Abstract

Background: In vitro studies reveal that LMP1 induces the expression of many factors for invasion and metastasis. Objective: The aims of this study were to investigate the role of LMP1 expression on the status of Nand M that take place either directly or sequentially through its relation with the expression IL4, IL6. IL 10. IFN-gamma. Gran2yme B and with protease MMP2. MMP9. either in the serum or in the tumour tissue. Methods: Forty one 41 EBER-RISH confirmed NPC cases with parallel serum were used for this study. Immunohistochemistry was used to study the expression of proteins in the tissue. The serum level of proteins were determined with ELISA. Statistical analysis used Lisrel 8.50. Results: No correlation between the expression of LMP1 with the status of Nand M of the tumour either directly or that sequentially took place through the expression of the cytokines and MMPs either in the tumour tissue or the serum level. The serum level of MMP9 was correlated with the serum level of IL6, the status of N. and the expression of IL 10. Status N was correlated with serum level of MMP2. Conclusion: There is a process that sequentially took place through the increase of the cytokines. MMPs, either in the tumour tissue or serum level and N. M status. LMP1 expression was not involved in the process. Key words: NPC - metastasis - MMP - cytokine

Abstract

Five major structural types of immunoglobulin in man can be distinguished. i. e. immunoglobulin G (abbreviated to IgG), IgM, IgA, IgD and IgE. Each type of immunoglobulin can synthesize specific antibody according to the inducing antigen. The specificity of immunoglobulin molecule is found in the antigen-binding fragment.Actually the specificity and the diversity of the immunoglobulin is controlled by the gene in the chromosome. Abstract

Telah diketahui bahwa untuk aktivasi limfosit B diperlukan faktor pertumbuhan yang disebut limfokin atau sitokin. Banyak bukti menunjukkan bahwa limfosit T yang teraktivasi menghasilkan substansi yang dapat memacu proliferasi atau diferensiasi limfosit B. Dewasa ini telah dapat diidentifikasi berbagai limfokin yang disebut sebagai interleukin (IL) seperti IL-2, IL-3, IL-4, IL-5, IL-6 dan IL-7 yang ikut berperan dalam aktivasi sel B. Namun, mekanisme aktivasi sel B oleh limfokin sampai sekarang masih terus berkembang dan dipelajari. Untuk dapat lebih memahami aktivasi sel B, dalam penelitian ini telah dilakukan pembuatan suatu T cell line, dan telah dikarakterisasi, yakni menunjukkan fenotipe Thyl+, Lyt1+2+, Dalam penelitian ini berbagai aktivator poliklonal lipopolysaccharide (LPS), ConA, dextran sulfat dipakai untuk memacu T cell line menghasilkan limfokin. Aktivitas limfokin pada supernatannya kemudian dianalisis dengan menggunakan blastogenesis assay dengan 3H-thymidine dan hemolytic plaque assay.Hash penelitian menunjukkan bahwa T cell line menghasilkan faktor pertumbuhan yang memacu proliferasi sel B (B cell growth factor II = BCGF II atau IL-5) dan faktor yang memacu sel B untuk berdiferensiasi (B cell differentiating factor = BCDF atau IL- 6) dan menghasilkan igM dan IgG.Key words : blastogenesis assay -- hemolitic plaque assay -- B cell activation -- T cell line -- lymphokines

