Abstract

High intensity exercise (HI) induces oxidative stress. Running for 2400 meters is one of the simple exercise form which can be counted as HI. During HI, increase of oxygen consumption happened in muscle cell resulted in an increase of oxidant level. The aim of the study was to measure the effects of Indonesian green tea supplementation, before and after HI to the level of lipid peroxidation (TBARSMalonyldialdehyde (MDA)). The subjects of study were 42 male students from Faculty of Medicine, Universitas Padjadjaran (FMUP) and Faculty of Physical Exercise, Indonesia University of Education (FPOK-UPI). The subjects were divided into trained and untrained subject. Subjects were divided into groups using Astrand Harvard step test. The purpose of this test was measuring the level of VO2 max. Blood samples for MDA level were collected at 3 hours before and after high intensity aerobic  exercise. Oxidative stress was induced by 2400 meters run. The result showed that green tea supplement-ation was effective to decrease MDA plasma level especially after exercise. Green tea could serve as natural antioxidant resources, which potentially can be used as potential sport supplemental drink.Key words: Aerobic exercise, Malonyldialdehyde (MDA), pre and post exerciseStudi Pra dan Pasca Pemberian Suplemen Ekstrak Teh Hijauterhadap Kadar MDAHigh intensity exercise (HI) dapat menginduksi stres oksidatif. Berlari sejauh 2400 meter merupakan salah satu bentuk latihan sederhana yang dapat digolongkan sebagai HI. Selama HI, konsumsi oksigen pada sel otot meningkat yang dapat menyebabkan peningkatan kadar oksidan. Penelitian ini bertujuanuntuk mengukur efek dari suplemen teh hijau Indonesia sebelum dan setelah HI terhadap kadar lipid peroksida (TBARS-Malonyldialdehyde (MDA)). Subjek penelitian ini adalah 42 mahasiswa laki-laki dari Fakultas Kedokteran Universitas Padjadjaran dan Fakultas Pendidikan Olahraga dan Kesehatan,Universitas Pendidikan Indonesia. Subjek penelitian dibagi ke dalam kelompok terlatih dan tidak terlatih dengan menggunakan cara pengujian Astrand Harvard. Tes ini bertujuan untuk mengukur kadar VO2 max. Sampel darah untuk kadar MDA diambil 3 jam sebelum dan setelah high intensity aerobic exercise. Stres oksidatif diinduksi dengan berlari sejauh 2400 meter. Hasil penelitian menunjukkan bahwa suplemen teh hijau efektif dalam menurunkan kadar MDA dalam plasma khususnya setelah berlari. Teh hijau dapat menjadi sumber antioksidan alami yang berpotensi sebagai minuman suplemen olahraga.Kata kunci: Aerobic exercise, Malonyldialdehyde (MDA), sebelum dan setelah berlari

Abstract

Aerobic physical activity is known to induce skeletal muscle adaptation. Some genes, including MyoD, are known to have a major role in the process of muscle adaptation. Several studies has stated that the expression of MyoD increases during the aerobic activity process; however, the studies were carried out in a single bout. Studies on the effects of acute phase exercise (<2 weeks) are still rare, especially those regarding the difference effect of acute training phase at different times. This study was performed in 2018 at the Central Laboratory and Animal Physiology Laboratory, Faculty of Medicine, Universitas Padjadjaran. The aim of this study was to determine the different effects of MyoD gene expression in a two-week acute aerobic physical activity. The effects were measured in the skeletal muscle in day 3, 7, and 14 of the activity. Twenty four male rats (rattus novergicus) were divided into 4 groups. Those rats in the treatment groups were run on an animal treadmill (P1=3 days, P2=7 days, and P3=14 days). On the last day of ran, rats were sacrificed and the soleus and gastrocnemius muscles were dissected. The expression of the MyoD gene in both muscles was then amplified using reversed PCR and detected by agarose gel electrophoresis. This study showed that there were differencesin MyoD gene expression levels in both muscles and that the level increased with days of treatment, although it was statistically insignificant. This study concludes that the MyoD gene expression level is not significantly affected by an acute aerobic physical activity in certain periods (3, 7 and 14 days).Key words: Acute, Aerobic physical activity, gastrocnemius, MyoD, rats, soleus Perbedaan Pengaruh Aktivitas Aerobik Akut Intensitas Sedang terhadap Ekspresi Gen MyoD pada Otot Rangka Tikus Galur WistarAktivitas fisik aerobik diketahui menginduksi adaptasi otot rangka. Beberapa gen diketahui memiliki peran utama dalam proses adaptasi otot termasuk MyoD. Dalam beberapa penelitian diketahui bahwa ekspresi MyoD meningkat selama proses aktivitas aerobik, tetapi penelitian dilakukan dalam pertarungan tunggal. Penelitian tentang efek latihan fase akut (<2 minggu) masih belum banyak dilakukan, terutama mengenai perbedaan efek fase latihan akut pada periode tertentu (3, 7, dan 14 hari). Penelitian ini dilakukan pada tahun 2018 di Laboratorium Sentral dan Laboratorium Fisiologi Hewan Fakultas Kedokteran Universitas Padjadjaran. Tujuan penelitian ini adalah mengetahui perbedaan efek ekspresi gen MyoD dalam aktivitas fisik aerobik akut pada berbagai hari (3, 7, 14 hari) pada otot rangka. 24 tikus jantan Rattus novergicus dibagi menjadi 4 kelompok. Kelompok perlakuan dijalankan dengan treadmill hewan (P1=3 hari, P2=7 hari, dan P3=14 hari). Pada hari terakhir, tikus dikorbankan dengan soleus dan otot gastroknemius dibedah. Ekspresi gen MyoD pada kedua otot kemudian diamplifikasi menggunakan PCR terbalik dan dideteksi dengan elektroforesis gel agarosa. Penelitian ini menunjukkan perbedaan level ekspresi gen MyoD pada kedua otot, hasil ini menunjukkan bahwa ada peningkatan level perawatan per hari, walaupun secara statistik tidak signifikan. Penelitian ini menyimpulkan bahwa pada otot gastrocnemius tidak terlihat efek yang berbeda dari durasi aktivitas fisik aerobik terhadap tingkat ekspresi gen MyoD, hanya otot soleus menunjukkan perubahan dan peningkatan ekspresi konsisten terlihat pada hari ke 14 (3, 7, dan 14 hari).Kata kunci: Aktivitas fisik aerobik, akut, gastroknemius, MyoD, soleus, tikus

