Abstract

Mercury is well known for its high toxicity and strong affinity toward the thiol group of proteins. Mercury-resistant bacteria can  be used for detoxify mercury wastes due to the ability of these bacteria to reduce toxic inorganic mercury into mercury metal which is volatile and less toxic. The aim of this study was the selection of bacterial strains resistant to inorganic mercury, as well as to show their capacity to reduce mercury  in pure culture media Nutrient Broth. Six strains bacteria was isolated  and one of them,  A1.1.1. isolate was selected for its capacity to reduce mercury HgCl2 in culture media nutrient broth. The one bacterial strains belong to the spesies Klebsiella pneumoniae. Inoculated in pure culture, these strain showed a mercury reduction of 75% in 1 hour, 92% in 12 hours, and  99,4% in 24 hours. Therefore, this bacterium could be useful in detoxification of inorganic mercury wastes.   Keywords : Bacteria,  Klebsiella pneumoniae, Hg-resistant, detoxification

Abstract

The aims of this research were to study antibacterial activity, effective concentration and effect ofincreasing concentrations of ethanolic extract from mayana leaf (Coleus atropurpureus [L] Benth) onbacteria growth inhibition of Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa.Extraction was done by maceration using ethanol 96%. Antibacterial activity test was done by usingpitting method, Kirby and Bauer agar difussion with modification. The result was analyzed by Oneway anova, followed by Duncan test. The result showed that extract concentrations (5, 10, 20, 40 and80%) possess bacterial inhibition activity. Extract concentrations (20, 40 and 80%) were effectivelyinhibit S. aureus growth and on effective concentration to inhibit E. coli were at 10, 20, 40 and 80%,while for P. aeruginosa were at 5, 10, 20, 40 and 80% extracts. Abstract

UJI AKTIVITAS ANTIBAKTERI EKSTRAK KULIT BUAH MANGGIS (Garcinia mangostana L.) TERHADAP BAKTERI Klebsiella pneumonlaeBrando Melkianus1), Fatimawali1), Sri Sudewi1)1)PS. Framasi FMIPA Unsrat Manado, 95115ABSTRACTResearch has been carried out on the Antibacterial Activity Test of Mangosteen (Garcinia mangostana L.) Fruit Bark Extract against Klebsiella pneumoniae bacteria. This study aims to determine the antibacterial activity of mangosteen peel extract using the diffusion method with positive control of Ciprofloxacin and negative control of aquades and the test solution used by mangosteen peel extract. The result showed differences in extract concentrations of 25% (6.66 mm) categorized as medium, extract concentractions of 50% (8.83 mm) categorized as medium, extract concentractions of 75% (9.16 mm) categorized as medium, and extract concentractions of 100% (10.16 mm) categorized as strong in influencing the inhibition of the growth of Klebsiella pneumoniae bacteria. The result of this study also showed that mangosteen peel extract (Garcinia mangostana L.), has a broad-spectrum bioactive compound which means it can inhibit the growth of Gram negative bacteria.Keywords : Garcinia mangostana L, Klebsiella pneumoniae, CiprofloxacinABSTRAKTelah dilakukan penelitian tentang Uji Aktivitas Antibakteri Ekstrak Kulit Buah Manggis (Garcinia Mangostana L.) Terhadap Bakteri Klebsiella Pneumoniae. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak kulit buah manggis menggunakan metode difusi agar dengan kontrol positif Ciprofloxacin dan kontrol negatif aquades serta larutan uji yang digunakan ekstrak kulit buah manggis. Hasil penelitian menunjukkan Perbedaan konsentrasi ekstrak 25% (6,66 mm) termasuk sedang, konsentrasi ekstrak 50% (8,83 mm) termasuk sedang, konsentrasi ekstrak 75% (9,16 mm) termasuk sedang, dan konsentrasi ekstrak 100% (10,16 mm) termasuk kuat dalam mempengaruhi penghambatan pertumbuhan bakteri Klebsiella Pneumoniae. Hasil penelitian ini juga menunjukan bahwa Ekstrak kulit buah manggis (Garcinia Mangostana L.), memiliki senyawa bioaktif dengan spektrum luas artinya dapat menghambat pertumbuhan bakteri Gram negatif.Kata kunci : My Garcinia Mangostana L, Klebsiella Pneunoniae, Ciprofloxacin

