Abstract

Mahkota Dewa [Phaleria macrocarpa (Scheff). Boerl.] is used traditionally to treat cancers in Indonesia. Extract methanol of the leaves displayed a small LC50 value (63.16 μg/ml) on BST (Brine Shrimp Lethality test) assay; therefore a phytochemical study of this extract was undertaken. A new benzophenoic glucoside was isolated from the methanol extract and identified as 4,5-dihydroxy,4’-methoxybenzophenone-3-0-β-D-glucoside (Phalerin) based on its spectroscopic data. Phalerin was cytotoxic to myeloma cell line (NS-1) (in vitro) having IC50 of 83 μg/ml or 1.9 x 10-1 mM.Key words: Phaleria macrocarpa, Phalerin, myeloma

Abstract

Telah dilakukan penelitian tentang keanekaragaman jenis tumbuhan obat di darah aliran sungai Opak. Tujuan penelitian ini adalah untuk mengetahui komposisi jenis-jenis tumbuhan obat, distribusi dan potensi pengembangan sebgai bahan baku herbal.Area kajian dibagi menjadi 6 lokasi berdasarkan ketinggian tempat, yaitu wilayah Kecamatan Cangkringan (CNK), Ngemplak (NPL), Prambanan (PRB), Kalasan (KLS), Imogiri (IMO), dan Pundong (PDO).  Setiap lokasi diletakkan plot kuadrat berukuran 2x1 m sebanyak 30 plot. Di dalam plot kuadrat dihitung jenis tumbuhan dan cacah individu setiap jenis tumbuhan.Hasil penelitian menunjukkan terdapat sebanyak 79 jenis tumbuhan yang dikelompokkan dalam 27 suku. Berdasarkan kontribusi jenis di dalam lingkungan dan studi literatur Eupatorium riparium,  Andrographis paniculata, dan Scoparia dulcis merupakan jenis-jenis tumbuhan yang potensial untuk dikembangkan sebagai bahan baku obat herbal.

Abstract

With regard to collaboration research called Widya Nusantara Exploration (EWIN) in May-June 2013 and November 2014, a study on isolation of actinomycetes from sediments of Makassar Strait have been conducted. Actinomycetes is one of microbe which has an excellent track record in producing antimikrob and other active substances. But due to terrestrial actinomycetes has been widely explored, then recently researchers began focusing on wide variety of extreme environments, such as marine environment, to screening aktinomisetes in producing new secondary metabolites. A total of 36 strains of actinomycetes were isolated from 10 samples obtained from deepsea floor in Makassar Strait, Indonesia, Direct Dillution Method were best used to isolate the actinomycetes compare to Sodium Dodecyl Sulfida – Yeast Extract Method (SDS-YE Method) and Rehidration Centrifugation Method (RC Method). NBRC-802 media and Actinomycetes Isolation Agar(AIA)(Himedia)media were used as the isolation media. All the isolates were identified by morphological characteristic and by analysis of 16S rRNA gene sequence. Actinomycetes isolated from deepsea floor of Makassar Strait have been dominated by Micromonospora (58%), Verrucosispora (14%)Streptomyces (8%) and Luteipulveratus (5%), however genus Nocardiopsis, Micrococcus, Gordonia, Kytococcus, and Arthrobacter were not dominant (3%). Station 25 in 1.547 m depth was the most abundant of actinomycetes, 18 strains and dominated by the genus Micromonospora which is isolated using Direct Dillution Method and both NBRC 802 or AIA media.

Abstract

BACKGROUNDThe leaves of Nerium indicum Mill. have been utilized traditionally to cure cancer. By using Bioassay guided extraction and isolation method, three compounds (NiO-1, NiO-2, NiO-3) were isolated from an active fraction of the CHCl3 extract of N. indicum leaves. NiO-2 identified as 5α- oleandrin is the best cytotoxic compound on HeLa cervical cancer cell in vitro among the other two. However mechanism of action of the compound hasn’t been evaluated yet. The aims of this study were to determine the mechanism of action at molecular level.METHODSThe action mechanism of 5α-oleandrin on HeLa cervical cancer cells was analyzed by staining the cells with Hoechst 33342; the agarose gel electrophoresis was aimed to determine the DNA fragmentation and the western blotting was aimed to determine the Bcl-2 and Bax protein expression.RESULTSIncubation of HeLa cervical cancer cell with 5α-oleandrin at the concentration 3,47x10-4 mM (24 hours) followed by staining with Hoechst 33342, a broken up light blue color of nucleus was observed (compared with intensive color of untreated control). By gel electrophoresis (at the same concentration of the tested compound), a smear band at about 200 bp was observed. In addition, cells treated with 5α-oleandrin displayed a decreasing of the Bcl-2 protein expression and increasing of the Bax protein expressionCONCLUSION5α-oleandrin induced HeLa cervical cancer cells DNA fragmentation observed by the presence of a smear band at about 200 bp indicative the apoptotic occurrence. 5α-oleandrin induces apoptosis by reducing the Bcl-2 protein expression but the Bax protein expression increases.

