Abstract

Abstrak Gangguan metabolik merupakan gangguan peningkatan kadar asam urat (hiperurasemia) penyakit ini sudah sering dijumpai baik di negara Indonesia ataupun negara lainnya di benua Asia. Obat golongan xantin oksidase inhibitor seperti alopurinol dan febuxostat direkomendasikan sebagai lini pertama untuk pengobatan, tetapi perlu diperhatikan penggunaan xantin oksidase yang terus menerus dapat menyebabkan efek samping toksisitas pada gastrointestinal dan meningkatkan serangan akut gout pada awal terapi. Oleh karena itu, banyak penelitian yang melibatkan banyak tumbuhan herbal yang diharapkan memiliki aktivitas antihiperurisemia karena tanaman herbal memiliki resiko toksisitas dan efek samping yang lebih rendah. Berdasarkan penelitian yang telah dilakukan jahe merah (Zingiber officinale var. Amarum), daun salam (Syzygium polyanthum), Herba Suruhan (Peperomia pellucida (L.) Kunth), Meniran (Phyllanthus niruri Linn.) , kayu secang (Caesalpinia sappan L.) , Ekstrak Rebung (Schizostachyum brachycladum Kurz ), Pakis tangkur (Polypodium feei), Naga Putih (Hylocereus undatus), (Mimosa pudica L. ), Daun Sirsak (Annona muricata L.), Sparattosperma leucanthum , Mahkota Dewa (Phaleria macrocarpa (Scheff.) Boerl.), Kulit Buah Manggis (Garcinia mangostana L.), Dioscorea tokoro makino, dan chatupatika memiliki aktivitas antihiperurisemia. Oleh karena itu, pada review ini akan dibahas aktivitas antihiperurisemia pada lima belas tanaman tersebut. Hasil yang didapatkan dari beberapa artikel yaitu beberapa tanaman tersebut berkhasiat sebagai antihyperurisemia melalui uji daya hambat xantin oksidase dan menurunkan kadar asam urat pada hewan uji. Sehingga memiliki potensi yang dapat dikembangkan sebagai obat untuk hyperurisemia. Kata Kunci: Hiperurisemia, Xantin Oksidase, Tanaman Herbal AbstractMetabolic disorders that affect the increase in uric acid levels (hyperurasemia) are diseases that have often been found in both Indonesia and other countries in the Asian continent. Xanthine oxidase inhibitors such as allopurinol and febuxostat are recommended as a first line for treatment, but it should be noted that the continued use of xanthine oxidase can cause gastrointestinal toxicity side effects and increase acute attacks of gout at the beginning of therapy. Therefore, many people use medicinal plants as anti gout because they have relatively small side effects, are easy to obtain, and are relatively inexpensive compared to synthetic drugs. Based on the research, red ginger (Zingiber officinale var. Amarum), bay leaf (Syzygium polyanthum), Suruhan Herb (Peperomia pellucida (L.) Kunth), Meniran (Phyllanthus niruri Linn.) , secang wood (Caesalpinia sappan L.), Bamboo Shoot Extract ( Schizostachyum brachycladum Kurz), Pakis tangkur (Polypodium feei), White Dragon (Hylocereus undatus), (Mimosa pudica L.), Soursop leaf (Annona muricata L.), Sparattosperma leucanthum, Mahkota Dewa (Phaleria macrocarpa (Scheff.) Boerl.), Kulit Buah Manggis (Garcinia mangostana L.), Dioscorea tokoro makino, and chatupatika have antihyperuricemia activities. Therefore, this review will discuss the antihyperuricemia activity in these fifteen plants. The results obtained from several articles, namely some of these plants are efficacious as antihyperurisemia through the inhibitory test of xanthine oxidase and reduce uric acid levels in test animals. Abstract