Abstract

ABSTRACTDNA transformation is one of the methods for inserting DNA into bacterial cells. The current transformation method is widely used to transfer plasmids containing genetic material. This study aims to evaluate the results of plasmid transformation containing merB gene in Escherichia coli BL21(DE3) bacteria. The stages of the research carried out were preceded by the microbiological identification of the E. coli BL21(DE3) bacteria used as hosts. Then the plasmid transformation containing merB gene into the E. coli BL21(DE3) host cell using the heat shock method was carried out. The transformation results were evaluated by observing at the presence of E. coli BL21(DE3) colonies on agar Luria Bertani (LB) media containing ampicillin antibiotics. Plasmids in E. coli BL21(DE3) were isolated and analyzed by 1% agarose gel electrophoresis. The results showed the success of the transformation indicated by the growth of E. coli BL21(DE3) bacteria in agar LB media containing ampicillin and the visualization on agarose gel resulted that the plasmid which carried the merB gene could be transformed in to the E. coli BL21(DE3) bacteria.Keywords : Plasmids, merB genes, heat shock, Escherichia coli BL21(DE3)ABSTRAKTransformasi DNA merupakan salah satu metode untuk memasukkan DNA ke dalam sel bakteri. Metode transformasi saat ini dipakai secara luas untuk mentransfer plasmid yang mengandung bahan genetika. Penelitian ini bertujuan untuk mengevaluasi hasil transformasi plasmid yang mengandung gen merB pada bakteri Escherichia coli BL21(DE3). Tahapan penelitian didahului dengan identifikasi secara mikrobiologi bakteri E. coli BL21(DE3) yang digunakan sebagai inang. Selanjutnya dilakukan transformasi plasmid yang mengandung gen merB kedalam sel inang E. coli BL21(DE3) menggunakan metode heat shock. Hasil transformasi dievaluasi dengan melihat adanya koloni E. coli BL21(DE3) pada media agar Luria Bertani (LB) yang mengandung antibiotik ampisilin. Plasmid pada E. coli BL21(DE3) diisolasi dan dianalisis dengan elektroforesis gel agarose 1%. Hasil penelitian menunjukkan keberhasilan transformasi dengan adanya pertumbuhan bakteri E. coli BL21(DE3) pada media LB yang mengandung ampisillin dan hasil visualisasi pada agarose gel terlihat bahwa plasmid yang membawa gen merB dapat ditransformasikan ke dalam bakteri E. coli BL21(DE3).Kata Kunci : Plasmid, gen merB, heat shock, Escherichia coli BL21(DE3)

Abstract

ABSTRACTAcrylamide has been classified as a cancer causing compound or potentially carcinogenic in humans. Acrylamide is produced from foods that contain high carbohydrates with temperatures over 1200C in processing. One of foods that are popular and potentially produce acrylamide compounds was fried potato. This study aims to determine the levels of acrylamide contained in french fries using UV-Vis Spectrophotometry method. Samples were obtained from 3 fast food restaurants in the City of Manado. In this study the analysis of acrylamide compounds from 3 samples were carried out at 267 nm wavelength using an aquadest blank with 2 repetitions. The estimated result of three samples are K samples of 0.69 µg/g, M of 0.58 µg/g, and T samples of 0.67 µg/g. The three samples did not contain acrylamide because the levels obtained were still below the detection limit (LOD) of 1.54 µg/g. Keywords: Acrylamide, French Fries, UV-Vis Spectrophotometry, Fast Food Restaurants in Manado. ABSTRAKAkrilamida telah diklasifikasikan sebagai senyawa yang menyebabkan kanker atau berpotensi sebagai karsinogenik pada manusia. Akrilamida dihasilkan dari makanan yang mengandung karbohidrat tinggi dengan suhu lebih dari 1200C pada pengolahannya. Makanan yang banyak digemari serta berpotensi menghasilkan senyawa akrilamida salah satunya adalah kentang goreng. Penelitian ini bertujuan untuk menetapkan kadar akrilamida yang terkandung dalam kentang goreng dengan menggunakan metode Spektrofotometri UV-Vis. Sampel diperoleh dari 3 restoran cepat saji di Kota Manado.  Pada penelitian ini analisis senyawa akrilamida dari 3 sampel dilakukan pada panjang gelombang 267 nm menggunakan blanko aquadest dengan 2 kali pengulangan. Hasil perhitungan dari ketiga sampel adalah sampel K sebesar 0,69 µg/g, M sebesar 0,58 µg/g, dan sampel T sebesar 0,67 µg/g. Ketiga sampel tidak mengandung akrilamida karena kadar yang didapat masih berada di bawah batas deteksi (LOD) yang didapat yaitu 1,54 µg/g. Kata Kunci:   Akrilamida, Kentang Goreng, Spektrofotometri UV-Vis,  Restoran cepat saji di Manado

