Abstract

ABSTRACTNanoparticles can increase the solubility of compounds, reduce the treatment dose and increase absorption. Flavonoid and phenol compounds in red galangal can inhibit the growth of bacteria that have been resistant to antibiotics. The bioactivity of red galangal compounds is expected to increase when the size of nanoparticles increases antibacterial activity. This study aims to determine the inhibition of the content of Red Galangal nanoparticles (Alpinia purpurata K.Schum) using chitosan on the growth of Klebsiella pneumoniae bacterial urine isolates of UTI antibiotic-resistant Ciprofloxacin. Nanoparticles extracts of red galangal rhizome was made using ionic gelation method, using extract concentration of 0.4%, tested for antibacterial activity using the wells method with a comparison of chitosan 0.4%, acetic acid 5%, red galangal extract 25%. Red galangal rhizome nanoparticles gave antibacterial activity value to the bacteria Klebsiella pneumoniae of 13.5 mm. Therefore, red galangal extracts nanoparticles can inhibit the growth of Klebsiella pneumoniae urine isolate in patients with urinary tract infections with strong inhibitory categories based on Davis and Stout categories.Keywords: Alpinia purpurata K. Schum, Klebsiella pneumonia, Nanoparticles. ABSTRAK Nanopartikel dapat meningkatkan kelarutan senyawa, mengurangi dosis pengobatan dan meningkatkan absorbsi. Senyawa flavonoid dan fenol dalam lengkuas merah mampu menghambat pertumbuhan bakteri yang telah resisten terhadap antibiotik. Sifat bioaktivitas senyawa lengkuas merah diharapkan akan meningkat ketika berukuran nanopartikel untuk meningkatkan aktivitas antibakteri. Penelitian ini bertujuan untuk mengetahui daya hambat kandungan nanopartikel rimpang Lengkuas Merah (Alpinia purpurata K.Schum) menggunakan kitosan terhadap pertumbuhan bakteri Klebsiella pneumoniae isolat urin penderita ISK yang resisten antibiotik Ciprofloxacin. Nanopartikel ekstrak rimpang lengkuas merah dibuat dengan menggunakan metode gelasi ionik, dengan menggunakan konsentrasi esktrak sebesar 0,4 %, dilakukan pengujian aktivitas antibakteri mengunakan metode sumuran dengan pembanding kitosan 0,4%, asam asetat 5%, ekstrak lengkuas merah 25%. Nanopartikel rimpang lengkuas merah memberikan nilai aktifitas antibakteri terhadap bakteri Klebsiella pneumoniae sebesar 13,5 mm. Oleh karena itu nanopartikel ekstrak rimpang lengkuas merah dapat menghambat pertumbuhan bakteri Klebsiella pneumoniae isolate urin penderita infeksi saluran kemih dengan kategori daya hambat kuat berdasrkan kategori Davis dan Stout.Kata Kunci: Alpinia purpurata K. Abstract

ABSTRACT             White galangal rhizome (Alpinia galanga L. Swartz) is one of the plants which has the antibacterial properties from the zingiberaceae family. Empirically it can treat various diseases such as chest pain, throat inflammation, stomach inflammation, rheumatism, diabetes and skin diseases. White galangal rhizome contains flavonoids, tannins, quinones and terpenoids. This study aims to determine the antibacterial activity of white galangal rhizome extract (Alpinia galanga L. Swartz) and test its inhibition against the growth of Klebsiella pneumoniae bacteria which are resistant to ceftriaxone antibiotics. The white galangal rhizome was extracted by maceration method using 96% ethanol solvent and antibacterial activity was tested using the disc and well method with a concentration difference of 100%, 75%, 50%, 25% and 12.5%. The results showed that white galangal rhizome extract (Alpinia galanga L. Swartz) had antibacterial activity against the bacterium Klebsiella pneumoniae and could inhibit the growth of Klebsiella pneumoniae bacteria. On the disc method for concentrations of 100%, 75%, 50%, 25% and 12.5%, the inhibitors were categorized as medium. Whereas in the well method for concentrations of 100% and 75%, the inhibitors were categorized as strong, for concentrations of 50%, 25%, and 12.5%, which have a medium category.Keywords: White Galangal Rhizome (Alpinia galanga L. Swartz), Klebsiella pneumoniae ABSTRAK Rimpang Lengkuas putih (Alpinia galanga L. Swartz) merupakan salah satu tanaman yang memiliki khasiat sebagai antibakteri dari family zingiberaceae. Secara empiris dapat mengobati berbagai penyakit seperti nyeri dada, radang tenggorokkan, radang lambung, rematik, diabetes dan penyakit kulit. Rimpang Lengkuas putih mengandung senyawa flavonoid, tanin, kuinon dan terpenoid. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak rimpang Lengkuas putih (Alpinia galanga L. Swartz) dan menguji daya hambatnya terhadap pertumbuhan bakteri klebsiella pneumoniae yang resisten terhadap antibiotik seftriakson. Rimpang Lengkuas putih diekstraksi dengan metode maserasi menggunakan pelarut etanol 96% dan dilakukan pengujian aktivitas antibakteri dengan menggunakan metode cakram dan sumuran dengan perbedaan konsentrasi 100%, 75%, 50%, 25% dan 12,5%. Hasil penelitian menunjukkan bahwa ekstrak rimpang Lengkuas putih (Alpinia galanga L. Swartz)  memiliki aktivitas antibakteri terhadap bakteri Klebsiella pneumoniae dan dapat menghambat pertumbuhan bakteri Klebsiella pneumoniae. Pada metode cakram untuk konsentrasi 100%, 75%, 50%, 25% dan 12,5% memiliki daya hambat yang dikategorikan sedang. Sedangkan pada metode sumuran untuk konsentrasi 100% dan 75%  memiliki daya hambat yang dikategorikan kuat, untuk kosentrasi 50%, 25%, dan 12,5% memiliki daya hambat yang dikategorikan sedang.Kata Kunci: Rimpang lengkuas putih (Alpinia galanga L. Abstract