Sintoc (Cinnamomum sintoc Bl.) is a plant which is used as medicine. This plant has been known to have an analgesic antiinflamatory activity, therefore it is predicted to have an antioxidant activity. An investigation on antioxidant activity of sintoc essential oils and ethanolic extract of its cortex using ascorbic acid as standard has been carried out. Essential oils and ethanol extract of sintoc cortex was tested using DPPH (1,1-diphenyl-2-pikril-hidrazil) by measuring absorbance using visible spectrophotometer at 518 nm. The methods of this research were distillation of essential oils and extraction of sintoc cortex, determination of the essential oil and extract concentrations required for 50% inhibition of DPPH radical scavenging effect (IC50) with ascorbic acid as the possitive control. The variation concentration  of essential oils are 15, 5, 1, 0.1, 0.5 ppm and 25, 20, 17, 15, 10 ppm for ethanolic extracts. The results showed that the essential oil showed antioxidant activity with IC50 value was 16.29 ppm (5 times lower than ascorbic acid) and then ethanolic extract showed IC50 value 38.89 ppm (11 times lower than ascorbic acid, IC50 of ascorbic acid was 3.35 ppm).

Abstract

Hasil penelitian kami tentang pencarian tanaman berkhasiat anti radang telah membuktikan bahwa jahe merah dapat menurunkan laju pembentukan prostaglandin secara in vitro, bahkan senyawa kimia di dalam jahe merah terbukti dapat menghambat enzim siklo oksigenase, yaitu enzim yang berperan penting pada terjadinya radang, melalui pembentukan ikatan hidrogen dengan residu asam amino penting di dalam enzim tersebut. Oleh karena itu, jahe merah dipilih sebagai tanaman yang akan dipopulerkan oleh Tim Pengabdian pada Masyarakat (PPM) Fakultas Farmasi Universitas Padjadjaran, kepada kader PKK di Desa Cikidang, Kecamatan Lembang, Kabupaten Bandung Barat. Kegiatan PPM ini meliputi (1) konsultasi kesehatan dan pemeriksaan tekanan darah peserta; (2) pendidikan masyarakat tentang radang dan pemanfaatan jahe merah sebagai swamedikasi radang; (3) difusi ipteks berupa demo pembuatan jahe merah instan sebagai minuman nutrisi. Dari sejumlah tujuh puluhun dangan yang disebarkan, kegiatan PPM dihadiri oleh empat puluh delapan peserta, terdiri dari 46 peserta wanita (95,83%) dan 2 peserta pria (4,17%). Hasil pre-test dan post-test menunjukkan adanya peningkatan pengetahuan peserta, dengan demikian dapat disimpulkan bahwa pembekalan pemanfaatan jahe merah kepada kader PKK di Desa Cikidang, Kecamatan Lembang, Kabupaten Bandung Barat, dapat meningkatkan pengetahuan mereka.

Abstract

The purpose of this study was to predict the bond mode, absorption and distribution profile as well as the mutagenicity and carcinogenicity of the asam gelugur fruit compounds which have the best inhibitory activity in neuraminidase influenza subtype H7N9 active pocket by in silico. Pharmacophore modeling and molecular docking using Ligandscout software. Absorption and distribution profiles using PreADMET software. Mutagenicity and carcinogenicity using Toxtree software. The results showed that 10 of the 13 compounds of asam gelugur fruit formed hydrogen bonds with important amino acid residues in the active sac neuraminidase H7N9. Organic acid group compounds have good absorption and distribution profiles. All acid gelugur fruit compounds are predicted not to be mutagenic and carcinogenic. Hydroxycitric acid is a compound that is predicted to have the best inhibition with the lowest Ki value of 1.09 µM. Abstract

The investigation efforts of an alternative treatment of the kidney stone disease by utilizing Indonesian native plants have still continued both for treatment and prevention. This study aims to strengthen the scientific evidence of the activity of celery as an anticalculi by in vivo assessment using hydroxyprolineinduced rat method. The white male wistar rats used in this study were intraperitoneally induced by hydroxyproline in order to form kidney stones, the celery extract preparations were then given at a dose of 50, 100, and 200 mg/kg of body weight orally for 5 days and observed following parameters: concentration of filtrate urinary calcium, concentration of urinary sediment calcium, as well as kidney to body weight ratio. The results showed that the concentration of filtrate urinary calcium in the test group at a dose of 200 mg/ kg body weight was significantly higher than those of negative control group, whereas the other test groups were not significantly different with the negative control group. The similar result was also shown on the parameter of calcium concentration in the urine sediment, while the ratio of kidney to body weight showed significant differences between all test groups and the negative control group. It can be concluded that the ethanol extract of the herb celery has an anticalculi activity in rats at a dose of 200 mg/kg of body weight.