Abstract

Rhodamin B is a synthetic dye that was banned for use in foods and is certified as a hazardousmaterial according to Minister of Health of Indonesian Republic No. 722/Menkes/Per/IX/1988because it can cause liver, kidney, and spleen damage, and followed by anatomical changes in theform of organ enlargement. Rhodamin B dye is often used for a food and beverage products. Thestudy was conducted to identify Rhodamin B on cakalang fufu in the market of Manado city.Cakalang fufu samples were taken from four markets in the city of Manado, namely : the Paal 2market, Bahu market, Karombasan market and Bersehati 45 Market. Samples were extracted by twomethods, the method of extraction followed by purification and extraction method using wool yarnand absorption followed by identification using thin layer chromatography (TLC) and detected withUV light 254 nm and 366 nm. Rhodamin B content is read using a UV-Vis spectrophotometry. Theresult showed that out of eight samples examined with two tests, there are four samples containingRhodamin B with higher levels of the average value of Rhodamin B in samples, that is from themerchant two of Bahu Market is 0,054 ug/ml, at merchant two Karombasan market is 0.184 ug/ml,34merchant one at Bersehati 45 Market is 0.103 ug/ml and merchant two at Bersehati 45 is 0.514 ug/ml.Based on these results, some of cakalang fufu circulating in Manado City Markets are not safe forconsumption.Keywords: Rhodamin B, Cakalang fufu, TLC and UV-Vis spectrophotometry

Abstract

ABSTRACTJackfruit leaves is a plant that work as medicine on healing process of skin diseases,especially in wounds. The purpose of this study is to determine the quality of ointment fromethanolic extract of Jackfruit leaves at the concentration on 5%, 10% and 15% and the effectof the open wound healing in rabbits. Type of this research is laboratory experimental. In thisresearch maceration was done as the extraction method to got the extract of Jackfruit leaves.The ointment of ethanolic extract from jackfruit leaves made within 3 concentrations of 5%,10% and 15%. On the testing ointment using organoleptic test, homogenity test and pH test.The effectiveness assay to heal the open wounds using 5 groups, Betadine ointment (positivecontrol), basic ointment (negative control), the ointment of ethanolic extract from Jackfruitleaves 5%, 10%, and 15% of the 3 rabbits with a 1.5 cm was done and observed until 7 days.The data were processed statistically using One Way ANOVA (Analysis Of Variant) andfollowed by LSD (Least Significant Different). Results of this study showed that the ointmentof ethanolic extract from Jackfruit leaves on the concentration of 5%, 10% and 15% producequalified ointment of organoleptic test, homogenity test and pH test, also give the effect ofopen wound healing in rabbits and the most effective cure is the ointment of ethanolic extractfrom Jackfruit leaves 15%.Keywords: ethanolic extract of Jackfruit leaves, ointment, wound, rabbit.