ABSTRACTDNA transformation is one of the methods for inserting DNA into bacterial cells. The current transformation method is widely used to transfer plasmids containing genetic material. This study aims to evaluate the results of plasmid transformation containing merB gene in Escherichia coli BL21(DE3) bacteria. The stages of the research carried out were preceded by the microbiological identification of the E. coli BL21(DE3) bacteria used as hosts. Then the plasmid transformation containing merB gene into the E. coli BL21(DE3) host cell using the heat shock method was carried out. The transformation results were evaluated by observing at the presence of E. coli BL21(DE3) colonies on agar Luria Bertani (LB) media containing ampicillin antibiotics. Plasmids in E. coli BL21(DE3) were isolated and analyzed by 1% agarose gel electrophoresis. The results showed the success of the transformation indicated by the growth of E. coli BL21(DE3) bacteria in agar LB media containing ampicillin and the visualization on agarose gel resulted that the plasmid which carried the merB gene could be transformed in to the E. coli BL21(DE3) bacteria.Keywords : Plasmids, merB genes, heat shock, Escherichia coli BL21(DE3)ABSTRAKTransformasi DNA merupakan salah satu metode untuk memasukkan DNA ke dalam sel bakteri. Metode transformasi saat ini dipakai secara luas untuk mentransfer plasmid yang mengandung bahan genetika. Penelitian ini bertujuan untuk mengevaluasi hasil transformasi plasmid yang mengandung gen merB pada bakteri Escherichia coli BL21(DE3). Tahapan penelitian didahului dengan identifikasi secara mikrobiologi bakteri E. coli BL21(DE3) yang digunakan sebagai inang. Selanjutnya dilakukan transformasi plasmid yang mengandung gen merB kedalam sel inang E. coli BL21(DE3) menggunakan metode heat shock. Hasil transformasi dievaluasi dengan melihat adanya koloni E. coli BL21(DE3) pada media agar Luria Bertani (LB) yang mengandung antibiotik ampisilin. Plasmid pada E. coli BL21(DE3) diisolasi dan dianalisis dengan elektroforesis gel agarose 1%. Hasil penelitian menunjukkan keberhasilan transformasi dengan adanya pertumbuhan bakteri E. coli BL21(DE3) pada media LB yang mengandung ampisillin dan hasil visualisasi pada agarose gel terlihat bahwa plasmid yang membawa gen merB dapat ditransformasikan ke dalam bakteri E. coli BL21(DE3).Kata Kunci : Plasmid, gen merB, heat shock, Escherichia coli BL21(DE3)