Abstract

Fitness becomes an important benchmark for public health in every age group. Activities with poor adverse fitness are at risk of injury. Good fitness is gained from a balance in everyday physical activity with conditions that are free of metabolic syndrome. This study investigated the relationship of physical activity, energy balance, and metabolic syndrome to the fitness of 166 professors of Universitas Padjadjaran who underwent a 1-month fitness checkup at BAPELKES Bandung in the period of 2017. Based on the regression analysis, the results showed a significant relationship between physical activity, energy balance, and metabolic syndrome on the physical fitness of the elderly group (F = 25.9290> F table = 2.427) with normal data distribution (Kolmogorov-Smirnov test). The results also showed a moderate relationship (R = 0.569 and R2 = 0.324). It also partially showed significant influence between physical activity and fitness (t = 8.541> t table = 1.975), significant influence between energy balance and physical fitness (t = 2.248> t table = 1.975), and no significant partial influence between metabolic syndrome and physical fitness of the elderly group (t = -0.029 <t table = 1.975). Hence, there is a relationship between physical activity, energy balance, metabolic syndrome and physical fitness  of Professors in Universitas Padjadjaran.Key words: Energy balance, metabolic syndrome, physical fitness  Hubungan Aktivitas Fisik, Keseimbangan Energi, dan Sindrom Metabolik terhadap Kebugaran Fisik pada Kelompok Usia LanjutKebugaran menjadi tolak ukur penting bagi kesehatan masyarakat pada setiap kelompok usia. Aktivitas dengan kebugaran usia lanjut yang kurang baik berisiko terjadi cedera. Kebugaran yang baik diperoleh dari keseimbangan dalam aktivitas fisik sehari-hari dengan kondisi bebas dari sindrom metabolik. Penelitian ini mempelajari hubungan aktivitas fisik, keseimbangan energi, dan sindrom metabolik dengan kebugaran 166 orang Guru Besar di Universitas Padjadjaran yang dilakukan pemeriksaan kebugaran selama 1 bulan di BAPELKES Bandung periode tahun 2017. Berdasar atas analisis regresi hasil penelitian menunjukkan hubungan yang signifikan antara aktivitas fisik, keseimbangan energi, dan sindrom metabolik dengan kebugaran fisik kelompok usia lanjut (F=25,9290 > Ftable=2,427) dengan distribusi data normal (tes Kolmogorov-Smirnov). Hasil juga menunjukkan hubungan yang moderat (R= 0,569 dan R2= 0,324). Secara parsial juga menunjukkan pengaruh yang signfikan aktivitas fisik dengan kebugaran (t= 8.541> t tabel= 1.975), pengaruh signifikan keseimbangan energi dengan kebugaran fisik (t= 2,248> t tabel= 1,975), dan tidak ada pengaruh signifikan secara parsial terhadap sindrom metabolik dengan kebugaran fisik kelompok usia lanjut (t= -0,029 < t tabel= 1,975). Berdasar atas hasil penelitian dapat disimpulkan hubungan = aktivitas fisik, keseimbangan energi, dan sindrom metabolik dengan kebugaran fisik guru besar Universitas Padjadjaran.Kata kunci: Kebugaran, keseimbangan energi, sindrom metabolik

