Abstract

Malaria is one of the most prevalent tropical disease in the world. Until now, the use of antimalarials in malaria patients generated a lot of resistance so many scientists began the development of research using herbal plant ingredients, for example mistletoe (Dendrophthoe pentandra) This study was conducted to determine the effect of antiplasmodium in vivo. Antiplasmodium test conducted on P. berghei which were injected into mice suppression method. The results show that the Dendrophthoe pentandra extract has antiplasmodium activity with ED50 of 146,2 mg/kgBW. It can be concluded that Dendrophthoe pentandra has the potential as an antimalarial agent in vivo. Keywords: Dendrophthoe pentandra, malaria, antiplasmodium, mice, parasitemia,  Plasmodium berghei.

Abstract

A formulation containing 40% of honey, 30% of black cumin, 20% of propolis and 10% of royaljelly has been available in the market for the treatment of diabetes. Although each content ofthe formulation is proven to possess antioxidant and antihyperglycemic activities, its combinationeffect has not been evaluated yet. The aim of this study was to evaluate the effect of thisformulation on blood glucose level and pancreatic -cells of streptozotocin (STZ)-induced diabeticrats. Thirty six male Wistar rats (Rattus novergicus) aged 11 weeks with body weight 100-150 gwere used in this study. The rats were divided into 6 groups with 6 rats in each group. Group 1was non diabetic rats that were given aquadest. Group 2 was diabetic rats that were givenaquadest. Group 3 was diabetic rats that were given metformin at dose of 45 mg/kg BW. Group4-6 were diabetic rats that were given formulation tested at dose of 3, 6 and 12 mL/kg BW,respectively. All rats were induced by intraperitoneal injection of STZ at 60 mg/kg BW anddiabetic rats were then orally administered the formulation tested or metformin twice daily for14 days. Blood glucose level was monitored on day 10 and 17 after STZ induction. Rats weresacrificed and pancreas samples were taken for histopathological examination. The results showedthat the blood glucose level decreased significantly after seven days of treatment with metforminor fomulation tested and continued after 14 days of treatment. The blood glucose level ofdiabetic rats after 14 days of treatment returned to the normal level. The vacuolization of thepancreatic -cells of diabetic rats treated with metformin or with formulation tested were lowerthan untreated diabetic rats but still higher than non diabetic rats. In conclusion, the formulationtested has antihyperglicemic and protective effect on -cells damage in diabetic rats.Keywords: diabetic rats - honey - black cumin - propolis - royal jelly - antihyperglicemic

Abstract

Background: The alcoholic extract of kluthuk banana (Musa balbisiana Colla), a seeded banana, has been known to reduce gastric acid secretion on rats, but there is no explanation which part of kluthuk banana having that effect.Objective: To determine the effect of kluthuk banana both flesh and seed on rats gastric acid secretion in vitro.Methods: This study was conducted using isolated Wistar rat stomach. Fifty-four rats of 175-225 g weight were devided into 9 groups, each consisted of 3 male and 3 female rats. The stomach was suspended in an organ bath containing buffered serosal solution, and the gastric lumen was perfused continuously with unbuffered mucosal solution. The isolated preparation was stabilized for 5 times within 1 h and perfusate was spilled out. The perfusate was allowed to flow continuously, collected for 10 minutes duration and Fl+ concentration was measured, as basal FI4- concentration. Pretreatment was added to the unbuffered mucosal solution for 30 minutes, after which, histamine was added to the unbuffered mucosal solution for 80 minutes to induce gastric secretion. Perfusate from gastric lumen were collected every 10 minutes and FP-concentration was measured. The elevation of H* concentration was calculated in percentage, and was expressed as means ± SEM. ANOVA was used to analysis the data.Results: The elevation of H* concentration of groups treated with alcoholic extract of kluthuk banana seed at 12.46, 24.92 and 49.84 mg/kgBW was significantly lower than control group (p0.05).Conclusion: Alcoholic extract of kluthuk banana seed reduced rat gastric acid secretion in vitro, but did not those of alcoholic extract of kluthuk banana flesh.Keywords: Musa balbisiana Colla - gastric acid secretion - histamine - rat - stomach

Abstract

The inhibitory activity of heme polymerization of (1)-N-(2-nitrobenzyl)-1,10- phenantrolinium iodide and (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide have been done. This study aims to analyse the (1)-N-(2-nitrobenzyl)-1,10-phenantrolinium iodide and (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide as inhibitory of polimerization heme. Analysis of heme inhibtory polimerization activity used the experimental in vitro method. The activity showed by IC50 (the capable concentration of extract to inhibiting polymerization heme by 50% ). The IC50 value acquired by probit analysis. Assess IC50 of (1)-N-(2-nitrobenzyl)- 1,10-phenantrolinium iodide not to be identified, (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide and chloroquine by successively are 0,571±0,071; 25,498±1,876 mg/mL. The result showed the (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide had the highest value of the heme polymerization inhibitory activity than chloroquin, (1)-N-(2-nitrobenzyl)-1,10- phenantrolinium iodide hadn’t the heme polymerization inhibitory activity.