Abstract

ABSTRACT Algae have long been used for the treatment of various types of diseases. Ulva lactuca is one of the algae, which contains bioactive substances as antimicrobial, antifungal, and antioxidant. This study aims to determine the microorganisms growth inhibitory activity from Ulva lactuca algae obtained from the of Lembeh Strait waters in City of Bitung against microorganisms Escherichia coli, Staphylococcus aureus and Candida albicans. Ulva lactuca algae was extracted using maceration method with ethanol solvent and fractionated with methanol, n-hexan and chloroform solvents. Testing of antimicrobial activity using agar diffusion method. The result showed that extracts and fractions of Ulva lactuca algae did not have antimicrobial activity against the microorganisms Escherichia coli, Staphylococcus aureus and Candida albicans. The chemical composition of algae is influenced by season, geographical distribution, as well as environmental factors such as temperature, water, salinity, light, nutrition, and mineral availability.Keywords: Ulva lactuca, Antimicrobials, Maseration, Fractions, Agar DiffusionABSTRAK Alga telah lama digunakan untuk keperluan pengobatan berbagai jenis penyakit.  Ulva lactuca merupakan salah satu alga yang memiliki kandungan zat bioaktif sebagai antimikroba. Penelitian ini bertujuan untuk mengetahui adanya aktivitas penghambat pertumbuhan mikroorganisme dari alga Ulva lactuca yang diperoleh dari perairan Selat Lembeh kota Bitung terhadap mikroba Escherichia coli, Staphylococcus aureus, dan Candida albicans. Alga Ulva lactuca diekstraksi menggunakan metode maserasi dengan pelarut etanol dan difraksinasi dengan pelarut methanol, n-hexan, dan kloroform. Pengujian aktivitas antimikroba menggunakan metode difusi agar. Hasil penelitian menunjukkan ekstrak dan fraksi dari alga Ulva lactuca tidak memiliki aktivitas antimikroba terhadap Escherichia coli, Staphylococcus aureus, dan Candida albicans. Komposisi kimia alga dipengaruhi oleh musim, distribusi geografis, serta faktor lingkungan seperti suhu, air, salinitasi, cahaya, nutrisi, dan ketersediaan mineral.Kata kunci : Alga Ulva lactuca, Antimikroba, Maserasi, Fraksi, Difusi Agar

Abstract

ABSTRACTLactic Acid Bacteria (LAB) are a group of Gram-positive bacteria that produce lactic acid as the major metabolic end product. They are cocci or rods, nonsporulating and are anaerobic or facultative anaerobes bacteria. Most LABs are probiotics that are known to have good benefits to health, such as inhibiting some pathogens. This study was aimed to examine the potential probiotics properties of LAB isolates from Romaine lettuce fermentation which is included  antibacterial activity of isolates. The isolates are spread on MRS agar  supplemented with 1% of CaCO3 and then purified by using streak method to obtain pure isolates. The results showed that there are 4 isolates from Romaine lettuce fermentation which have the potential to inhibits some pathogens.Key words: Lactic acid bacteria, fermentation, Romain lettuce, potential probiotic ABSTRAKBakteri Asam Laktat (BAL) merupakan sekelompok bakteri Gram positif yang menghasilkan asam laktat sebagai produk akhir metabolisme. Berbentuk kokus atau batang, tidak memiliki spora dan bersifat anaerob atau fakultatif anaerob. Sebagian besar BAL merupakan probiotik yang diketahui memiliki manfaat baik bagi kesehatan, seperti memiliki aktivitas antibakteri. Penelitian ini bertujuan untuk menguji kemampuan isolat BAL dari hasil fermentasi selada Romain sebagai probiotik potensial yang mencakup aktivitas antibakteri isolat. Isolat bakteri ditumbuhkan pada media MRS agar  yang ditambahkan 1% CaCO3 kemudian dimurnikan menggunakan metode gores (streak) sehingga diperoleh isolat murni yang kemudian diuji. Hasil penelitian menunjukkan bahwa ke empat isolat yang diperoleh dari hasil fermentasi selada romain yang berpotensi memiliki aktivitas penghambatan bakteri patogenKata kunci: Bakteri asam laktat, fermentasi, selada Romain, probiotik potensial
