Abstract

Fatty hydroxamic acids (FHAs) have been successfully synthesized from coconut oil by a one-step lipase catalyzed reaction. FHAs are theacids that are based on fatty acids. Their hydrophobicity can be use for some applications such as surfactant and metal ions extraction fromaqueous media. This paper describes enzymatic synthesis of FHAs from coconut oil by reacting hydroxylamine with the substrate catalyzedby a lipase. The lipase used was Lipozyme, a commercially lipase of Mucor meihe fixed on a polymer anion exchange resin. The use ofimmobilized lipase as the catalyst for the preparation reaction provides an easy isolation of the enzyme from the products and othercomponents in the reaction mixture. In addition, it also allows the reaction to be carried out under mild conditions, which reduces thereaction’s side products. The optimum preparation conditions obtained were as follows; the reaction temperature was 30 oC, the reactiontime was 30 h, the ratio of coconut oil : lipozyme (g : g) was 29.5, and the ratio of hydroxylamine : coconut oil (mmol : mmol) was 6. Thesolvent used was hexane. The purified products were characterized by qualitative test, such as FTIR spectroscopy and HPLC.

Abstract

Bekasam is a traditional fermented fish from Indonesia. It is made from fish  in which  the gills and  gut  had been  discarded,  washed,  and  mixed with salt, after two days, it was covered  with  rice  and  salt,  and then  fermented  for  5-7 days  at room  temperature.  Bekasam  is perceived to have antihypertensive activity, which is though to be the activity of bioactive peptides, the product of protein degradation  during  the bekasam fermentation. The research was aim to isolate the indigenous lactic  acid  bacteria  that  produce strong  proteolytic  capabilities.  The lactic  acid bacteria were collected from 3  kinds of bekasam (tilapia, milkfish, and tuna bekasam). Isolation  of the lactic acid bacteria was done         on agar medium containing MRS + CaCO3, and  the screening of the proteolytic bacteria  was done by growing the isolated  culture on  skim agar.   The colonies  that produce clear zone  were identified  as proteolytic  lactic acid  bacteria. Some of 180  isolated strains of acid  producing bacteria   that produce clear zone on MRS agar were found 150 strains of  lactic acid  bacteria, and  84  strains  showed  their  proteolytic activity.  The later  were identified morphologically and biochemical as Lactobacillus, Pediococcus and Leuconostoc genera.  Selection  was further conducted based on the height of the proteolytic and homofermentative fermentation activities. Upon species identification using API CH 50 kit,  the  selected strains belong to species of Lactobacillus plantarum, Lactobacillus pentosus, and Pediococcus pentosaseus.

Abstract

Malondialdehyde, a product of lipid peroxidation, is often used as an indicator to detect the oxidative stress damaged in the body. Lipidperoxidation is associated with progression of atherosclerosis. It is known that onion (Allium ascalonicum L.) can be used as an antioxidantand hypolipidemic agent, but whether onion can decrease hypercolesterolemia mice MDA plasma induced by yolk is still unknown. Thepurpose of this study was to investigate the effect of onion’s juice on hypercolesterolemia mice MDA plasma level. This was an experimentallaboratory research with post test only design with control. The measured parameters were cholesterol and MDA level on tested groups.Groups with treatment consist of yolks group, and 80% shallot juice, simvastatin and aquadest were separately used as positive and negativecontrol. The result showed that there was significant difference between high fat feed group which added 80% onion’s juice and high fat feedgroup only. Addition of 80% onion’s juice in high fat feed groups decreased MDA plasma level equal to normal feed group. In conclusion, itwas proved that onion’s juice decreased mice MDA plasma level.

Abstract

Oil palm plantations at peatland are faced with problems of land degraded, low productivity and produced to green house gas (GRG) emission.The objectives of this study were to land clearing effect of peatlands and to identified factors that affect the peatlands in the biophysicalcharacteristics. This study was conducted on District Bengkalis-Riau at marine and brakish peat. The data was collected using field survey anddocumentation. The descriptive analysis was used to determine biophysical characteristics and linier regresion was used to corelation performimportant factors. The research results showed that the biophysical characteristics of peatland experienced changes in the horizon profile,peat thickness, decomposition level, moisture content, ash content, pH, C-organic and biomass. The secondary peat swamp forest ofbiophysical characteristics have to different in the horizon profile, peat thickness, decomposition level, moisture content, ash content, pH,C-organic and biomass compared with marine and brackish peat.

