Abstract

Androgen receptor (AR) is the member of steroid hormone receptor involved in the progression of prostate cancer growth due to receptor over-activation. On the other hand, mangosteen (Garcinia mangostana L.) as a medicinal plant contains xanthone-derived compounds which were known to have cytotoxic activity towards any types of human cancer cells. This research aims to determine xanthone-derived isolates potency from mangosteen as AR antagonists. The study was carried out through molecular docking assay utilizing AutoDock 4.2.6 using androgen receptor obtained from PDB ID 2AM9, testosterone as native ligand, and bicalutamide, flutamide, and nilutamide as reference. The results indicated that three isolates (1,3,7-trihydroxy-2,8-di-(3-methylbut-2-enyl)xanthone, mangostinone, and trapezifolixanthone) have the highest potency to be AR antagonist seen from the lower bond-free energy value than all of reference ligand. The lowest bond-free energy was provided by mangostinone with a ΔG value of -10.05 kcal/mol. However, the highest difference of residual amino acids interaction with testosterone and similar interaction with bicalutamide was provided by 1,3,7-trihydroxy-2,8-di-(3-methylbut-2-enyl)xanthone, with five different amino acids with testosterone and nine similar amino acids with bicalutamide, respectively. Interestingly, 1,3,7-trihydroxy-2,8-di-(3-methylbut-2-enyl)xanthone has similar hydrogen bond with the key residue amino acids of AR (705-Asn and 711-Gln) which indicates probably partial agonist activity while mangostinone has the highest amount of hydrogen bond in the absence of hydrogen bond towards key residual amino acids of AR. The results concluded that three specific derived-xanthone compounds were predicted to have activity as AR antagonists.Keywords: Prostate cancer, Androgen receptor, Mangosteen, Xanthone, Molecular docking.

Abstract

The aim of this study is to examine the possible use of ion-exchange resin on Dowex AG 50x8 in separating Y-90 from Sr-90 which then could be applied in the basic of manufacture of producing Sr-90/Y-90 generator for the production of radiopharmaceutical cancer therapy. The findings showed that a distributive coefficient of Sr-90 and Y-90 with HCl 6 N solvent showed that distributive coefficient of Sr-90 in cation-exchange resin on Dowex 50x8 was greater than Y-90. Y-90 in HCl 6 N solvent was more easily eluted, while Sr-90 was bound to resin.. In conclusion, resin on Dowex AG 50x8 with HCl 6 N elution solvent might be able to separate Y-90 from Sr-90 as the basis for Sr-90/Y-90 generator.

Abstract

Sekitar 80%, alternatif dalam terapi pemeliharaan kesehatan bergantung pada penggunaan obat tradisional. Obat tradisional berasal dari bahan dan teknik pengobatannya yang bersifat alami, sehingga efek sampingnya relatif rendah, memiliki kandungan senyawa yang beraneka ragam bersifat sinergis serta biaya pengobatannya lebih ekonomis. Tanaman genus Garcinia merupakan famili dari Clusiaceae memiliki 400 spesies, 77 spesies diantaranya tersebar di wilayah Indonesia dan tumbuh di daerah tropis. Tanaman ini memiliki potensi besar sebagai obat tradisional dalam  mengobati berbagai jenis kelainan patofisiologi. Pemanfaatan tanaman sebagai obat tradisional sebagian besar didasarkan pada pengalaman, namun informasi tentang bukti keamanan atau toksisitas tanaman obat kepada masyarakat masih terbatas sehingga perlu dilakukan pengkajian toksisitas genus Garcinia. Spesies tanaman dari genus Garcinia yang telah diuji toksisitasnya antara lain, Garcinia brasiliensis Mart., Garcinia xanthochymus Hook.f. ex T.Anderson, Garcinia cowa  Roxb., Garcinia pedunculata Roxb. ex Buch.-Ham., Garcinia husor, Garcinia kola Heckel, Garcinia benthami Pierra, Garcinia mangostana L., Garcinia indica (Thouars) Choisy, Garcinia hombroniana Pierre, Garcinia rubroechinata Kosterm., Garcinia achachairu Rusby, Garcinia atroviridis Griff. ex T. Abstract