Abstract

Background : Mahkota dewa (Pheleria macrocarpa (Scheff, Boerl) is an herbal medicine which has been used since many years as traditional medicine in Indonesia against infections.Objective: The study was aimed to know the effect of mahkota dewa extract on cellular immune response, in particular NK1.1 cell activities.Methods: Forty C57BL/6 mice used in this study were divided into 8 groups. In this study, different doses of etanolic extract of mahkota dewa leaf were used i.e. 1.05 mg, 2.1 mg, and 4.2 mg/20g BW. Groups I, II, and III were treated with 1 mg, 2.1 mg, and 4.2 mg/20 g BW of the extract daily for 30 days. In group IV, V, and VI the extract was given simultaneously during 30 days, then the mice were infected with 104 cfu of Listeria monocytogenes (A TCC-191151 for stimulating their immune responses. Whereas group VII was untreated control group, and group VIII received only Listeria monocytogenes. All mice were then sacrificed 48 hours after the last treatment. Splenocyte NK1.1 cells were collected then cultured with YAC-1 (ATCC:TIB 1601 target cells for killing activity assay, expressing NKG2D, CD122 and IFNy assay. Results: The results showed that an oral administration of the extract significantly increased the killing activity of splenic NK 1.1 cell against the target, Y AC-1 cell. Moreover, the extract promoted the secretion of IFN-y from NK1.1 cells, and also induced expression of both surface molecule NKG2D and CD 122. The strongest effect stimulation was on the dosis of 2.1 mg/20 g BW.Conclusion: The extract had effect to augment splenic NK1.1 cell activities, as indicated by increasing their killing activity, expression of surface molecules and IFN-y production.Key words: pha/eria macrocarpa - splenic NK1.1 cell activity - YAC-1 cell - IFNy - NKG2D and CD122 surface molecules

Abstract

Cancer is a disease, a public health problem, which is found in the world as well as in Indonesia. Ingeneral, some of cancer theraphies are ineffective, characterized by the resistance performance of cancer cell line,the exposed normal cell and by the side effects. Nowadays, studies to fi nd the specifi c and safely anti-cancerdrugs were increased by the time. Several studies revealed that Curcuma mangga Val. Rhizome contains somesecondary metabolites, essential or non-essential oil, which has cytotoxic activities to the cancer cells. Basedon these anti-cancer potentials, this study has several aims to recognize anti-cancer selectivity and molecularmechanism by inducting apoptosis and inhibiting myeloma cell proliferation. To C. mangga Val. essential oil,immunocyto chemical test was performed to determine the expression of p53, caspase-9, Bcl-2, H-Ras proteinwhile TUNEL test was performed to determine the number of apoptosis cells.The results of this study shown that anti-cancer molecular mechanism of C. mangga Val. essential oil tomyeloma cell line was performed by increasing apoptosis; by increasing the expression of pro-apoptosis p53,caspase-9 protein and reducing protein which is increasing proliferation Bcl-2 and H-Ras.

Abstract

Mahkota Dewa [Phaleria macrocarpa (Scheff). Boerl.] is used traditionally to treat cancers in Indonesia. Extract methanol of the leaves displayed a small LC50 value (63.16 μg/ml) on BST (Brine Shrimp Lethality test) assay; therefore a phytochemical study of this extract was undertaken. A new benzophenoic glucoside was isolated from the methanol extract and identified as 4,5-dihydroxy,4’-methoxybenzophenone-3-0-β-D-glucoside (Phalerin) based on its spectroscopic data. Phalerin was cytotoxic to myeloma cell line (NS-1) (in vitro) having IC50 of 83 μg/ml or 1.9 x 10-1 mM.Key words: Phaleria macrocarpa, Phalerin, myeloma

Abstract

BACKGROUNDThe leaves of Nerium indicum Mill. have been utilized traditionally to cure cancer. By using Bioassay guided extraction and isolation method, three compounds (NiO-1, NiO-2, NiO-3) were isolated from an active fraction of the CHCl3 extract of N. indicum leaves. NiO-2 identified as 5α- oleandrin is the best cytotoxic compound on HeLa cervical cancer cell in vitro among the other two. However mechanism of action of the compound hasn’t been evaluated yet. The aims of this study were to determine the mechanism of action at molecular level.METHODSThe action mechanism of 5α-oleandrin on HeLa cervical cancer cells was analyzed by staining the cells with Hoechst 33342; the agarose gel electrophoresis was aimed to determine the DNA fragmentation and the western blotting was aimed to determine the Bcl-2 and Bax protein expression.RESULTSIncubation of HeLa cervical cancer cell with 5α-oleandrin at the concentration 3,47x10-4 mM (24 hours) followed by staining with Hoechst 33342, a broken up light blue color of nucleus was observed (compared with intensive color of untreated control). By gel electrophoresis (at the same concentration of the tested compound), a smear band at about 200 bp was observed. In addition, cells treated with 5α-oleandrin displayed a decreasing of the Bcl-2 protein expression and increasing of the Bax protein expressionCONCLUSION5α-oleandrin induced HeLa cervical cancer cells DNA fragmentation observed by the presence of a smear band at about 200 bp indicative the apoptotic occurrence. 5α-oleandrin induces apoptosis by reducing the Bcl-2 protein expression but the Bax protein expression increases.