Abstract

This study was aimed to investigate diversity of sea cucumber. Samples were collected  in  the coastal  waters  of  Natuna,  Riau  Islands  in  June 2009. Samples were taken from 2 locations: Sepempang and Pengadah beach. Sampling was done by using a transect quadrant of 5x5 m. This  sampling and observation on  its  microhabitat  were conducted by snorkeling. Analyses on the sea cucumber community structure were based on its diversity, equitability, density and frequency of occurance. The result showed that at both  locations there  are 16  species from two families: Holothuriidae and  Stichopodidae.  The higher  diversity index  (H) was  3.256  and  equitability  index  (e) was 0.993.  The higher  density  of species were 0.0032 ind/m2. The higher frequency of occurance was 27.71% by H. atra.

Abstract

This research is about effectivity analysis of two kinds of fungi i.e. Beauveria bassiana and Metharrizium anisopliae as Aedes aegyptilarvacide, it has been conducted at laboratories Zoology in Biology Department, Mathematics and Natural Sciences Faculty, Syiah KualaUniversity. Aims of this research were estimating maximum time storage of fungi powder as effective larvacide and the number of bacteriacolonies were found in water used for experiment. This research was used Completely Randomized Design (CRD). The experiments consistof two factors i.e kind and saving time of fungi. The result show that the longer the storage time of Beauveria bassiana the more ineffectiveit was as larvacide and the least fungi colonies were found. Metharrizium anisopliae on the other hand, the longer it was kept, the moreeffective it was as larvacide and the more fungi colonies were found.

Abstract

The chitosan from shell of crabs (Portunus sanginolentus L.) waste was produced from deproteinization, demineralization, depigmentation,and deacetylation steps. This process was obtained 22.66% chitosan which identified by FTIR spectrophotometry and had 65.47% degree ofdeacetylation. This chitosan was used to adsorb methylene blue dye which fit to the Langmuir isotherm with 0.0312 l mg -1 equilibriumconstant of adsorption (K) and 4.20 mg g -1 maximum capacity of adsorption (q m).

Abstract

The aim of this research was to identify chemical compounds in cat eye dammar. The method included functional groups characterizationby infrared spectrophotometer and identification using Pyrolisis-GC/MS. Infrared spectra of crude sample showed that there were somefunctional groups such as alkyl, carbonyl, vinyl, and hydroxyl. Identification by Pyrolisis-GC/MS showed that dammar consisted of at least67 compounds. This natural gum components could be categorized into 4 groups, i.e. tetra cyclic hydrocarbon (30 compounds, 49.57%),penta cyclic (3 compounds, 2.56%), C 15 compounds (11 compounds, 17.09%), and other group (23 compounds, 18.26%). According to thePy-GC/MS data, brassicasterol is the highest relative concentration in dammar, i.e. 20%.

Abstract

The aim of this study was to bacteria isolation and characterization proteases from water swamp. The water swamp samples collected fromIndralaya for microbial isolation. Three isolates showed proteolytic index >1.00. The optimum pH of extraceluller proteases from A6S3,A4S3 and A15S3 were 7.5; 8.0; 8.0, respectively. The optimum temperature of A6S3, A4S3 and A15S3 protease were 40; 50; 50 oC,respectively. Effect of metal ion (Fe, K, Mn, and Zn) shown Fe and K were inhibit protease A6S3, all metal ion were inhibit protease A4S3and K, Mn and Zn inhibit protease A15S3. Study on the effect of metals ion and spesific inhibitors indicated that all protease weremetaloprotease. Their moleculer weights were determined by using SDS-PAGE and zymogram technique for A6S3 isolate were 70, 88, 106and 121 kD respectively. Whereas for isolate A4S3 was 138 kD and isolate A15S3 was 131 kD.

Abstract

The species of trees have different secondary compounds that need optimum extraction techniques. Appropriate extraction techniquesdetermine the quality and quantity of DNA produced. This research aims to found optimal of extraction methods and DNA isolation, thento created genome DNA in high quality and quantity, so that it can be using for genetic variation analyses in Suren (Toona sureni Merr.) byRandom Amplified Polymorphic DNA (RAPD). This study shows that DNA concentrates were 763.3 μg/ml, 180.0 μg/ml, 383.3 μg/ml, and436.7 μg/ml. While based on the results of PCR amplification using the primers OPD 03 shows that the four extraction methods used, the extraction method of number 3 has been able to produce genomic DNA with better quality and more number of bands, although the quantityis lower.