Abstract

This research was conducted to evaluate the anticancer activity of new compounds of benzyl-1,10- phenanthroline derivatives and their selectivity. In vitro anticancer activity of 11 benzyl-1,10-phenanthroline derivatives were conducted on three human cancer cell lines, cervical cancer (HeLa), myeloma (NS-1), and breast cancer (MCF-7) using MTT-based cytotoxicity assay. The cytotoxicity of each compound was assessed to normal Vero cell line by the same method. The in vitro anticancer activity and cytotoxicity was expressed by the concentration inhibiting 50% of the cell growth (IC50), and the selectivity index (SI) was determined by calculating ratio of the IC50 on Vero cell line and the human cancer cell lines. The results showed that among the 11 compounds tested, the (1)-N-(4-butoxybenzyl)-1,10-phenanthrolinium bromide exhibited the best in vitro anticancer activity with an IC50 27.60 ± 2.76 µM on HeLa, 6.42 ± 5.53 µM on NS-1 and 9.44 ± 2.17 µM on MCF-7 cell lines. Its SI were 377.65 ± 39.97 on HeLa, 6158.72 ± 5306.34 on NS-1 and 1140.11 ± 261.85 on MCF-7 cell lines. This study demonstrated that (1)-N-(4-butoxybenzyl)-1,10-phenanthrolinium bromide possessed a potential in vitro anticancer activity on cancer cell lines with high selectivity

Abstract

Introduction: Brucein A has been known to have antiplasmodial activity. Some new compounds were synthesized to increase their antiplasmodial activity, i.e 3-benzoylbruceinA, 3-dimethyl sulphate bruceine A, 3-choro benzoyl brucein A, and 3-chloro acetyl bruceine A.However, their antiplasmodial activity have not studied yet.Objectives: To know the in vitro antiplasmodial activity of 3-benzoyl brucein A, 3-dimethyl sulphate bruceine A, 3-choro benzoyl brucein A, and 3-chloro acetyl bruceine A.Methods: Antiplasmodial activity was conducted by incubating FCR-3 strain of P. falciparum with 3- benzoyl brucein A, 3-dimethyl sulphate bruceine A, 3-choro benzoyl brucein A, and 3-chloro acetyl bruceine A in various concentrations for 72 hours. Parasitemia after incubation period of each compound was calculated by making a thin smear stained with 5% Giemsa.Results: Semisynthetic compounds of bruceine A have antiplasmodial activity in vitro with IC50 value were 2.648 ± 1.30 ng/mL for 3-benzoyl bruceine A, 1.098 ± 0.510 ng/mL for 3-dimethyl sulphate bruceine A, 50.246 ± 0.207 ng/mL for 3-chloro benzoyl bruceine A and 67.951 ± 11.517 ng/mL for 3- chloro acetyl brucein A. The IC50 value of Bruceine A as the lead compound was 3.87 ± 2.530 ng/mLConclusion: The 3-dimethyl sulphate bruceine A showed the highest antiplasmodial activity among 4 semisynthetic compounds of Bruceine A.Keywords:bruceineA, semisynthetic compound, Plasmodiumfalciparum, invitroantiplasmodial activity.

Abstract

ABSTRACTIntroduction: Malaria infection could activate T cell helper 2 CD4+ which release cytokines IL-4, IL-5, IL-10 was a kind of immunosuppressant cytokines and prevented a cerebral tissue damage from the expression intercellular adhesion molecule-1 (ICAM-1) by TNF-α, ended with a cerebral malaria. The red fruit was proved as antioxidant but its effect to reduce TNF-α and expression of ICAM-1 in cerebral tissue endothelial cell remained unknown.Objectives: to know the effect of red fruit ethanol extract toward the change of TNF-α secretion number and ICAM-1 in Swiss male mice’s cerebral endothelial cell infected with P.berghei.Methods: Sixty Swiss male mice were divided in to 6 groups, group I was not given the red fruit (BM) ethanol extract and not infected P.berghei, Group II was given BM 260mg/kg BB but not infected P.berghei, group III, IV, V were given BM  130, 260, and 520 mg/kg respectively and infected with P.berghei. Red fruit extract or carrier substance was given for 4 weeks, P.berghei was given in the early of third week. TNF-α level check was done in third and ninth day after the injection, with TNF-α ELISA kit mouse. ICAM-1 cerebral endotel cell expression check was done in third and nine day with immunohistochemistry (IHC) coloring.Results: TNF-α level in third day after infected to the groups given red fruit extract (group III, IV, and V) showed no significant difference (p=0,839) compared to group which was not given red fruit but infected (group VI). TNF-α level in ninth day after infected to groups given red fruit ( group III,IV,V) lower (p<0,05) compared to groups which was not given red fruit but infected. ICAM-1 expression in third and ninth day after treatment to group given red fruit extract was lower (p<0,05) compared to groups given red fruit extract but infected. Conclusion: Red fruit ethanol extract ( P conoideus Lam) could decrease TNF-α level and ICAM-I expression in Swiss mice’ endothelial cerebral cells after infected by P.berghei.Key word: Pandanus conoideus Lam, TNF-α, ICAM-1 expression, Plasmodium berghei, Swiss mice