Coronary Heart Disease (CHD) is a heart disorder caused by blockage of blood vessels. CHD can bedetected by Myocardial Perfusion Imaging (MPI). MPI is performed by injecting a radiopharmaceuticalinto the patient's body. 99mTc-sestamibi is a radiopharmaceutical that is commonly used in MPI.Sestamibi is available in the form of a multidose vial, but the cost of the examination will be expensiveif it is only used for one patient. Cost effectiveness can be increased by fractionating the sestamibi kitbefore labelled by 99mTc. 99mTc-sestamibi needs to be tested for quality control before it is administeredto the patients. One of the tests is the radiochemical purity test. The aim of this study was to determinethe radiochemical purity of the fractionated sestamibi kit labelled by 50 mCi 99mTc. 2 vials of sestamibikit were fractionated by adding 5 mL of 0.9% NaCl solution to each vial and divided into 10 new vials.Radiochemical purity was measured using the thin layer chromatography (TLC) method. The resultsof this study indicated that all samples had radiochemical purity of 100% up to 6 hours after labelling.

Abstract

Leukemia Limfoblastik Akut (LLA) merupakan penyakit hematologi heterogen yang ditandai dengan proliferasi sel limfoid imatur di sumsum tulang, darah tepi, dan organ lainnya karena adanya mutasi pada reseptor JAK. Beberapa tahun terakhir, strategi baru yang menargetkan protein terkait penyakit untuk degradasi seperti Proteolysis targeting chimerics (PROTAC) diteliti. Penelitian ini bertujuan untuk studi in silico aktivitas senyawa penghambat STAT 3 dan JAK 2 pada tanaman ranti (Solanum nigrum L.). Pengujian dilakukan menggunakan perangkat lunak meliputi, ChemOffice 2019, AutoDockTools 1.5.6., BIOVIA Discovery Studio 2021, dan Ligandscout.  Dari 17 senyawa tanaman ranti yang diuji, hanya 11 senyawa yang memenuhi aturan Lipinski rules of five dan prediksi ADMETOKS yang kemudian dilanjutkan dengan penambatan molekul. Hasilnya, senyawa N-Acetyl-L-tryptophan ethyl ester dinilai paling berpotensi sebagai antikanker, meskipun memiliki energi gibbs yang lebih rendah sebesar -6.38 kcal/mol apabila dibandingkan dengan ligan alaminya yakni -8.19 kcal/mol. Hasil menunjukkan tanaman ranti (Solanum nigrum L.) berpotensi sebagai kandidat pengobatan LLA, namun masih butuh penelitian lebih lanjut dengan perangkat lunak yang lebih lengkap.

Abstract

Kampung Batu Lonceng, Desa Suntenjaya, Kecamatan Lembang merupakan salah satu daerah usaha tani dan ternak yang ada di Indonesia. Penelitian ini dilakukan untuk mengetahui tingkat pengetahuan dan pemahaman masyarakat di Desa Suntenjaya, Kecamatan Lembang mengenai budidaya cacing dan digital marketing. Cacing tanah juga memiliki manfaat lain yaitu pada bidang medis. Cacing tanah dapat digunakan sebagai obat penurun panas, demam dan obat tipus. Penggunaan cacing tanah sebagai obat diproses dengan cara dikeringkan dan diekstrak dalam bentuk kapsul. Penelitian ini menggunakan random sampling survey terhadap 21 warga Desa Suntenjaya yang mengisi kuesioner secara luring di tempat. Kategori tingkat pengetahuan responden ditentukan berdasarkan pengkategorian Arikunto (2013). Hasilnya tingkat pengetahuan masyarakat di Desa Suntenjaya mengenai budidaya cacing dan digital marketing dikategorikan kurang baik dengan rata-rata skor ≤ 9%.

