Abstract

Sembung (Blumea balsamifera) Leaf Extract Improves Testis Histology of High-Fat Diet-Induced RatsABSTRACTHigh fat and high cholesterol diet cause hyperlipidemia, leading to various health problems including reproductive health. The purpose of this study was to examine the effect of sembung (Blumea balsamifera) leaf extract on testicular histology profile of high-fat-diet-induced wistar. This research used 16 adult male rats (Rattus norvegicus), aged 3-4 month, weighing 150-200 g, and randomly divided into two groups. Eight rats were treated with distilled water and eight rats were treated with 2 mg/mL B. balsamifera extract. High-fat diet was a 30 days of porcine fat feed. The results showed that the diameter of seminiferous tubules, the number of spermatogenic cells of spermatogonium A, spermatocytes Pakiten and spermatid 16 were increased by giving sembung leaf extract for 50 days (p <0.05). These results suggested that the sembung leaf extract improves testis histology of high-fat diet-induced rats. Keywords: high-fat diet, rat, sembung leaf, seminiferous tubules, spermatogenic cellsABSTRAKMakanan tinggi lemak dan tinggi kolestrol menyebabkan hiperlipidemia yang menimbulkan masalah pada sistem reproduksi. Penelitian ini bertujuan untuk mengetahui manfaat pemberian ekstrak daun sembung (Blumea balsamifera) terhadap profil tubulus seminiferus dan sel-sel spermatogenik pada tikus wistar yang diinduksi pakan tinggi lemak. Sebanyak 16 ekor tikus wistar jantan dewasa (Rattus norvegicus) umur 3-4 bulan, berat 150-200 g, secara random dibagi dua kelompok, yaitu 8 ekor tikus kelompok kontrol (aquades steril) dan 8 ekor tikus kelompok perlakuan (ekstrak daun sembung dosis 2 mg/mL). Tikus diinduksi pakan tinggi lemak berupa lemak babi selama 30 hari. Pada prettest dilakukan pemeriksaan diameter tubulus seminiferus dan sel-sel spermatogenik. Hasil penelitian menunjukkan bahwa rata-rata diameter tubulus seminiferus tikus meningkat signifikan setelah pemberian ekstrak daun sembung. Peningkatan secara signifikan juga diikuti oleh spermatogonium A, spermatosit Pakiten dan spermatid 16 (p<0,05). Dapat disimpulkan bahwa ekstrak daun sembung memperbaiki histologi testis tikus wistar yang diinduksi pakan tinggi lemak.Kata Kunci: daun sembung, pakan tinggi lemak, sel spermatogenik, tikus, tubulus seminiferus

Abstract

ABSTRAK?Seiring dengan proses penuaan, kemampuan gen yang mengontrol viabilitas sel mengalami penurunan, sejalan dengan meningkatnya aktivitas gen yang menyebabkan kematiansel dan berujung pada patogenesis? penyakit. Dengan memanipulasi aktivasi dan non-aktivasi gen yang terlibat dalam proses penuaan, maka kesehatan akan meningkat dan proses penuaandapat dihambat bahkan dikembalikan. Saat ini telah ditemukan teknologi CRISPR/Cas-9 yangmemungkinkan manipulasi pada tingkat genetik. CRISPR sebenarnya merupakan bagian prosesfisiologis pada bakteri, yang merupakan salah satu sistem imunitas bakteri. Namun saat iniCRISPR telah banyak dibuktikan dan diaplikasikan untuk reprogram DNA pada sel mamaliadan dapat mengembalikan perubahan genetik yang tidak diinginkan.Kata kunci: Penuaan, CRISPR/Cas-9, anti-aging medicine, manipulasi genetik ?ABSTRACTAs all living thing undergo aging, genes that maintain cellular viability and vitality are downregulated, in line with upregulated expression of genes that promote cells senescence anddisease-related pathogenesis. By manipulating the initiation and inhibition of aging-relatedgene expression, the youthful health may be systemically restored. Recently, a new technologycalled CRISPR is found to be usefull for manipulating gene and rapidly transform senescentcells to regain youthful function and structure. CRISPR is originally developed in nature by bacteria as a way to destroy the DNA of viruses which part of their immune system. A naturalversion of CRISPR has been adapted by scientists to enable the reprogramming of cellular DNAto rid cells of unfavorable genetic changes. Abstract