Abstract

ABSTRACTIntroduction: The immunity against malaria infection was very complex, involving humoral immunity and cellular immunity.  One of the methods to improve body immune response was using antioxidant. Red fruit (Pandanus conoideus Lam) containing carotenoid and tocopherol that have antioxidants effects increase phagocytosis activity of macrophage and proliferation activity of lymphocyte as immune response againts parasite infection.Objectives: This study was intended to know the effect of red fruit (P. conoideus Lam) ethanol extract on IFN-γ and nitric oxide (NO) levels in Plasmodium berghei infected Swiss mice malaria model.Methods: This quasi experimental study used post test only control group design. Sixty eight-week aged male Swiss mice were devided into 6 groups. Group I was not given red fruit ethanol extracts neither infected by P.berghei, group II was given 260mg/kg/day of red fruit ethanol extracts, and it was not infected by P.berghei, groups III, IV and V were given red fruit ethanol extracts at 130, 260 and 520mg/kg/day respectively and infected by P.berghei. Group VI was not given the extract, however it was infected by P.berghei as control group. The red fruit ethanol extracts or the carrier was given for 28 days and the mice were infected by P.berghei in the beginning of week 3. On the day 3 and 9 after infection, 2 mL blood was taken from 3 mice of each group for serum isolation.Results: There was not any significant difference (p>0.05) mean of IFN-γ levels in all groups neither on day 3 nor day 9 after infection. The nitric oxide levels mean of mice group which have received red fruit extracts at 130 and 260mg/kg/day on the day 9 and  in mice group which have received red fruit extracts 260mg/kg/day on day 3 were lower than they were in control group without any extract (group VI). However, the nitric oxide levels mean of mice group which have received 520mg/kg/day did not show any significant difference (p>0.05) compared with control group.Conclusion: The red fruit (P. conoideus Lam) ethanol extract did not show any significant effect on the IFN-γ levels, however at doses of 130 and 260mg/kg/day it decreased the nitric oxide level of P. berghei infected Swiss mice malaria model significantly. Keywords: Pandanus conoideus Lam, Interferon gamma (INF-γ), Nitric oxide (NO),  Plasmodium berghei, and Malaria ModelKeywordsPandanus conoideus Lam, Interferon gamma (INF-γ), Nitric oxide (NO), Plasmodium berghei, and Malaria Model

Abstract

Anemia is the most common hematologic abnormality in patients with human immunodeficiency virus (HIV) infection. This abnormality is associated with HIV infection itself, HIV-related opportunities infections or drug use. Zidovudine (AZT) is the most common cause of anemia in HIV patients. Recent study showed anemia in HIV patients is also associated with CD4 cell counts. Aim of this study was to evaluate the risk of anemia on HIV patients with different CD4 cell counts after AZT-based antiretroviral therapy (ART).This retrospective cohort study was conducted using medical record of HIV patients in Dr. Soetomo General Hospital, Surabaya. Subjects who fulfilled the inclusion and exclusion criteria were divided into two group i.e. HIV patients with CD4 cell counts 200-350 cell/mm3 and those with CD4cell counts ≥350 cell/mm3. All available demographics, clinical and laboratory data of subjects before and after AZT-based ART were then recorded and evaluated. Ninety-seven HIV patients (50 male and 47 female) were involved in this study. The result showed that the anemia incidence significantly increased after AZT-based ART (p<0.05), however no significantly different in anemia incidence, mean Hb level reduction and Hb level time reduction were observed between HIV patients with CD4 cell counts 200-350 cell/mm3 and those with CD4cell counts ≥350 cell/mm3(p>0.05). Gender, age, weight and clinical stage were not associated with anemia incidence (p>0.05). In contrast, anemia incidence is associated with Hb level before AZT therapy (p<0.05). In conclusion, the anemia incidence in HIV patients after AZT based ART is not associated with the level of CD4 cell counts, however it is associated with Hb levels before AZT therapy.
