weight of dried seed, and weight of 1000 seeds, (2) The best treatment was Cu2+ ameliorant on IR-64 withuptake of N ( 254,0 mg/plant), P (32,8 mg/plant), K (76,0 mg/plant), productive stem (30,0 stems/plant), weight ofdried seed (63,9 g/plant), weight of 1000 seeds (23,7 g).
