Abstract

During a visit to Bali Island in September 2011, we found chilipepper plants exhibiting yellowing symptoms. The infected plants showed quite similar symptom to those of the recently reported Pepper yellow leaf curl virus from Israel and Japan, but there was no shortening of internodes or leaf rolling. We have amplified part of the genome of a virus associated with this disease using degenerate primers for members of the genus Polerovirus. The reverse transcription-polymerase chain reaction (RT-PCR) from symptom-bearing samples resulted in the amplification of a 650 bp band which is the expected size. RT-PCR from healthy samples did not produce an amplicon. Based on our results, we concluded that yellowing disease on chilipepper observed in Payangan, Gianyar, Bali is associated with a virus belonging to the genus Polerovirus. This is the first report on Polerovirus infection in chilipepper in Indonesia. Some researches for elucidation of the virus species and biological/molecular characters are being conducted.Key word: chilipepper, Indonesia, Polerovirus

Abstract

Leaf yellowing symptoms was commonly found in cucumber plants in Bali provinces, i.e. in Apuan and Bangli villages recently. Begomovirus infection is suspected as the causal agent, due to similar symptoms previously reported from cucumber plants in Java. In addition, Bemisia tabaci was observed in the field. The objective of this research was to identify the causal agent of leaf yellowing disease of cucumber in Bali. Virus detection and identification was conducted by polymerase chain reaction method using universal primers for Begomovirus, i.e. SPG1/SPG2. DNA fragment of 912 bp in size was successfully amplified from leaf samples. Analysis of nucleotide sequencing indicated that Begomovirus infecting cucumber plants in Bali has the highest homology (91%) with Squash leaf curl China virus (SLCCNV) isolate from Malaysia. This is the first report of SLCCNV infection in Bali.

Abstract

Yellowing vein banding disease has been reported infecting cucurbit plants in Bali since 2014. Similar vein banding symptom on chilli pepper was observed previously, and early diagnosis indicated infection of Polerovirus. The objective of this research was to confirm the presence of Polerovirus infection on cucumber plant showing yellow vein banding symptom in Bali. Reverse transcription polymerase chain reaction – based detection method was conducted using specific primer pairs PeVYV-CP-F-BamH1/ PeVYV-CP-R-Pst1followed by sequencing and nucleotide sequence analysis.  Specific DNA fragments of ± 650 bp was successfully amplified from field samples.  Nucleotide sequence analysis showed that the sequence has the highest similarity > 95% with Pepper vein yellow virus (PeVYV) infecting chili pepper from Indonesia (Bali, and Rembang), Japan, and Greece.

Abstract

The research about impact of using barrier of net color (red and white) for whitefly population on cayenne pepper plant in Kerta Village was conducted in order to know firstly about species of whitefly that existing on the cayenne pepper, secondly to know about impact of net color for populations of whitefly on the cayenne papper. The research was done using the randomized block design (RBD) with 3 treatments and 9 replications. The observation covered of the population of whitefly and symptom of virus infection. The results of this research showed whitefly were identified as Bemisia tabaci. The result indicated the lowest average populations of Bemisia tabaci occurred in the white net barrier treatment.

Abstract

Chili peppers (Capsicum frutescens L.) is one of the important vegetable in the world and one of the leading horticultural commodities in Indonesia. Pepper plants grown in all provinces in Indonesia and got priority for development because it has great potential economic value. These researches was conducted in order to determine the type of infecting Potyvirus on chili and know the proximity nucleotide sequence of Potyvirus infecting chili crop in Kerta village, Payangan, Gianyar, Bali, with the kind of Potyvirus in other areas. This study use RT-PCR technique to detect plant viruses and continued with tracing sequence coat protein gene nucleotide to its proximity to some of the isolates analyzed from GeneBank. The results showed that the infecting Potyvirus chili crop is Chili veinal motle virus species (ChiVMV) with size of approximately 900 bp DNA band, and based on phylogenetic analysis of coat protein gene nucleotide sequence, PayanganIndo isolate had high similarity (93%) and small genetic distance (0.041) with isolate of origin Thailand state.