Abstract

Seringkali kejadian memar pada manusia maupun hewan menjadi bahan kajian yang menarik untuk diteliti sehingga banyak peneliti yang mencari dan melakukan formulasi penanganan/pengobatan untuk kesembuhan memar. Ragam efek dari pengaplikasian suatu zat/obat baru pada manusia dapat diketahui dengan mempelajari efek kumulatif dari dosis pada tahap praklinik. Informasi tersebut diperoleh dari melakukan eksperimen laboratorium secara in vivo dengan menggunakan hewan coba sebagai model yang dirancang pada serangkaian uji praklinik, seperti menggunakan tikus (Rattus norvegicus). Sejauh ini pengembangan mengenai pemanfaatan tikus sebagai hewan model memar masih sangat potensial untuk dipelajari lebih lanjut. Kajian literatur ini membahas mengenai penggunaan hewan sebagai model memar, tren tikus sebagai hewan model memar, prinsip etik dan animal welfare hewan model, penjelasan dan klasifikasi memar, metode pembuatan memar, dan aspek pengembangan hewan model masa depan sebagai subjek penelitian.

Abstract

Exercise-induced hearth muscle adaptation is important for physiological process after exercise. This adaptation will ensure basal mitochondrial homeostasis and as a part of the mitochondria quality control. This process is reflected by equal level of biogenesis stimulation and as well as the selective degradation of old and undesirable mitochondria through fusion or fission cycle and Mitophagy. There is limited information about genetic regulation stimulated by training in cardiomyocytes. We believe there is a specific myokines or protein release in the serum and initiate cardiac muscle adaptation process. In the present study, twelve male wistar rats were appointed to two group: sedentary control and aerobic-intensity (AE, 15m/minute). Rats were trained for running with specific protocol as follows: 30 minutes/day with a 5 times/week interval for 8 weeks. On the last day, serum form control and exercise groups were taken via retro-orbital sinus. Then, 3.105  H9C2 cells (Rat cardiomyocytes cell line)  were cultured and incubated by this serum for 24 hours. After treatment, cell were extracted using trisure for RNA purification and continue with reverse transcriptase PCR. Our data showed that expression of the Pgc-1α, Mfn1, Mfn2, Opa1, Drp1, Pink, and Parkin genes were altered and modulated. Specifically, Mfn1, Mfn2, and Opa1 gene expression levels significantly increased. Interestingly, we did not find significant modulation for  Pgc-1α, Drp1, Pink, and Parkin. Taken together, serum of exercise rats might be contained with myokines or specific protein which was released during training and it altered mitochondrial genes expression in cardiomyocytes culture cell. We believe that myokines release in the serum had a contribution in cardiacmyocyte adaptation.

Abstract

The prevention of liver disease could be conducted through preventive strategies, such as antioxidants and exercise. Moringa leaf extract has substances that could act as antioxidant, while exercise is also known protecting the liver from disease by changing hepatic metabolism and autophagy. This study aimed to investigate the effect of moringa leaf extract and moderate intensity exercise on histopathological appearance and autophagy gene expression of wistar rat liver. The method used in this study was animal experiment using 24 male wistar rats divided into 4 groups, including control group, moringa group, exercise group, and moringa + exercise group. Moringa leaf extract was given in low dose (5.7 mg/kgW) per oral, 5 times a week, for 4 weeks. Meanwhile, the 20 m/minute treadmill exercise was given for 30 minutes per day, 5 times a week, for 4 weeks. Results showed a significant change on histopathological scoring in exercise group (p=0.011) compared to control group. Increased autophagy gene expression was found in moringa + exercise group compared to control group (LC3 0.90 fold; p62 0.87 fold). In summary, this study presented that moderate intensity exercise induced changes on histopathological appearance of wistar rat liver that might be associated with physiological inflammation. Moringa, with its antioxidant properties, combined with increased autophagy might improve histopathological changes in moringa + exercise group.