Buah anggur dikenal memiliki aktifitas antioksidan yang tinggi dan bermanfaat mencegah terjadinya stres oksidatif. Penelitian ini bertujuan mengetahui peran antioksidan dalam buah anggur untuk mencegah terjadinya stres oksidatif yang diakibatkan aktivitas fisik berlebih dengan indikator kadar AST darah mencit. Penelitian ini adalah penelitian eksperimental murni dengan rancangan completely randomized menggunakan post-test only control group design yang terdiri dari empat kelompok mencit yaitu kontrol, perlakuan satu dengan 1ml/hari ekstrak buah anggur, perlakuan dua dengan 0,5ml/hari ekstrak buah anggur dan perlakuan tiga dengan 0,25ml/hari ekstrak buah anggur. Pemberian ekstrak buah anggur dilakukan selama tujuh hari, kemudian diberi perlakuan overtraining berupa renang, pengambilan darah dan pengecekan kadar AST. Hasil penelitian menunjukan kadar AST antar perlakuan tidak berbeda nyata (p>0,05), dan dengan uji lanjutan Least Significant Difference (LSD) didapatkan hasil tidak terjadi penurunan AST secara bermakna pada perlakuan tiga (P3) jika dibandingkan dengan kontrol (p>0,05). Sedangkan pada perlakuan satu (P1) dan perlakuan dua (P2) terjadi penurunan kadar AST secara bermakna (p<0,05). Dari hasil penelitian ini dapat disimpulkan bahwa ekstrak buah anggur menurunkan kadar AST darah mencit dengan aktivitas fisik berlebih dengan dosis optimal 0,5ml/hari.

Abstract

Dokter hewan praktisi seringkali mengabaikan dampak psikologi hewan terhadap kondisi kesehatan hewan. Penatalaksanaan penyakit hewan selalu dikaitkan dengan tindakan medis veteriner. Namun kenyataannya timbulnya gejala klinis penyakit seperti anoreksia, diare, dan demam tidak selalu disertai dengan penyebab yang jelas. Hal ini kemungkinan besar terkait dengan kondisi psikologi hewan. Psikoneuroimunologi (PNI) merupakan cabang ilmu kedokteran yang merupakan gabungan dari ilmu psikologi, neurologi, endokrinologi serta imunologi yang memiliki paradigm sendiri. PNI mengkaji bagaimana sistem imun tubuh merespon keadaan homeostasis dan patologis dipengaruhi oleh keadaan psikologis. Hubungan otak dengan sistem imun terjadi melalui sel di HPA axis (hipotalamo-pituitary-adrenal axis), yang melibatkan hormon sitokin, dan melalui sel di jalur ANS (autonomic nervous system). Berbagai macam stresor ini akan mengaktifkan hipotalamus mensekresikan CRH, yang kemudian merangsang pituitary menghasilkan ACTH. ACTH kemudian berikatan dengan reseptornya di kelenjar adrenal untuk menginduksi sekresi Epinefrin (EPI) dan Norepinefrin (NE). Sudah dipahami bahwa limfosit memiliki reseptor untuk EPI dan NE sehingga stres ini akan mengakibatkan modulasi imunitas. Psikologi menjadi kajian yang sangat penting dalam dunia kedokteran hewan terkait dengan modulasi imun akibat psikologi. Oleh karena itu pengembangan Psikoneuroimunologi dalam tatalaksana penyakit pada hewan menjadi sangat penting, disamping penanganan medis.

