Abstract

Rizosfer tanaman karet menyimpan potensi yang besar melalui keberadaan mikroorganisme yang bermanfaat untuk berbagai kepentingan, misalnya produk bioindustri seperti biofungisida dan biofertilizer. Pengembangan produk dari hasil eksplorasi mikroorganisme dari rizosfer tanaman karet akan menjadi peluang untuk meningkatkan peran dari perkebunan karet. Protein Sel Tunggal (PST) merupakan sel kering atau biomassa mikroorganisme seperti cendawan, khamir, bakteri, dan ganggang yang dapat digunakan sebagai sumber protein untuk pangan, selain mengandung protein tertentu, juga mengandung karbohidrat, lemak, vitamin, mineral, dan nutrisi lain yang dibutuhkan manusia. Penelitian ini bertujuan untuk mengetahui potensi mikroorganisme dari rizosfer tanaman karet (Hevea brasiliensis Muell.Arg.) untuk produksi protein sel tunggal. Penelitian dilakukan di Laboratorium Bioteknologi Tanah, Kimia dan Kesuburan Tanah di Departemen Ilmu Tanah dan Sumberdaya Lahan dan Laboratorium Cendawan Departemen Hama dan Penyakit Tanaman mulai  Februari 2011 sampai dengan Mei 2011. Analisa protein menggunakan metode Kjeldahl. Mikroorganisme dari rizosfer tanaman karet yang mampu menggunakan metanol dapat digunakan untuk produksi protein sel tunggal dan dapat dimanfaatkan sebagai sumber pangan dan pakan. Eksplorasi mikroorganisme dari rhizosfer tanaman karet memperoleh 22 isolat dan terseleksi 8 isolat untuk pengujian kadar protein yang terdiri 5 bakteri K2, K4, K8, K11, dan K13, serta 3 cendawan K15, K17, dan K18. Pengujian Hypersensitive Response, menghasilkan 2 isolat bakteri patogen K10 dan K13. Kandidat yang terbaik adalah isolat K4 dengan kadar protein 0,91%. Abstract