Abstract

“Bihormonal hypothesis” is disturbances of both insulin and glucagon in diabetes mellitus. It resulted in a high blood glucose level. Interestingly, as one of Diabetes Mellitus’s risk factors and metabolic disorders, obesity may also play role in altering homeostasis and regulation of glucose metabolism in blood and its utilization in tissues. Unfortunately, there is limited information about the alteration of glucagon levels in various degrees of obesity. This research objective is to learn the plasma glucagon levels alteration in pre-obese, obese, and very obese persons in Jatinangor in 2015. We had observed 31 obese female subjects in one village. This study was conducted using descriptive quantitative with cross-sectional design. Blood vein samples from the left arm were collected, stored, and transferred to Dr.Hasan Sadikin General Hospital. Glucagon plasma was measured by the ELISA method. We discovered an interesting pattern that showed a correlation between glucose level and the glucagon level in a very obese group. We observed average glucose level is declined and linearly associate with the glucagon level from pre obese to obese and to very obese group. The average level of glucagon in the pre-obese group is 158.62 pg/mL, the obese group is 149.99 pg/mL, and the very obese group is 111.98 pg/mL.

Abstract

Exercise is an activity that has many health benefits, including bone, heart, and muscle health. One of the proteins found to be increased after exercise was FGF23. FGF23 is one of the bone mineral regulators which previously served as an indicator for chronic kidney disease severity and cardiovascular morbidity. However, some studies have found no increase in FGF23 after exercise. The purpose of this study is to elucidate the effect of exercise on serum FGF23. This study is a literature review with a qualitative approach comparing previous studies related to exercise and FGF23. From 13 selected kinds of literature, serum FGF23 elevation was found on exercise for one day to three weeks and did not change after six weeks to 12 months of exercise. The conclusions of the study are serum FGF23 was found to be increased after one day to three weeks of exercise and serum FGF23 in human studies found to be increased after long-duration maximal-intensity exercise.

Abstract

Pencak silat refers to performance sport requiring good physical condition. This study objective was to investigate the correlation between the physical activities and the body composition (fat percentage) to the cardiopulmonary endurance in female and male player-categorized athletes of pencak silat at Pelatda PON XVII Jawa Barat. The subjects of the study consisted of 20 pelatda athletes. The athletes conducted test of cardiopulmonary endurance using Astrand’s bench, daily intake was measured by food records 7x24 hours, physical activity was measured by activity records 7x24 hours, body composition was measured by skinfold calliper. The data was analyzed using Pearson’s correlation test. The conclusion, there were a close positive correlation of the physical activity to the cardiopulmonary endurance of all subjects, there was also a close negative correlation of the body composition (fat percentage) to the cardiopulmonary endurance of male athletes, while a weak positive correlation of female athletes.

Abstract

Under specific circumstances such as during aerobic predominant exercise, the production of free radicals increases and creates free radicals-endogen antioxidants imbalance. It causes oxidative stress. The level of oxidative stress can be recognized by measuring the level of plasma malondialdehyde (MDA). But the effect of physical activity and the effect of exogenous antioxidants supplementation is still not clearly known in the literature. Based on the background above, as a preliminary study, the writers conduct a study to investigate the plasma MDA level on mice which is given and not given a combination of Brastagi’s oranges and carrots juice before physical activity using mice’s treadmill for 10,20, and 30 minutes. The research method used in this study is an experimental laboratory study. As objects of this study are 24 mice(mus musculus), white-colored, male, weighting 25-30 grams, which is randomly chosen. The objects are divided into 2 groups, Group A: 12 mice (given a combination of Brastagi’s oranges and carrots juice before physical activity using mice’s treadmill) and group B: 12 mice (not given a combination of Brastagi’s orangesand carrots juice before physical activity using mice’s treadmill). Group A is divided into 3 subgroups: A1 (doing treadmill for 10 minutes), A2 (doing treadmill for 20 minutes), and A3 (doing treadmill for 30 minutes). The same procedure is employed for group B. Plasma MDA level measured after doing a physical activity using mice treadmill. The homogeneity of the result then was tested using Levene’s test and the normality of the result was tested using the Kolmogorov-Smirnov test (p>0.05). Further, the data were analyzed using an independent t-test (p≤0.05), one-way ANOVA (p≤0.05) then Duncan’s test were used. The analyzed data indicated a combination of Brastagi’s oranges and carrots juice supplementation given to mice before physical activity, for 10,20, and 30 minutes caused lower plasma MDA levels than mice not given a combination of Brastagi’s oranges and carrots juice before physical activity. The differences are 38,44% (0,5277 vs 0,8571) (A1-B1); 37,96% (0,5866 vs 0,9455) (A2-B2); dan77,79% (0,8438 vs 1,6161) (A3-B3).The conclusion of the study suggested that a combination of Brastagi’s oranges and carrots juice supplementation given to mice before physical activity for 10,20, and 30 minutes caused lower plasma MDA levels than mice not given a combination of Brastagi’s oranges and carrots juice before physical activity.