Abstract

Moderate Physical Exercise Increases Serum Vascular Endothelial Growth Factor and Stromal Cell-Derived Factor-1 Level Physical exercise affects many systems in the body that play a role in maintaining physical fitness and slowing down the aging process. This study aimed to identity VEGF and SDF-1 as the mechanism of exercise-related anti-aging therapy. This study was a randomized pretest-posttest control group design study, using 24 Wistar rats (Rattus norvegicus), male, 2.5-3 months old, weighing 180-200 g, and healthy. Rats were divided into two groups (n = 12). The control group was a sedentary lifestyle-treated group (P0) and the treatment group was subjected to moderate physical exercise for 4 weeks (P1). Both VEGF and SDF-1 levels were examined using the ELISA method. The results showed that moderate physical exercise increased VEGF (43.88 ± 6.24 to 69.80 ± 10.04 pg mL-1; p< 0.001) and SDF-1 (1.82±0.17 to 3.81±0.39 ng mL-1; p< 0.001) levels significantly. This study, therefore, suggested the possibility of VEGF and SDF-1 as a mediator of moderate physical exercise-induced anti-aging effect. Pelatihan fisik mempengaruhi berbagai sistem dalam tubuh yang berperan mempertahankan kebugaran fisik dan memperlambat proses penuaan. Penelitian menunjukkan bahwa kadar vascular endothelial growth factor (VEGF) dan stromal cell-derived factor-1 (SDF-1) menurun seiring dengan penuaan. Penelitian ini bertujuan membuktikan bahwa pelatihan fisik sedang meningkatkan kadar VEGF dan SDF-1 sebagai salah satu mekanisme penghambat proses penuaan. Penelitian ini menggunakan randomized pre-post test control group design dengan menggunakan 24 ekor tikus wistar (Rattus norvegicus) jantan sehat dengan umur 2,5 - 3 bulan, berat badan 180 -200 g yang dibagi dalam dua kelompok (n= 12 ekor). Kelompok kontrol tidak diberikan perlakuan atau sedentary lifestyle (P0) dan kelompok perlakuan diberi pelatihan fisik sedang selama 4 minggu (P1).  Kadar VEGF dan SDF-1 diperiksa menggunakan ELISA. Hasil penelitian menunjukkan bahwa pelatihan fisik sedang meningkatkan kadar VEGF (43,88±6,24 menjadi 69,80 ± 10,04 pg mL-1; p<0,001) dan SDF-1 (1,82 ± 0,17 to 3,81±0,39 ng mL-1; p< 0,001). Hasil penelitian ini menunjukkan kemungkinan VEGF dan SDF-1 sebagai mediator efek yang memperlambat penuaan. Namun, perlu dilakukan penelitian lebih lanjut untuk mengidentifikasi korelasi antara peningkatan kadar VEGF dan SDF-1 dengan fenotip penuaan.

Abstract

ABSTRAKTujuan dari penelitian ini adalah untuk menggambarkan prevalensi penyakit Baliziekte pada sapi potong di Kabupaten Majene, Sulawesi Barat. Penelitian ini menggunakan metode penelitian kuantitatif dengan pendekatan analisis data sekunder. Data kasus kejadian Baliziekte pada sapi potong di Kabupaten Majene, Sulawesi Barat pada tahun 2018-2020 didapatkan dari Dinas Pertanian, Peternakan dan Perkebunan Kabupaten Majene. Hasil analisis menunjukkan bahwa prevalensi penyakit Baliziekte pada sapi potong di Kabupaten Majene mengalami peningkatan dalam rentang waktu  dua tahun terakhir. Jumlah prevalensi penyakit Baliziekte pada tahun 2018 adalah 0,789 %, pada tahun 2019 terjadi penurunan menjadi 0,716 % dan meningkat pada tahun 2020 menjadi 1,257 %. Kecamatan Sendana merupakan satu-satunya daerah dengan peningkatan jumlah kasus Baliziekte pada sapi potong setiap tahunnya, sedangkan Kecamatan Banggae Timur mengalami penurunan jumlah kasus Baliziekte yaitu dari prevalensi 0,078% di tahun 2018 menjadi 0% di tahun 2019 dan 2020. Kecamatan Tubo Sendana merupakan daerah dengan jumlah kasus dan prevalensi tertinggi pada tahun 2020, sedangkan Kecamatan Banggae dan Pamboang merupakan daerah dengan data kasus dan prevalensi 0%. Kata kunci: sapi potong, prevalensi, baliziekte, Kabupaten MajeneThe Prevalence of Baliziekte Disease in Beef Cattle in Majene, West SulawesiABSTRACTThe purpose of this study was to illustrate the prevalence of Baliziekte disease in beef cattle in Majene, West Sulawesi. This study was quantitative research by analyzing the secondary data analysis approach. Data on the Baliziekte incidence in beef cattle in Majene, West Sulawesi in 2018-2020 were obtained from the Department of Agriculture, Livestock and Plantation of Majene Regency. The results of the analysis showed that the prevalence of Baliziekte disease in beef cattle in Majene Regency has increased in the last two years. The prevalence of Baliziekte disease in 2018 was 0.789%, decreased to 0.716% in 2019, and increased sharply to 1.257% in 2020. Sendana District was the only area with an increase in the number of Baliziekte cases in beef cattle every year, while the East Banggae District underwent depletion in the number of Baliziekte cases, from a prevalence of 0.078% in 2018 to 0% in both 2019 and 2020. Tubo Sendana District was the area with the highest number of cases and prevalence in 2020, while the Districts of Banggae and Pamboang were both maintained a 0% case and prevalence throughout the years.Keywords: beef cattle, prevalence, baliziekte, Majene

Abstract

One important aspect that determines the quality of human life is sexual life. Therefore,  sexual activity to be one part in the assessment of the quality of human life. In everyday life, not infrequently encountered sexual problem in various communities, an estimated 322 million people both men and women experience sexual dysfunction worldwide. One of the factors that cause sexual dysfunction that is very important is lifestyle. Lifestyle that includes physical activity would interfere with organ function so that the body is not healthy and does not fit include decreased sexual fitness. Many studies have reported that the results of unhealthy lifestyle, especially diet and sleep patterns, can interfere with a person's sexual ability. In addition to lifestyle, excessive physical activity also decreases sexual function. But in contrast, a good control on lifestyle and physical activity will enhance the ability of sexual activity.

