Abstract

ABSTRACTCitrus aurantifolia is known contains flavonoid compound and high Vitamin C. One of theeffects of flavonoid and Vitamin C are antioxidant. This research aims to knowing the antioxidantactivity of citrus aurantifolia etanolic extract by using the DPPH (1, 1-difenil-2-pikrilhidrazil) methodand to knowing the active compound that containing whithin.The research was done by making level series of citrus aurantifolia etanolic extract, they were10, 20, 40 and 80 μg/ml. As a standard of comparison was used vitamin C with concentrations 1, 2, 4and 8 μg/ml. As a blank was used DPPH 0,1 mM. The antioxidant activity test was done by the DPPHmethod. The achieved data was counted to know it’s antioxidant activity. The statistical analysis wasused T-Test. To know IC50 (Inhibition Concentration)50 was used the probit analysis and to know it’sactive compound content was done an identification with TLC (Thin Layer Chromatography).The result of the research shows that etanolic extract of citrus aurantifolia has antioxidantactivity IC50 about 54,458 μg/ml and 4,768 μg/ml for vitamin C. The statistical test result of antioxidantactivity shows that there is no any significant difference. The TLC result shows that compoundcontained in etanolic extract of citrus aurantifolia are flavonoid and Vitamin C.Keywords: Citrus aurantifolia extract, DPPH method, antioxidant and IC50.

Abstract

ABSTRACT Free radicals can lead to various dangerous diseases, such as cancer. Active compounds from medicinal plants such as flavonoids and alkaloids have been shown the antioxidant effects and may serve to prevent the negative effects of free radicals. Two of these compounds have been identified in leaf extract (Carica pubescens) and can be separated from other compounds with fractionation method. The purpose of this study was to determine the antioxidant activity of n-hexane fraction of karika leaves ethanol extract (HFKLEE) with DPPH method and identify its alkaloids and flavonoids compounds. Antioxidant activity of HFKLEE series concentration (50; 100; 150; 200; 250 and 300) ppm was performed by the DPPH method. Vitamin C series concentration (0.5; 1.0; 1.5; 2.0 and 2.5) ppm was used as a standard solution. Identification of alkaloids and flavonoids compound in HFKLEE determined by the thin layer chromatography (TLC) method. The results showed that HFKLEE has antioxidant activity with IC50 values of 157.134 ppm and Vitamin C at 2.875 ppm. TLC results indicate the presence of alkaloids and flavonoids compounds in HFKLEE. Abstract

ABSTRACT The chemicals contained in an extract must be maintained its quality. The aim of this study is to prove both that the ethanol extract of kecapi (Sandoricum koetjape, Merr.) and keluwih (Artocarpus communis) leaves meet the non-specific parameters on the standardization of the extract. The ethanol extract of kecapi and keluwih leaves were extracted using the ultrasonic method. The extract was tested with non-specific parameters including water content, total ash content and metal contamination level. The test results are compared with standard parameter references. Ethanol extract of kecapi leaf have water content of 27.75% ± 1.708, total ash content of 4.856% ± 0.040 and levels of As metal contamination < 0.030 mg/Kg, Hg < 0.002 mg/Kg, Pb < 0.400 mg/Kg, while ethanol extract of keluwih leaf have water content of 23.206 ± 0.091%, total ash content of 9,678 ± 0.306% and levels of heavy metal contamination of As < 0.03 mg/Kg, Hg < 0.006 mg/Kg, Pb < 0.02 mg/Kg. The both extract meet the standard parameters of water content, total ash content and levels heavy metal contamination. Abstract

ABSTRACT The content of active compounds and the quality of extracts cannot be guaranteed to always be in a constant amount, so it is necessary to standardize. This study aims to prove the ethanol extract of jamblang leaves (Syzygium cumini (l.) Skeels) fulfill the standardization of non-specific and specific parameters as a natural medicine. The ethanol extract of jamblang leaves was extracted by extracting using ultrasonic methods The extract was tested for non-specific parameters including: determination of water content, total ash content, acid insoluble ash content, drying losses and heavy metal contamination. Whereas the specific parameter tests include: identity, organoleptic extract, compounds dissolved in water and ethanol, and chemical content. Test results were analyzed descriptively and compared to standard parameters. Abstract

ABSTRACTImmunostimulatory agent has an important role in the treatment of infectious disease caused by pathogen. Roselle calyx (Hibiscus sabdariffa L.) used by community as a health drink to enhance body’s immune system. The aims of this research was to know immunostimulatory activity of ethyl acetate fraction of ethanolic extract of roselle calyx on lymphocyte cell proliferation swiss strain mice and to identified the chemical compounds of the fraction. Roselle calyx was extracted by maceration method using ethanol 96% as a solvent. The ethanolic extract was partitioned gradually by n-hexan and ethyl acetate. Series of ethyl acetate fraction were made to concentration 10, 20, 50, 100, 200, 400 μg/mL and positive control PHA 10 μg/mL for tested to lymphocyte cell culture. Immunostimulatory activity test was done according to MTT Assay method. Lymphocyte proliferation activity was analyzed statistically to the value of Optical Density (OD) result of the Enzyme Linked Immunosorbent Assay (ELISA) reader using Kruskal Wallis Test continued by Mann Whitney Test (p<0,05). Identification of the contents phenolic compounds and flavonoids performed by Thin Layer Chromatography (TLC). The result of the research showed that ethyl acetate fraction had lymphocyte cell proliferation activity at the concentration of 50, 100, 200, and 400 μg/mL. Abstract