Abstrak. Degradasi bahan organik dari tanah gambut menghasilkan berbagai asam fenolat, CO2 dan CH4. Asam fenolat bersifat toksik dan menyebabkan terhambatnya pertumbuhan tanaman. Sebagian pestisida yang diaplikasikan jatuh ke tanah dan bereaksi dengan bahan organik tanah. Penelitian ini bertujuan mempelajari pengaruh penambahan pestisida terhadap ikatan asam fenolat pada tanah gambut serta menganalisis tingkat emisi CO2 dan CH4. Penelitian ini menggunakan 2 jenis pestisida yaitu herbisida paraquat diklorida dan insektisida Buthylphenylmethylcarbamate (BPMC). Dosis pestisida yang digunakan mengacu kepada dosis anjuran, yaitu 4 liter ha-1 untuk paraquat dan 1 liter ha-1 untuk BPMC. Perlakuan menggunakan 3 dosis (setengah, setara dan dua kali dosis anjuran: ?g kg-1): 1.104, 2.208, dan 4.416 untuk paraquat dan 485, 970, dan 1.940 untuk BPMC dengan satu kontrol. Setelah diinkubasi 1, 7, 14 dan 28 hari dilakukan analisis yang meliputi asam fenolat, residu pestisida, gugus fungsional, serta emisi CO2 dan CH4. Hasil penelitian menunjukan bahwa pemberian pestisida menurunkan jumlah asam fenolat di dalam tanah gambut. Penggunaan bahan aktif paraquat dan BPMC memperlihatkan pola yang sama terhadap perubahan konsentrasi asam fenolat dalam tanah. Dosis bahan aktif BPMC 485 ?g kg-1 menyebabkan penurunan residu pestisida lebih tinggi dibandingkan dosis yang lainnya yaitu sebesar 95% setelah 28 hari masa inkubasi. Secara umum penggunaan bahan aktif paraquat pada semua dosis yang diuji tidak meninggalkan residu setelah 28 hari masa inkubasi. Pemeriksaan gugus fungsional dengan FTIR tidak mendeteksi adanya penambahan gugus fungsional baru pada semua dosis dan jenis bahan aktif, namun perubahan intensitas puncak serapan dapat merupakan indikasi bahwa terjadi perubahan komposisi senyawa kimia dalam tanah gambut selama periode inkubasi. Penambahan pestisida tidak nyata mempengaruhi emisi CO2 dan CH4.Abstract. Degradation of organic matter from peat soil produces various phenolic acids, CO2, and CH4. The phenolic acid is known toxic and causes plant growth retardation. Some of pesticide is unintentionally drip on peat soil surface and reacts with organic matter. This study was aimed at evaluating the influence of pesticides on phenolic acid bondings as well as CO2, and CH4 emissions. Two types of pesticides were used: herbicide paraquate dichloride and insecticide Buthylphenylmethyl-carbamate (BPMC). The pesticide dosages were based on the recommended application, i.e. 4 liter ha-1 for paraquat and 1 liter ha-1 for BPMC. Treatments were consisted of 3 levels of dosage (half, equal and two-fold the recommended rates: ?g kg-1): 1,104; 2,208; and 4,416 for paraquat and 485, 970, and 1,940 for BPMC and a control treatment. After 1, 7, 14, and 28 days of incubation, soils in each treatment were analyzed for phenolic acids, pesticide residues, functional groups, and CO2 and CH4 emissions. The results showed that pesticide application reduced the total phenolic acid concentrations of peat soil. Paraquat and BPMC applications showed a similar pattern on soil phenolic acid concentrations. BPMC dosage of 485 ?g kg-1 reduced 95% of pesticide residues, which was the highest reduction compared to any other dosages. In general, paraquat application at the tested dosages left no pesticide residues after 28 days of incubation. The FTIR analysis did not detect appearance of any new functional groups under the tested rates and types of pesticides. However, altered intensity of the absorption peaks could be an indication of compositional changes of the chemical substances within peat soil during the incubation period. Pesticide application did not significantly affect CO2 and CH4 emissions.

Abstract

Abstrak. Biodiversitas tanah dipengaruhi oleh ekosistem lahan pertanian dan biodiversitas menggambarkan kesuburan tanah. Penelitian ini bertujuan untuk mempelajari pengaruh ekosistem lada (Piper nigrum) dan ubi kayu (Manihot esculenta) terhadap keragaman fauna tanah, mikroba tanah fungsional dan sifat kimia tanah. Pengambilan contoh tanah dilaksanakan di Kabupaten Lampung Timur pada akhir musim kemarau pada bulan Agustus 2013. Hasil identifikasi pada ekosistem lada dan ubi kayu menunjukkan bahwa terdapat 11 taksa biota tanah yang terdiri dari kelompok mesofauna (Acari dan Collembola) dan kelompok makrofauna ( Aranea, Chilopoda, Coleoptera, Diplura, Diplopoda, Hymenoptera, Isopoda, Isoptera dan Pseudoscorpion). Total kelimpahan fauna tanah yang ditemukan sebanyak 17529 individu terdiri dari 7210 individu pada ekosistem lada dan 10319 individu pada ekosistem ubi kayu. Populasi azotobacter, mikroba selulotik dan total bakteri pada ekosistem lada berbeda secara signifikan dengan ekosistem ubi kayu, sedangkan populasi bakteri pelarut fosfat dan total fungi tidak berbeda nyata. Sifat kimia tanah tidak berbeda signifikan antara kedua ekosistem ini. Berdasarkan analisis diskriman diketahui bahwa KTK, ldd, C-organik, pasir, liat dan total bakteri memiliki nilai fungsi sebagai faktor pembeda dan sifat-sifat ini dapat dimanfaatkan sebagai basis untuk pengelolaan tanah pada ekosistem lada dan ubi kayu.Abstract. Soil biodiversity is influenced by agricultural ecosystems and the biodiversity is an indicator of soil fertility. The objective of this research were to evaluate the effects of pepper (Piper nigrum) and cassava (Manihot esculenta) ecosystems on soil fauna biodiversity, population of functional microbes, and soil chemical properties. Soil samples were taken from East Lampung District in the end of dry season (August) 2013. The identification revealed 11 taxa of soil biota, i.e. group of mesofauna (Acari and Collembola) and group of macrofauna (Araneae, Chilopoda, Coleoptera, Diplura, Diplopoda, Hymenoptera, Isopoda, Isoptera, and Pseudoscorpion). The total abundance of soil fauna was 17529 individuals; 7210 in pepper and 10319 in cassava, but there was no significant difference in mean soil fauna abundance between the two ecosystems. Azotobacter, cellulotic and total bacteria showed significant differences in the abundance between pepper and cassava ecosystems, but the significant difference was not detected for phosphate solubilizing bacteria and total fungi. The soil chemical characteristics were not different between the two ecosystems. Based on the discriminant analysis, the variables that differentiated the two ecosystems were cation exchange capacity, exchangeable Al, organic carbon content, sand content, clay content, and total bacteria. These variables can be used as the basis for managing the pepper and casava ecosystems.