Abstract

The aims of this experiment were to compare the shelf life of chili pepper seeds given dry heat treatment (DHT) and non dry heat treatment (NT). This experiment was conducted in Laboratory of Seed Technology Faculty of Agriculture, Udayana University. The study consisted of two experiments that attempt to use lots of chili seeds treated with dry heat treatment (DHT) and experimental use a lot of chili seeds without dry heat treatment (NT). This experiment uses a randomized completely design (RCD) with 10 replications.  Each type of chili pepper seeds tested each power shelf with 10 replications. Storage treatment performed at room temperature, which consists of non-storage (T0), storage for 2 weeks (T2), storage for 4 weeks (T4), storage for 6 weeks (T6) and storage for 8 weeks (T8). Thus, for each lots of chili pepper there are 50 experimental units. Treatment until 8 weeks of storage time did not affect the viability and vigor of pepper seeds either DHT or NT with value up to 80%, so they do not affect seed longivity.  The dry heat treatment (DHT) showed the values of seed and seedling vigor are higher compared to that of no dry heat treatment (NT).

Abstract

ABSTRACT Influence of Plastic Mulch on Crop Chili Pepper (Capsicum frutescens L.) Off  Season in the Kerta village. Chili is one of the important horticultural commodities in Indonesia and has a high economic value. To maintain its supplies, cultivation technology that able to produce the commodities continuously is needed by applying technology in the off-season cultivation. This study aims to determine the effect of plastic mulch on chili crop in the off-season. The experimental design was used a randomized block design with 3 treatments and 9 replications. The treatment given is the use of mulch consisting of silver plastic mulch (MP), black plastic mulch (MH), and without mulch/control (K). The variables observed in this study, ware, plant height, number of branches, soil temperature, symptoms of viral diseases, and yield per hectare. The results showed the plastic mulch give good results on chili crop in the off-season. However, the highest results of the chili plants was in the silver plastic mulch treatments with plant height (95.64 cm), number of branches (13.11 piece), and yield (12.31 ton/ha).   Keywords: chili pepper, off season, plastic mulch, yield

Abstract

Tolerance of Processing Delay to Physical and Physiological quality of Soya Bean (Glycine max L. Merril) The aim of this research is to figure out the time of the tolerance processing delay so that the physical quality and the physiological quality of soya bean seeds could be defended. This research was started in April 2017 at Jalan Cekomaria, Denpasar and also at the Lab. Pemuliaan Tanaman dan Teknologi Benih Faculty of Agriculture Udayana University. The design method was used for this research is a randomized complete block design (RCBD) using five treatment and five-time repetitions consist of W0= Without using process, W1= 1 day of Delaying process W2= 2 days of Delaying process, W3 = 3 days of Delaying process, W4 = 4 days of Delaying process. The statistic test and analyzing variance statistical test shows different result, the procedure followed by LSD method (Least Significance Different) with 95% of confidence level using costat program.The result of physical analysis test, shows that the role of the seeds which is without delaying process showing that the percentage of the seeds was 87,36% and the result of 4 days delaying process seeds are 11,256%. The result of the physiological test of the seeds using germinating test was, W0 (95,16%) and W4 (94,72%). The percentage of the vigor will be decreasing, which is W0 (80,52%) and W4 (34,4%). The equality control of the percentage was W0 (90,3%) and W4 (70,68%). Result of the observation of electrical conductivity showing that the lowest point of the electrical conductivity control is W0 (5,112 ? mhos cm¯² g¹) and the highest value of electrical conductivity was shown on the W4 (7,302 ? mhos cm¯² g¹).

Abstract

Influence Mosaic Diseases and Yellow to Long Bean (Vigna sinensis L.)Production at Perean Village, Baturiti, TabananThis study aims to determine how far the influence of viral infection with symptoms of mosaic and yellow against a decrease in crop yield long beans. This research consists of four activities are: 1. Determining the spread of disease areas; 2. Observation and data collection; 3. Calculation yields; and 4. Data analysis. Design used was a randomized block design (RBD) with three treatments and nine replications.Research result show that long bean plants in several locations in Perean village infected by BCMV which has mosaic symptoms and MYMV which has yellow symptoms. Percentage of mosaic virus symptoms is higher than yellow virus. The average of percentage of mosaic virus symptoms was 42,85%, while yellow virus was 32,70%. Plant beans with yellow virus symptoms causes the highest yield loss which amounted to 53,87% and plants with symptoms of mosaic virus by 43,02% when compared with healthy plants. Based on these studies it is necessary dissemination of information, about the importance of sanitation and the use of healthy seeds to controlling the long bean’s viral infection to related parties.