ABSTRAK Teh kombucha diperoleh dari hasil fermentasi larutan teh, gula dan koloni kombucha.  Fermentasi teh hitam jamur kombucha (FTHJK) secara empiris, terbukti dapat meningkatkan sistem kekebalan tubuh. Penelitian ini bertujuan untuk mengetahui aktivitas imunomodulator hasil FTHJK terhadap proliferasi sel limfosit mencit galur Balb/C secara in vitro. Kombucha difermentasi selama 5, 10, dan 15 hari dengan menggunakan seduhan teh hitam yang telah diberi gula. Hasil fermentasi teh hitam jamur kombucha (FTHJK) dibuat beberapa seri konsentrasi yakni 6,25; 12,5; 25; 50 dan 100 µg/ml dan sebagai kontrol positif Phytohemagglutinin (PHA) 10 µg/ml diuji secara in vitro terhadap proliferasi sel limfosit mencit galur Balb/C jantan usia 2 bulan dengan menggunakan metode MTT Assay. Data absorbansi berupa nilai Optical Density (OD) dianalisis secara statistik dengan Friedman Test dilanjutkan Mann Whitney Test. Hasil penelitian menunjukkan bahwa FTHJK memiliki aktivitas imunostimulator terhadap proliferasi sel limfosit pada konsentrasi 50 dan 100µg/mL fermentasi hari ke-5, dan konsentrasi 100µg/mL fermentasi hari ke-10 dan 15. Aktivitas imunostimulator tertinggi terjadi pada konsentrasi 100µg/mL dengan lama fermentasi hari ke-15. kata kunci: Teh hitam jamur kombucha, imunomodulator, MTT Assay,Optical Density (OD).

Abstract

Beberapa metode analisis telah dikembangkan untuk menentukan kadar natrium diklofenak, diantaranya dengan metode spektrofotometri UV dan KCKT (Kromatografi Cair Kinerja Tinggi). Metode spektrofotometri UV dan KCKT masing-masing mempunyai kelebihan dan kekurangan. Oleh karena itu, penelitian ini bertujuan untuk mengetahui perbandingan kadar natrium diklofenak secara spektrofotometri UV dan KCKT dalam hal ketepatan, ketelitian dan sensitivitas. Penetapan kadar natrium diklofenak dianalisis dengan metode spektrofotometri UV dengan melakukan penentuan panjang gelombang maksimal, penentuan operating time, pembuatan kurva baku, pengukuran serapan sampel, perhitungan kadar natrium diklofenak dalam sampel. Sedangkan metode KCKT dengan melakukan optimasi instrumen dan optimasi fase gerak, identifikasi natrium diklofenak dalam sampel, pembuatan kurva baku, pengamatan kromatogram sampel, perhitungan kadar natrium diklofenak dalam sampel. Pelarut yang digunakan adalah aquabidestilata dan fase gerak yang digunakan adalah campuran asetonitril  dan buffer fosfat 0,01 M pH 3,5. Sedangkan fase diam yang digunakan adalah Oktadesil Silikat (ODS) C18 (4,6x150mm). Data kadar yang diperoleh dari masing-masing metode dibandingkan ketepatan, ketelitian dan sensitivitas dari dua metode tersebut. Hasil penelitian menunjukkan bahwa kadar rata-rata natrium diklofenak secara Spektrofotometri UV adalah 17,9 µg/ml. Sedangkan secara KCKT adalah 17,3 µg/ml. Kadar rata-rata natrium diklofenak dalam sampel secara spektrofotometri UV dan KCKT memenuhi persyaratan Farmakope Indonesia yaitu memiliki nilai rekoveri yang masih dalam range 90,0 % -110 % dan memiliki nilai CV < 5%. Akan tetapi metode KCKT memiliki sensitivitas yang lebih tinggi dibandingkan metode Spektrofotometri UV.   Kata kunci:  Spektrofotometri UV, Kromatografi Cair Kinerja Tinggi, Natrium  Diklofenak