Abstract

Human life expectancy increasing progressively declines with technological developments and poor lifestyle. The cause of death due to an unhealthy lifestyle among other physical activity that is not in accordance with scientific principles accelerates the aging process. The development of the science of Anti - Aging Medicine is expected to improve human welfare through increased life expectancy. Several attempts have been made, both in research and clinical reports, to find the factors of aging that can do prevention and control. These efforts are expected to be a reference to slow down the aging process and preventing the aging occur. One of the efforts that have been made in various countries is the use of stem cells. The utilization of stem cells is a breakthrough in the field of medicine to address degenerative diseases and slow down the aging process. The ability of proliferation, differentiation and self- renewal of stem cells can be used to replace the cells, tissues, and organs damaged by the aging process. The characters and the unique benefits of this increase the number of stem cell field research. One area of ??research that is related to the formation of the stem cell is physical exercise factor. The benefits of physical training on endogenous stem cells give new hope in the field of anti - aging medicine. Regular physical training in accordance with scientific principles (exercise physiology), is expected to prevent anti-aging through activation of endogenous stem cells in the body naturally.

Abstract

BACKGROUND: Hypoxia has been shown to be able to induce adiposity. However, the mechanism and factors involved in this effect still remains unclear. Hence, we sought to investigate the role of oxygensensitive factors regarding hypoxia-induced adiposity in nematode Caenorhabditis elegans.METHODS: The C. elegans were grown on nematode growth medium (NGM) agar plates seeded with Escherichia coli OP50 at 20°C. The ratio of width/body length was measured using the morphometry analysis. Fat accumulation was examined using Sudan Black methods. Protein levels of sterol binding protein (SBP)-1 were assessed by immunoblotting. Lifespan assay was performed at 20°C and was monitored every two days.RESULTS: The results showed that of all mutant used, only hif-1 mutant which did not experience an increase in the ratio of width/body length (p>0.05) and fat accumulation (p>0.05), indicating that hypoxia-inducible factors (HIF)-1 plays an important role in the pathogenesis of hypoxia-induced adiposity. Both siah-1 and skn-1 mutants experienced SBP-1 protein elevation (p<0.05), and increased fat-6 mRNA expression (p<0.05) which were not experienced by a hif-1 mutant (p>0.05) further supporting that HIF-1 acts as an upstream regulator fromSBP-1.CONCLUSION: In general, the results of this study provide evidences of the involvement of the transcription factor HIF-1 in inducing adiposity under the hypoxic conditions. However, we did not find the involvement of seven in absentia homolog-1 (Siah-1) and skinhead-1 (SKN-1).KEYWORDS: hypoxia, adiposity, fat, HIF-1, Siah-1, SKN-1, C. elegans

Abstract

Overtraining is a high-volume, high-intensity, long-duration, and high-frequency training. The heavier physical training, the more oxygen needed for metabolism. Increasing oxygen usage causes an elevation of electron leakage in mitochondria;thus,produce a higher amount of reactive oxygen species (ROS). Antioxidant inhibits oxidative damage in a target molecule. Grapes contain a lot of antioxidants, such as polyphenols and anthocyanins. The purpose of this study was to examine the effect of ethanol extract of Balinese grapes (Vitis vinifera) on β-cells and muscle damage in overtraining-induced rats. This study was a completely randomized experimental study using a posttest only control group design. Samples were 36 male albino rats (Rattus norvergicus), aged 2.5-3 months, divided randomly into two groups. The control group (P0), 18 rats, were given overtraining and placebo of 2 ml distilled water; the treatment group (P1) 18 rats, were given overtraining and 25g/kg.BW ethanol extract of Balinese grapes. The result showed the average amount of pancreatic β-cells in the P0 group was higher than the P1 group (p< 0.001). In addition, the average level of creatine kinase was also higher in the P1 group than those of the P0 group (p<0,001). It can be concluded that the administration of ethanolic extract of Balinese grapes mitigates the damage on pancreatic β-cells and muscle cells induced with overtraining.
