Abstract

The aim of the research is to analyse germination percentage, effect of coconut water addition in the media to number of last stage embryos and development of final stage D. anosmum Lindl. embryo. Research was conducted at Plant Structure and Development of Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences University of Udayana and UPT. Balai Benih Induk Tanaman Pangan dan Hortikultura Provinsi Bali from February 2015 to July 2015. Three type of media were employed, i.e. PDA, Organic, MS. Three different concentration of coconut was added to media, i.e. 0 ml/L 50 ml/L, 100 ml/L. Results showed that seed D. anosmum Lindl. on PDA, Organic and MS has not germinated yet on all media, only swollen cells observed. Seed dormancy and maturity of seed were the main factors affecting seed germination.  Addition of 100 ml / L coconut water in MS showed that media is capable to increase the formation of final stages embryos (> 32 cells). Embryo development on PDA and Organic largely showed cell stage embryos 1 to 8 cells, while MS media already showed embryo development stages of > 32 cells.

Abstract

Longtail tuna (Thunnus tonggol) is one of the oceanodromus neritic species and the migration pattern follows the water currents. Currently, this species has not been widely studied in Indonesian waters, so it is nesessary to study the identification of morphology and genetic diversity. The molecular approach employed in this study is DNA barcoding using mitochondrial D – loop locus. This study can explain the importance of species genetic information in stability and resilience. This study aims to determine the identification of morphology, phylogenetic and genetic diversity of longtail tuna at two locations in PPI Kedonganan, Bali dan PPP Muncar, Banyuwangi. The molecular analysis was done in several stages, i.e DNA extraction, Polymerase Chain Reaction, electrophoresis and sequencing. Based on the result of sequencing and analysis, 33 samples of longtail tuna was found. The result of phylogenetic tree reconstruction from two locations showed one clade with genetic distance value among longtail tuna species ranging from 0.000 – 0.042 for all close kinship samples. The haplotype diversity (Hd) value of longtail tuna was 0.9905 and nucleotide diversity (?) was 0.020. The high value of genetic diversity indicated that two longtail tuna populations have a high survival ability to adapt on environmental changes. Index fixation analysis (Fst) has a value of 0.0299, p - value > 0.05. The index fixation value indicated no significant population difference. The result of this study can be use as basic data in planning genetic conservation strategies with sustainable fisheries management efforts. Keywords: Tuna; Thunnus tonggol; Genetic Marker

Abstract

AbstractSeven Brassica napus cultivars were field tested to find out their adaptability andproductivity in tropical dry land environment. The aim of this research is to identifypotentially high oil yielding and agronomically stable Brassica napus genotypes as a newsource of biodiesel in Indonesia. Crop evaluation includes germination rate, plant height, number of leaves and stomata, flowering time, harvesting time, seed yield and oil content.Oil extraction using ’cold pressing’ technique were trialed. Results show that all genotypeshave good adaptability, therefore can be an alternative for biodiesel sources in Indonesia.Variation occurs on plant height, number of leaves, flowering time, seed set and oil content.‘‘Tanami’’, ‘‘Trigold’’ and ‘‘Trilogy’’ has similar performance, short plants, high pod set andearly flowering, while other cultivars have vigorous, tall plant habit but lower seed set. Some cultivars have an early flowering type while others has intermediate or late flowering.Pod set was excellence; however, seed set was low due to pest attack, namely Diamondbackmoth. Oil content of the seeds ranging from 2.6 to 11 %. In conclusion, B. napus areadaptable in dry land environment. Further study is needed to maximize seed productionand oil content.