Abstract

ABSTRACTAntibodies play a role in maintaining the body’s immune system from various microorganism. When the body experiences an imbalance, the body needs a compound that can increase antibodies. Flavonoids, polyphenols and alkaloids can increase antibodies. Flavonoids, polyphenols and alkaloids can increase antibodies. Ethanol extract of kedondong leaves contains compounds of flavonoid, alkaloids, and polyphenols. The purpose of this study was to determine the immunomodulatory activity of ethanol extract in IgM and IgG antibodies in balb /c mice induced by hepatitis B vaccine. Ethanol extract of kedondong leaves was macerated with 96% ethanol. The doses of the concentration of ethanol extract of kedondong leaves used were 5, 10 and 20% p.o for 46 days and vaccination was carried out on the 7th, 28th and 43rd days. Positive control was used with levamisole at a dose of 0.45mg / 20gBB by p.o. Analysis of Optical Density (OD) IgM and IgG on days 14, 35 and 46 using Friedman and Mann Whitney statistical tests. The results showed that the ethanol extract of kedondong leaves had activity of increasing IgM and IgG antibodies at concentrations of 5, 10 and 20% on days 14, 35 and 46 and contained flavonoids, alkaloids and polyphenols.Keywords: Hepatitis B Vaccine, IgM, IgG, Optical Density (OD)

Abstract

ABSTRACT Recently, some research showed that there is an immunomodulator effect in the fermentation of Rosella kombucha mushroom  tea and it can be caused by flavonoid and organic acid. The purpose of this research was to show the activity of immunomodulator Rosella kombucha mushroom  tea to against the proliferation of lymphocyte cells of Balb/C mice in vitro and to show the dose and length of fermentation that induces highest immunomodulator activity.  Kombucha mushroom was fermented for 5 days, 10 days and also 15 days with boiled Rosella tea, water and sugar. The yield of fermentation in the concentration of 6,25; 12,5; 25; 50 and 100 µg/mL were tested on lymphocyte cells proliferation by the MTT Assay. The OD (Optical Density) data obtained was  statistically analyzed by nonparametric Friedman test and continuoused by Mann-Whitney test. The result of this research showed that the fermentation of Rosella kombucha mushroom tea  has an immunostimulatory activity on lymphocyte cells proliferation with the concentration 6,25; 12,5 and 25 µg/mL in the 10 days and the highest immunostimulatory activity  showed by the concentration 25 µg/mL and 10 days of fermentation time. Abstract

INTISARI Teh hitam dan kombucha adalah minuman kesehatan yang diketahui mampu meningkatkan sistem kekebalan tubuh. Penelitian ini bertujuan untuk membuktikan aktivitas imunomodulator fermentasi teh hitam jamur kombucha (FTHJK) terhadap fagositosis makrofag mencit galur Balb/C secara in vitro. Fermentasi kombucha dilakukan selama 5, 10 dan 15 hari menggunakan media seduhan teh hitam yang telah ditambahkan gula. Hasil FTHJK dibuat beberapa seri konsentrasi yaitu 6,25; 12,5; 25; 50, 100 µg/mL dan sebagai kontrol positif digunakan lipopolisakarida (LPS) 10 µg/mL untuk diujikan aktivitas fagositosis makrofag secara in vitro pada mencit jantan galur Balb/C usia 2 bulan. Data yang diperoleh berupa nilai Indeks Fagositosis  (IF) (jumlah lateks yang dimakan oleh 100 sel makrofag) dan Kapasitas Fagositosis (jumlah sel makrofag yang memakan lateks). Hasil IF dan kapasitas dianalisis secara statistik menggunakan  SPSS  16  for windows   metode Friedman test, dilanjutkan uji Mann Withney. Hasil penelitian menunjukkan, FTHJK memiliki aktivitas imunomodulator terhadap peningkatan fagositosis makrofag pada konsentrasi 6,25; 12,5; 25; 50 dan 100 µg/mL dengan lama fermentasi 5, 10 dan 15 hari. Aktivitas imunostimulator tertinggi ditunjukkan pada konsentrasi 100 µg/mL  dengan  lama fermentasi 10 hari berdasarkan nilai IF.  Kata kunci:    Imunomodulator,  IF  (Indeks Fagositosis),  Kapasitas Fagositosis, Fermentasi teh hitam jamur kombucha. This study aims to not only determine the  fermented black tea kombucha as an  immunomodulatory againts macrophages cell phagocytic of mice strains Balb/C in vitro, but also to determine concentration with duration fermentation that produces the highest activity Fermentation kombucha was carried out  for 5, 10 and 15 days with steeping black tea has been given sugar. Fermented black tea kombucha was made to any series concentrations of 6,25; 12,5; 25; 50 and 100 µg/mL, as a positive control used lipopolysaccharide (LPS) 10 mg/mL there were tested in vitro on activity phagocytic macrophage cell of mice strains Balb/C.   Value of Phagocytic Index (phagocytic of latex in 100 macrophage)   and phagocytic capacity (amount of macrophage that phagocytic latex)  were analyzed  using SPSS 16  for windows with methode Friedman test followed by Mann Withney test.  The results showed that fermented black tea kombucha has the highest as immunostimulatory activity on macrophage phagocytic at the  concentration of 100  µg/mL  and 10 days duration fermentation.  Keywords:  Immunomodulatory, Phagocytic  Index, Phagocytic Capacity, Fermented  black  tea kombucha.