Abstract

ABSTRACT Algae have long been used for the treatment of various types of diseases. Ulva lactuca is one of the algae, which contains bioactive substances as antimicrobial, antifungal, and antioxidant. This study aims to determine the microorganisms growth inhibitory activity from Ulva lactuca algae obtained from the of Lembeh Strait waters in City of Bitung against microorganisms Escherichia coli, Staphylococcus aureus and Candida albicans. Ulva lactuca algae was extracted using maceration method with ethanol solvent and fractionated with methanol, n-hexan and chloroform solvents. Testing of antimicrobial activity using agar diffusion method. The result showed that extracts and fractions of Ulva lactuca algae did not have antimicrobial activity against the microorganisms Escherichia coli, Staphylococcus aureus and Candida albicans. The chemical composition of algae is influenced by season, geographical distribution, as well as environmental factors such as temperature, water, salinity, light, nutrition, and mineral availability.Keywords: Ulva lactuca, Antimicrobials, Maseration, Fractions, Agar DiffusionABSTRAK Alga telah lama digunakan untuk keperluan pengobatan berbagai jenis penyakit.  Ulva lactuca merupakan salah satu alga yang memiliki kandungan zat bioaktif sebagai antimikroba. Penelitian ini bertujuan untuk mengetahui adanya aktivitas penghambat pertumbuhan mikroorganisme dari alga Ulva lactuca yang diperoleh dari perairan Selat Lembeh kota Bitung terhadap mikroba Escherichia coli, Staphylococcus aureus, dan Candida albicans. Alga Ulva lactuca diekstraksi menggunakan metode maserasi dengan pelarut etanol dan difraksinasi dengan pelarut methanol, n-hexan, dan kloroform. Pengujian aktivitas antimikroba menggunakan metode difusi agar. Hasil penelitian menunjukkan ekstrak dan fraksi dari alga Ulva lactuca tidak memiliki aktivitas antimikroba terhadap Escherichia coli, Staphylococcus aureus, dan Candida albicans. Komposisi kimia alga dipengaruhi oleh musim, distribusi geografis, serta faktor lingkungan seperti suhu, air, salinitasi, cahaya, nutrisi, dan ketersediaan mineral.Kata kunci : Alga Ulva lactuca, Antimikroba, Maserasi, Fraksi, Difusi Agar

Abstract

ABSTRACT            Lactic acid bacteria (LAB) is a group of bacteria that produce lactic acid as the major metabolic end product of carbohydrate fermentation. LAB are highly beneficial because of their probiotic potential properties and as functional starter cultures in food fermentation. This study was aimed to identify the LAB species isolated from romaine lettuce fermented product by using molecular identification method with 16S rRNA gene marker. The fermented product was diluted and spread onto MRS agar supplemented with 1% of CaCO3 and then purified by using streak method. Both isolates were positive Gram bacteria and gave negative results from catalase test. The result of molecular identification showed that LAB from romaine lettuce fermented product have each 99 and 100% similarity to Enterococcus faecium that known have properties as probiotic potential and functional culture starter.Key words: lactic acid bacteria, fermentation, probiotic, Enterococcus faecium, Lactuca sativa var. Longifolia Lam.             ABSTRAK             Bakteri asam laktat (BAL) merupakan bakteri yang menghasilkan asam laktat sebagai metabolit utamanya dalam fermentasi karbohidrat. BAL merupakan bakteri yang menguntungkan karena memiliki aktivitas probiotik potensial dan dapat berguna sebagai starter dalam proses fermentasi makanan. Penelitian ini bertujuan untuk mengidentifikasi spesies BAL yang diisolasi dari hasil fermentasi selada romain menggunakan metode identifikasi molekuler dengan gen penanda 16S rRNA. Cairan hasil fermentasi diencerkan dan ditebar pada media MRS agar yang telah dicampur dengan CaCO3 1% dan kemudian dimurnikan menggunakan metode streak plate. Kedua isolat merupakan bakteri Gram positif dan menunjukkan hasil negatif pada uji katalase. Hasil identifikasi molekuler menunjukkan bahwa  kedua BAL hasil fermentasi selada romain memiliki kemiripan masing-masing 99 dan 100 % dengan Enterococcus faecium yang diketahui memiliki aktivitas probiotik.Kata Kunci: bakteri asam laktat, fermentasi, probiotik, Enterococcus faecium, Lactuca sativa var. longifolia Lam.