Abstract

Invertase Diversity, Novel Strain and Production Technology DevelopmentInvertase or β-fructofuranosidase (E.C.3.2.1.2.6) have been a valuable enzyme in food industry, so that research about enhancing invertase activity on an industrial scale has been reported massively. So far, the production of the invertase enzyme is highly dependent on the potential activity of the invertase enzyme derived from microorganisms. The development of enzyme production technology is also a concern in invertase research to obtain an efficient and inexpensive production system. So far, various developments in enzyme production technology and enzyme utilization have been carried out, including various innovations in immobilizing and increasing the stabilization of the invertase enzyme during the production process. The development of superior strains and enzyme production technology continues amid the discovery of the latest approaches such as genetic engineering, protein modification, and nanotechnology. Selain itu, pengembangan teknologi produksi juga menjadi perhatian dalam pengembangan produksi dan pemanfaatan enzim untuk mendapatkan sistem produksi yang efisien dan murah. Sejauh ini berbagai pengembangan teknologi produksi enzim dan pemanfaatan enzim telah dilakukan, diantaranya dengan berbagai inovasi dalam melakukan imobilisasi dan peningkatan stabilisasi enzim invertase selama proses produksi. Pengembangan strain unggul dan teknologi produksi enzim terus dilakukan di tengah ditemukannya pendekatan-pendekatan terkini seperti rekayasa genentika, modifikasi protein, dan teknologi nano. Artikel ini bertujuan untuk membahas variasi enzim invertase dari berbagai organisme, korelasi pengembangan strain mikrobia unggul terhadap peningkatan produksi enzim invertase, dan perkembangan teknologi produksi enzim invertase meliputi teknologi imobilisasi dan stabilisasi enzim invertase.

Abstract

Harapan forests is the first restoration forest in Indonesia, includes several different ecosystems. Different ecosystems have different characteristic to affect the diversity and abundance of Collembola. In the ecosystem, Collembola have an important role in biocontrol, decomposition, soil nutrient distribution, stimulation of soil microbial activity and as an alternative feed for predators. This study was aimed to investigate the diversity and abundance of Collembola in four ecosystems at Harapan forest, i.e. secondary forest, rubber forest, rubber smallholder and oil palm smallholder. To achieve the objective, soil samples were taken at 48 observation points in the four ecosystem. The soil samples were then extracted by Kempson Extractor. The diversity and abundance of Collembola in four ecosystems were categorized as medium to high. The total of individual and population of Collembola in the secondary and rubber forest ecosystems were likely to be higher than in the rubber and oil palm smallholders. This study had also identified four orders, 14 families and 31 genera of Collembola, where Isotomiella and Proisotoma dominated the genus of Collembola in the four ecosystems. Abstract