Abstract

Temulawak rhizome (Curcuma xanthorrhiza Roxb.) is a rhizome that comes from the Zingiberaceaetribe. Temulawak rhizome is commonly used as a traditional medicine in Indonesia as an antiinflammatory.The purpose of this study was to provide information on the potential of temulawakrhizome as a COX-2 inhibitor drug candidate and its toxicity to shrimp larvae (Artemia salina Leach.).The methods used are Lipinski Rule of Five prediction, PreADMET, molecular docking, pharmacophorescreening, and BSLT toxicity test. The results obtained show that the lowest Gibbs energy is producedby curcumin (-9.65 kcal/mol), has a pharmacophore hit value, meets the Lipinski rule of five, predictsa good pharmacophore profile, but curcumin has mutagenic properties and is classified as toxic afterbeing tested. with the BSLT method. So that it can be concluded that curcumin has the potential tobecome an anti-inflammatory drug, but further studies are needed and modifications to the molecularstructure of the compound can be carried out so that the tested compound can produce better activity.

Abstract

Abstraks-PhenylMercapturic Acid (s-PMA) merupakan salah satu biomarker spesifik yang diusulkan untuk pemantauan biologis terhadap paparan benzena dalam konsentrasi rendah. American Conference of Governmental Industrial Hygienists (ACGIH) juga mengharuskan perusahaan industri untuk mengevaluasi tingkat s-PMA pada pekerja yang bekerja terkait dengan benzena. ACGIH BEI (Biological Exposure Limit) menetapkan nilai normal s-PMA dalam urin maksimal 25 μg/g kreatinin (25 ppb) (ACGIH, 2012). Beberapa metode telah dikembangkan untuk mendapatkan kesesuaian prosedur dalam penetapan kadar s-PMA dalam urin. Metode yang digunakan untuk analisis s-PMA sampai saat ini adalah High Performance Liquid Chromatografi (HPLC) dengan detektor flouresens, HPLC dengan detektor DAD, HPLC-MS-MS, GC-MS, dan ELISA (Wang. Z, et al, 2013). Preparasi terhadap sampel dilakukan beberapa tahap, menggunakan teknik liquid-liquid extraction (LLE), solid-phase extraction (SPE) dan derivatisasi analit menjadi senyawa fluoresens terkonjugasi (Buratti M, 2001). Kata kunci: Biomarker, s-Phenyl MercapturicAcid (s-PMA), Benzena, HPLC Abstracts-PhenylMercapturic Acid (s-PMA) is one of the specific biomarkers purposed for biological monitoring of benzene exposure in low concentrations. The American Conference of Governmental Industrial Hygienists (ACGIH) also requires industrial companies to evaluate the level of s-PMA who workrs working with benzene. ACGIH BEI (Biological Exposure Limit) determines the normal value of s-PMA in urine to a maximum of 25 μg/g creatinine (25 ppb) (ACGIH, 2012). Several methods have been developed to obtain the suitability of procedures in the determination of s-PMA levels in the urine. The recently of analysis methods used for s-PMA are High Performance Liquid Chromatography (HPLC) with flouresens detector, HPLC with DAD detector, HPLC-MS-MS, GC-MS, and ELISA (Wang. Z, et al, 2013). Abstract

Mangosteen peel extract contains good antibacterial compounds and is thought to be better when microencapsulated is formed. This study aimed to determine the effects of formulation of mangosteen peel ethanol extract associated to maltodextrin on activities of Escherichia coli. It was conducted by disk diffusion method according to Kirby-Bauer test in Mueller Hinton Agar (MHA) using three different concentrations namely: 100, 75, and 50 (%). Each formulation was tested as many as 10 µL disk -1 consisted of each treatment. The employed maltodextrin was obtained by enzymatic processing of arenga starch. Nine formulations were tested by measuring their bacterial inhibition activity in a Mueller Hinton Agar (MHA) medium.  The results showed that the effects of formulation between mangosteen peel ethanol extract and maltodextrin had the significant effect (P<0.05) on the antibacterial activities of Escherichia coli. It had been shown the ability to actively inhibit the bacterial growth at medium levels.