Abstract

Genus Garcinia diketahui kaya dengan senyawa santon teroksigenasi, santon terprenilasi dan benzofenon poliisoprenilasi.  Beberapa dari golongan senyawa ini memiliki aktivitas biologi yang bervariasi antara lain sebagai antibakteri, antioksidan, sitotoksik, dan antimalaria.  Garcinia cowa termasuk famili Guttiferae dan di Indonesia dikenal dengan nama asam kandis.  Dalam rangka investigasi berkelanjutan dari tumbuhan genus Garcinia asal Indonesia suatu senyawa santon kowanin telah berhasil diisolasi dari bagian   kulit batang Garcinia cowa.  Struktur molekul dari senyawa ini telah ditetapkan berdasarkan data spektroskopi meliputi UV, IR, dan NMR 1-D serta data yang diperoleh  dibandingkan  dengan data  literatur. Abstract

BACKGROUND: Acetaminophen, when used at low doses is a safe drug, but at higher doses it induces apoptosis in hepatoma cells. Arcangelisia flava that grows widely in Kalimantan Island, Indonesia, contains berberine which is effective in protecting the liver. This work was aimed to study the effect of A. flava extract on the modulation of caspase-3 in acetaminophen-induced hepatotoxicity.METHODS: Thirty-five Wistar male rats were divided into groups: I the normal control (water); II the negative control (Arabic gum powder or PGA, 2% in suspension); III the positive control (silymarin); IV-VII (A. flava extract 100, 200, 400, and 800 mg/Kg of body weight (BW), respectively) for 14 days. At day 15th, group II-VII were induced with acetaminophen 1000 mg/Kg of BW per oral for 7 days along with the extracts. At day 22nd, the animals were measured for their serum glutamic oxaloacetic transaminase (SGOT), serum glutamic pyruvic transaminase (SGPT) and gamma-glutamyl transferase (GGT), histological examination, and Western blotting.RESULTS: Acetaminophen elevated the SGOT and SGPT (3x compared to normal group), and GGT (5x compared to normal group) of the animals in group II. Pre-treatment with higher doses of A. flava extract (group VI and VII) significantly prevented the biochemical changes induced by acetaminophen. Normal histology of the liver was showed by group I, III, VII, whereas dilated sinusoids, central vein (CV) lesion, and local haemorrage were observed in group II, IV, V and VI. Western blotting showed an inhibition of caspase-3 expression by A. flava extract in dose-dependent manner.CONCLUSION: A. Higher dose A. flava extract shows hepatoprotective activity by preventing the elevation of serum transaminases and transferase levels. Eventually, no damage in the acetaminophen-induced rat’s liver was observed. Abstract

BACKGROUND: Previous works indicated various pharmacology activities of bay plant (Syzygium polyanthum (Wight) Walp.), however only few studies investigated its toxicity. This work was aimed to study the subchronic toxicity of ethanol extract of this plant.METHODS: White Wistar rats were divided into 4 groups and were treated with 2% of Arabic gum (PGA) suspension, 1000 mg/kg of body weight (BW), 400 mg/kg of BW and 100 mg/kg of BW, respectively. The animals were observed on their body weight, hematology, clinical biochemistry parameters, organ index and histopathology.RESULTS: Flavonoids, tannins, polyphenols, saponins, quinones, monoterpenes and sesquiterpenes were detected in dried leaves and ethanol extract of bay plant. An increase of body weight in male and female groups treated with dose 100 and 400 mg/kg BW compared to controls, was observed. Moreover, there was an increase of white blood cell (WBC) in male and female groups treated with S. polyanthum extracts compared to controls, whereas a decrease of red blood cell (RBC) was observed in male groups treated with S. polyanthum extracts in dose-dependent manner compared to control. No significant changes of RBC were seen in female groups, haemoglobin values were not altered by extract treatment. Photomicrographs of liver, kidney, lungs, heart and spleen histopathology of male and female S. polyanthum extract-treated groups showed no significant alteration compared to controls.CONCLUSION: Our study revealed that S. polyanthum extracts does not show toxicity on the body weight, hematology, creatinine and serum glutamic pyruvic transaminase (SGPT), but fatty liver and necrosis are observed in female rats. This result can be beneficial for plant-based drug discovery, particularly this study provides information about the safety of S. polyanthum to be further developed as candidate of phytopharmaceutics.KEYWORDS: bay plants, salam leaves, hepatotoxicity, necrosis, SGOT, SGPT