Latar belakang : Kemenkes RI (2015), mencatat rasio perawat terhadap penduduk sebesar 87,65 per 100.000 penduduk. Target 2019, 180 per 100.000 penduduk. Meningkatkan pendidikan keperawatan dari Prodi Diploma III ke S 1 merupakan upaya memenuhi rasio perawat terhadap penduduk. Meningkatkan pendidikan perlu dukungan dari keluarga seperti: dukungan emosional, penghargaan, instrumental dan informasi yang memberikan sugesti memiliki motivasi meningkatkan pendidikan Prodi Diploma ke S 1 Keperawatan. Tujuan : Mengetahui Hubungan Dukungan Keluarga Dengan Motivasi Melanjutkan Studi S1 Keperawatan Mahasiswa Semester VI Prodi Diploma III Keperawatan STIKES Bethesda Yakkum Yogyakarta Tahun 2017. Metode : Jenis penelitian analitik deskriptif correlation dengan pendekatan cross sectional. Penelitian dilakukan di STIKES Bethesda Yakkum Yogyakarta 17 Maret 2017. Sampel 84 responden, teknik total populasi. Analisis data menggunakan uji Spearman Rank. Hasil : Hasil analisis bivariat uji Spearman Rank dengan komputerisasi hasil p value (0,000) < (0,05) dengan nilai koefisien (0,615) tingkat keeratan kuat, maka H0 di tolak dan H? diterima. Kesimpulan : Ada Hubungan Dukungan Keluarga Dengan Motivasi Melanjutkan Studi S1 Keperawatan Mahasiswa Semester VI Prodi Diploma III Keperawatan STIKES Bethesda Yakkum Yogyakarta Tahun 2017. Saran : Peneliti selanjutnya kiranya menggali variabel faktor-faktor yang mempengaruhi motivasi mahasiswa Prodi Diploma III Keperawatan melanjutkan studi S1 Keperawatan, menggunakan metode wawancara sehingga mendapatkan analisis mendalam. Purpose: To know relationship between family support and motivation to take Bachelor Degree of Forth Semester Diploma Degree Students of STIKES Bethesda Yakkum Yogyakarta in 2017. Method: This was an analytic descriptive correlation research with cross sectional approach. The study was conducted at STIKES Bethesda Yakkum Yogyakarta on March 17, 2017. The sample of the study was 84 respondents taken by total population technique. The data analysis used was Spearman Rank test. Results: The result by computer based Spearman Rank: Result P value (0,000) < (0,05) with coefficient point (0,615) with strong cohesion. It means Ho is rejected and Ha is accepted. Conclusion: There is a relationship between family support and motivation to take Bachelor Degree of Forth Semester Diploma Degree Students of STIKES Bethesda Yakkum Yogyakarta in 2017. Suggestion: Further research is suggested discover factors that affect motivation of Diploma degree students to continue their study by interview method so that in depth analysis is obtained. Abstract

The feeding activity of soil animals was measured by using bait lamina test in three main ecosystems, i.e. the teak forest, home garden and rainfed paddy field. Two additional ecosystems in rainfed paddy field, i.e. the old (permanently established bund around paddy fields) and new bunds were examined as well. Three blocks of bait-lamina sticks (each block consisting of 16 individual sticks) were exposed at each location. The bait lamina were retrieved from the soil after two days and visually assessed. Each hole is designated as &Atilde;&cent;&acirc;&sbquo;&not;&Aring;&ldquo;fed&Atilde;&cent;&acirc;&sbquo;&not;&Acirc; (perforated) or &Atilde;&cent;&acirc;&sbquo;&not;&Aring;&ldquo;non-fed&Atilde;&cent;&acirc;&sbquo;&not;&Acirc; hole. The feeding rate is measured as the absolute number of &Atilde;&cent;&acirc;&sbquo;&not;&Aring;&ldquo;fed&Atilde;&cent;&acirc;&sbquo;&not;&Acirc; holes. Soil animals in the old bunds showed the highest feeding activity (55.20%), followed by home garden (39.10%), rainfed paddy field (16.50%), teak forest (15.60%), and new bund (7.80%). The frequency of animals attack to the bait strips also indicated the similar pattern as their feeding activity, i.e. high in the old bunds (0.90), followed by home garden (0.70), teak forest (0.40), new bunds (0.40) and rainfed paddy field (0.30), respectively. Abstract