Abstract

ABSTRACT---In this paper, we analyze and compute distribution of thermal neutron flux in core of Kartini’s Reactor using “Swadaya” detector, both for axial (function of z) and radial (function r) direction. First, “Swadaya” detector is calibrated by activation plate method, which the output from detector is current. The result of calibration is thermal neutron flux (ф) dependent on current (I) that can be written as ф = 4,57x1020I - 2,74x1011. Distribution for neutron thermal flux for axial direction and radial direction respectively Ф = 5,10 x 1012 + 5,99 x 1013 and ф = 1.08 x 1012 + 1,00 x 103 which in reactor core ф has largest value in center position for radial direction.   Keywords: flux neutron distribution, nucleus reactor, Swadaya detector

Abstract

ABSTRAK   Temu kunci (B.pandurata) mengandung senyawa-senyawa yang berpotensi antikanker. Penelitian ini bertujuan untuk mengkaji efek kemoprevensi ekstrak etanolik rimpang temu kunci pada karsinogenesis kulit terinduksi ultra violet. Ekstraksi serbuk rimpang B. pandurata dilakukan dengan metode maserasi  menggunakan etanol. Hewan uji yang digunakan adalah mencit Balb/C betina umur 28 hari yang dicukur rambut punggungnya, dikelompokkan menjadi 5 kelompok perlakuan, yaitu kelompok kontrol dan empat  kelompok   perlakuan ekstrak etanolik temu kunci yang diberikan secara oral. Induksi karsinogenesis dengan paparan UV dosis 0,167 J/cm2/hari, sebanyak 60 paparan (5 kali paparan/minggu). Efek penghambatan karsinogenesis kulit  dipelajari pada tingkat insidensi dan multiplisitas kanker. Hasil uji menunjukkan bahwa ekstrak etanolik temu kunci mampu menurunkan angka insidensi dan multiplisitas kanker kulit pada mencit Balb/C terinduksi UV. Ekstrak tersebut berpotensi   dikembangkan sebagai agen kemoprevensi kanker.   Kata Kunci: Boesenbergia pandurata, kemoprevensi, karsinogenesis , ultra violet,

Abstract

Diabetes mellitus tipe 2 merupakan jenis diabetes yang paling banyak terjadi. Berbagai macam terapi telah digunakan untuk menangani DM (Diabetes Mellitus) tipe 2, namun masih terdapat keterbatasan. Penelitian ini dilakukan untuk mengetahui pengaruh MT-SPM terhadap histopatologi pankreas dan jumlah sel normal Langerhans pada tikus model diabetes melitus tipe 2. Penelitian ini merupakan penelitian eksperimental murni dengan desain Posttest Control Group. Sebanyak 27 ekor tikus Sprague Dawley jantan dibagi menjadi 3 kelompok: kontrol normal (K(-)) yaitu 9 tikus sehat, kontrol diabetes (K(+)) yaitu 9 tikus yang diinduksi DM tipe 2 dengan 60 mg/kg BB STZ (i.p) dan 120 mg/kg BB NA (i.p), dan kelompok perlakuan (P) yaitu 9 tikus yang diinduksi DM tipe 2 dan diberi perlakuan dengan 0,1 ml/200 g BB (i.p) setiap 3 hari selama 10 kali. Pada hari ke-30 setelah perlakuan, tikus dikorbankan dan diambil jaringan pankreasnya untuk diuji histopatologi dengan pewarnaan Hematoksilin-Eosin. Hasil tersebut dianalisis dengan program Image J. Hasil penelitian menunjukkan terjadi perbaikan pada profil histopatologi pankreas dan terdapat peningkatan jumlah sel normal di islet Langerhans pada kelompok perlakuan yang diberi MT-SPM dibandingkan kelompok kontrol positif.