Abstract

Inflamasi dapat menginduksi NOS (nitrat oksida sintase), yaitu enzim pengkatalisis pembentukan NO (nitrat oksida) berlebih sebagai pro-inflamasi. Akar kuning (Arcangelisia flava. L) banyak digunakan sebagai obat tradisional oleh masyarakat Asia Tenggara, baik sebagai obat luar maupun obat dalam. Senyawa kimia yang terkandung di dalam A. flava antara lain saponin, flavonoid dan tanin.  Akar tanaman ini juga mengandung glikosida dan alkaloid, terutama golongan isokuinolin, yaitu berberin, jatrorizin dan palmatin. Terdapat juga beberapa alkaloid minor seperti kolumbamin, dehidrokoridalmin, homoaromolin dan talifendin, serta diterpen fibraleusin. Fibraurin berpontensi sebagai anti-bakteri, anti-tumor, dan anti-inflamasi. Tujuan penelitian ini adalah untuk menelaah mode ikatan senyawa-senyawa kimia yang terkandung dalam A. flava terhadap enzim NOS dengan metode penambatan molekuler menggunakan perangkat lunak AutoDock. Hasil penelitian menunjukkan bahwa berberin, daidzein, dehidrokorildamin, epikatekhin, hidroksiekdison, jatrorizin, kaempferol, piknarin, kuersetin, dan talifendin dapat berinteraksi dengan enzim NOS pada kantung aktif  melalui pembentukan ikatan hidrogen dengan residu asam amino Glu377. Senyawa lainnya berinteraksi dengan residu asam amino tidak spesifik. Tidak ditemukan interaksi dengan Tyr347 pada semua senyawa. Hanya satu senyawa yang tidak membentuk interaksi dengan enzim NOS yaitu fibraurin.

Abstract

ABSTRAKTelah dilakukan penelitian mengenai aktivitas antiinflamasi fraksi-fraksi n- heksana, etil asetat, butanol dan air dari kulit batang sintok (Cinnamomum sintoc BL.) pada telapak kaki tikus putih jantan yang telah induksi edema dengan karagenan. Fraksinasi dilakukan dengan metode ekstraksi cair-cair. Penapisan fitokimia dilakukan terhadap serbuk simplisia kulit batang sintok. Pada uji aktivitas antiinflamasi, bahan uji diberikan pada tikus secara oral dengan dosis 1000 mg/kg berat badan (BB), indometasin10 mg/kgBB digunakan sebagai kontrol positif. Hasil penapisan fitokimia menunjukkanbahwa serbuk simplisia kulit batang sintok mengandung golongan senyawa polifenol, mono-seskuiterpen dan steroid. Hasil uji aktifitas antiinflamasi menunjukan bahwa fraksi air, fraksi butanol dan fraksi n-heksana memiliki aktifitas antiinflamasi dengan persentase inhibisi radang berturut-turut sebesar 64,36%, 53,67% dan 35,74% pada tingkat kepercayaan 95% dan 99% sementara indometasin sebagai kontrol positif memiliki persentase inhibisi radang sebesar 67,75%. Hasil ini menunjukkan bahwa fraksi air ekstrak kulit batang sintok memiliki potensi sebagai agen antiinflamasi alami.Kata kunci : Aktivitas antiinflamasi, Sintok, tikus putih jantan