ABSTRACTThe availability of nitrogen in soil is one of the limiting factors to support growth and rice productivity. Nitrogen-fixing bacteria have ability to utilize air nitrogen so it becomes available in the soil. The use of nitrogen-fixing bacteria could potentially reduce application of nitrogen fertilizer. The aim of the experiment was to determine the role of nitrogen-fixing bacteria in reducing inorganic N fertilizer on lowland rice. The research was conducted in April-August 2012 at the plastic house of Babakan Sawah Baru Experimental Station, IPB. The experiment was arranged in randomized block design with two factors, namely nitrogen fertilizer and type of bacteria. The dosage of N fertilizer (urea), i.e. 0, 50, 75 and 100 kg N ha-1. The types of bacteria, i.e. without bacteria, Azotobacter-like, Azospirillum-like, and consortium. The result of the experiment showed that N fertilization significantly affected to all variables except the percentage of empty grains per panicle, 1,000 grain weight, and N content of plant. The types of bacteria significantly affected to root dry weight, number of filled grain per panicle, greenness of leaf, uptake and content of nitrogen (shoot and grain), grain weight per plot. Consortium of bacteria were capable to reduce 25% the use of inorganic N fertilizer from recommendation doses (100 kg N ha-1) that based on the value of agronomic effectiveness.Keywords: Azospirillum-like, Azotobacter-like, consortium of bacteria

Abstract

Indigenous microorganisms effectively accelerate the decomposition of agricultural waste and agricultural waste that has been decomposed properly can be an organic fertilizer containing macro and microelements. The aim of the study was to determine the potential of indigenous cellulolytic fungi in the palm oil stems after replanting. The research was conducted at the Cikabayan Experimental Field Bogor Agricultural University, Dramaga, West Java. The experiment was conducted in October 2018 until January 2019, using a randomized complete block design consisting of four decomposition methods treatment: control, added with decomposer, chopping, and chopping+decomposer. The results showed that four isolates and isolate A were chosen to calculate the number of fungi at a dilution rate of 10-6 because they had the largest diameter. The identification results of isolate A were Trichoderma harzianum fungi and it was concluded that Trichoderma sp. in the palm oil stem after replanting, it has the potential as an indigenous cellulolytic fungus of oil palm trunks. The chopping+decomposer treatment can accelerate the weathering of the oil palm stem with a significant influence between the treatments that have met the requirements of compost maturity as specified in SNI 19-7030-2004 with ratio C/N 16.11 at 3 months after treatment. Abstract

Functional microbes of rhizosphere play important roles in nutrient transformation and controlling disease as well as in supporting plant growth and development. However, there is no study on the role of functional microbes on pineapple productivity. The purpose of this study was to investigate the abundance and diversity of soil functional microbes at different growth phases at two levels of productivity and their correlations to disease incidence. The research process included sampling of pineapple rhizospheric soil from vegetative and generative phases pineapples at low and high plant productivity sites, observations of disease incidence, and isolations of functional microbes. Functional groups of bacteria were Azotobacter, phosphate-solubilizing bacteria, potassium-solubilizing bacteria, antibiotics-producing bacteria, IAA-producing bacteria, and chitinolytic bacteria. The soil sampling method was simple randomized sampling at 6 locations with an area of each location ± 5 ha with a depth of 20 cm. Rhizosphere were taken in plants grown in high productivity area (>60tons/ha) and low productivity area (<60 tons/ha) in vegetative and generative phases. The results showed that potassium-solubilizing bacteria, chitinolytic bacteria, and IAA-producing bacteria were more abundant during the generative phase compared to those during vegetative phase. While Azotobacter, phosphate-solubilizing bacteria, and antibiotic-producing bacteria were more predominant during vegetative phase at various crop productivy. Total density of microbes was higher in soil with high crop productivity than that in soil with low crop productivity. The abundance of chitinolytic bacteria and IAA-producing bacteria had negative correlation with disease caused by Erwinia chrysanthemi and Phytophthora cinnamomi.