Abstract

ABSTRACTPalm horse weevil contains medicinal properties that are useful in the process of woundhealing. The purpose of this study is to to examine formulate an ointment of palm horse(Ipomoea pes-caprae (L) Sweet) weevil extract and to test the ointment to heal open woundon the skin of rabbits. This study is experimental descriptive in the laboratory. Palm horseweevil extracts used in the manufactured of ointment formulation 10%, 15 % and 20 %. Theresults showed that the palm horse weevil formulation meets the ointment test requirementaccording, making ointment formulation using cassava tuber extract used as 5 rabbits.Divided into 5 treatment gropus, namely, test the ointment treatment group, negative controlgroup (basic ointment) and positive control group (chloramphenicol ointment). Observed thehealing effects for 7 days. The results showed that from day-1 to day-7 observations burnssuffered diminution in diameter. It was concluded that concentration of 10 % cassava extractontment has healing effect and increasing the concentration of 15 % and 20 % containedshowed more effective and speed up the healing process.Keywords : Extract leaf palm horse , ointment, wound healing, rabbit.

Abstract

This study aimed at finding out the effectivity of Salacca (Salacca zalacca [Gaertn]Voss) rind extract on blood sugar decrease of white male wistar (Rattus norvegicus L.)induced with sucrose. Completely randomized design method was used in this experiment.The subject of the experiment were 15 white male wistar which were divided into 3 (three)groups namely negative control group, treatment group with the dose of 150 mg extract/kgbody weight, and positive control group using glibenclamide 0.45 mg/kg body weight. Dataobtained from the examination of the fasting blood sugar were 30 minutes after inductionwith sucrose and at minute 15, 30, 60, and 120 after treatment was given. The data isprocessed using ANOVA and LSD test to compare between the negative control group,treatment, and a positive control. The analysis revealed a significant difference betweennegative control and treatment, negative control and positive control, but did not show asignificant difference between treatment groups with the positive control. The conclusion ofthis study is Salacca rind extract has an effect on blood sugar levels decrease of white malewistar induced with sucrose.Keywords: Salacca zalacca (Gaertn.) Abstract

The aims of this research was to determine the effect of ethanol extract paku sisik nagaleaves against lipid peroxidation in male wistar liver induced by CCl4. Method of thisresearch used laboratory experiment using 9 white male wistar were divided into 3 treatmentgroups. All groups were given treatment during ten days. The first group (negative control)was given distilled water ad libitum, the second group was given ethanol extract of paku sisiknaga leaves 97.02 mg/kg body weight per oral and the third group (positive control) wasgiven Curcuma 45 mg/kg body weight per oral. On the tenth day, after two hours oftreatment all groups were given CCl4 1mL/kg body weight intraperitonial, eighteen hoursafter that rats in all groups were dissected and taken his liver. Then rat liver analyzed byTBARS method (Thiobarbiturate Acid Reactive Subtance) to measure the levels of MDA(Malondialdehyde) which is an indicator of lipid peroxidation in the rat liver. Data wereanalyzed by SPSS ver.20, significant difference between treatments was tested by one-wayANOVA. The results showed a significant difference with 95% confidence level (α = 0.05)with a mean value of MDA 3,828 μM negative control group, group of ethanol extract ofpaku sisik naga leaf was 2,666 μM and 2,211 μM for positive control group. Ethanol extractof paku sisik naga leaves with dose 97.02 mg/kg body weight can prevent the process of lipidperoxidation in rat liver.Keywords: Drymoglossum piloselloides L. Abstract

This paper reports the identification and determination of benzoate preservative level intomato sauce on the market in Manado. Extraction of the benzoate with diethylether anddetermination of the preservative (benzoic acid) was determined using UV-Visspectrophotometer. Qualitatively, it was found that the sauce samples contained benzoate.Quantitatively, it was found that four samples of tomato sauce used benzoate as apreservative. two of them used benzoic above the threshold set uot in Decree No. Permenkes.722/Menkes/Per/IX/88.Keywords: benzoate, ketchup, Manado, spectrophotometer UV- Vis.

Abstract

ABSTRACTThis research aims to determine the phytochemical compounds and testing the analgesiceffects of ethanol extract of kucing-kucingan plant (Acalypha indica Linn.). Phytochemicaltesting conducted according to Harbone methods and analgesic activity of rats reaction timeis determined based on the thermal response after giving the ethanol extract of A indica L.This study used 20 female Wistar rats were divided into a treatment group was given 0,035mg / kg extract of leaves, stems and roots, negative control receive aquadest and positivecontrol using aspirin. Reaction time data were analyzed using One Way ANOVA followed byDuncan's test multipe Range Test. The results showed that the ethanol extract of A.indica L.contains alkaloids and saponins in the extract while the third type of flavonoid and tanninfound only in leaf extracts. Testing the analgesic activity of ethanol extract A. indica L.showed that the leaf extract gave the highest reaction time of 3.16 seconds, and stem and rootextracts followed by 1.72 seconds and 2.85 seconds.