Abstract

Tumbuh-tumbuhan diketahui kaya antioksidan alami seperti vitamin E, vitamin C, beta karoten dan flavonoid.Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan10 jenis sayuran, Amaranthus lividus L.sensu Thell., Cichorium endivia, Brassica juncea (L.) Czern., Sauropus androgynus (L.) Merr., Ocimum bassilicum, Brassica oleracea var.botrytis  L.,  Brassica oleracea var.botrytis  L. subvar cauliflora DC., Brassica oleracea var.capitata L., Brassica oleracea var.capitata L. forma rubra, Cosmos caudatus H.B.K dan untuk mengetahui seberapa besar kontribusi senyawa fenolik dan flavonoid pada aktivitas antioksidannya. Uji aktivitas antioksidan dilakukandengan metode DPPH (2,2-difenil-1-pikril hidrazil) dengan menggunakan baku pembanding kuersetin. Analisis kandungan fenolik total ditentukan oleh kemampuan sampel untuk mereduksi reagen Folin Ciocalteau.Analisis flavonoid total dilakukan secara spektrofotometri didasarkan adanya kemampuan flavonoid  membentuk kompleks yang berwarna kuning dengan AlCl3 , yang selanjutnya akan bereaksi dengan  NaOH membentuk warna merah muda yang dapat diukur absorbansinya pada panjang gelombang 510 nm. Hasil penelitian menunjukkan ekstrak kenikir (Cosmos caudatus H.B.K.) memiliki aktivitas antioksidan yang paling tinggi dengan nilai IC50 19,49 µg/ml. Hubungan antara aktivitas antioksidan 10 jenis sayuran dengan kandungan fenolik total diperoleh persamaan regresi linear   y = -34,94 x +562,5, r2 = 0,235 , sementara itu hubungan aktivitas antioksidan dengan flavonoid totalnya diperoleh persamaan y = -16,57 + 755,9, r2 = 0,471.Kata kunci : antioksidan, DPPH, sayuran, fenolik total, flavonoid total

Abstract

Spermatogenesis effect of  ethanol extract senggani root (Melastoma affine D. Don) was proved. Furthermore, the extract was fractionated with ethyl acetate and was investigated toward spermatogenesis effect to explore natural product  as male  contraception. The subject of this research were five groups of thirty two fertile male Wistar rat,. Four groups were treated with ethyl acetate fraction of senggani root in gradually 500; 250; 125; and 62,5 mg/kg body weight dose. The fifth group as negative control was treated by CMC Na 0,5 %. Every groups was treated by orally administration for twenty two days period of time, once a day. In twenty third day, the rats were killed then observed toward the motility, sperm count, and testis histopathology. Results of the research indicated that there were influence toward spermatogenesis effect from ethyl acetate fraction of senggani root, especially at sperm motility and sperm count. Decrease of sperm motility after treatment was compared with negative control group. The percentage of the sperm count decreased at 500; 250 mg/kg body weight dose from ethyl acetate fraction was 49,74 % and 48,21.This decreased probably were caused by seminiferous tubules necrotic.   ABSTRAK Pemberian ekstrak etanol akar senggani (Melastoma affine D. Don) telah terbukti dapat menyebabkan berkurangnya jumlah dan motilitas sperma tikus. Berdasarkan hasil penelitian tersebut dilakukan fraksinasi ekstrak etanol akar senggani dengan etil asetat kemudian fraksi itu di uji cobakan pada tikus dengan tujuan untuk eksplorasi bahan alam yang dapat dipakai sebagai bahan kontrasepsi pria. Penelitian ini menggunakan 32 ekor tikus jantan fertil galur Wistar, umur 2,5-4 bulan dengan bobot 145-350 g dan dibagi menjadi 5 kelompok. Empat kelompok merupakan kelompok perlakuan fraksi etil asetat akar senggani dengan dosis 500; 250; 125; dan 62,5 mg/kgBB. Kelompok V merupakan kelompok kontrol negatif dengan perlakuan CMC Na 0,5 %. Semua kelompok diberi perlakuan secara per oral selama 22  hari dengan frekuensi pemberian sekali sehari. Pada hari ke-23 tikus dikorbankan dan diamati motilitas, dan jumlah atau angka sperma. Dilakukan juga pengamatan histopatologi organ testis. Hasil penelitian menunjukkan ada pengaruh pemberian fraksi etil asetat akar senggani terhadap motilitas dan jumlah (angka) sperma. Motilitas sperma menjadi berkurang pada semua dosis perlakuan jika dibandingkan dengan kelompok kontrol. Penurunan motilitas sperma terbesar pada dosis 250 mg/kgBB. Jumlah sperma juga mengalami penurunan untuk semua dosis perlakuan. Persentase penurunan jumlah sperma terbesar pada dosis 500 ; 250 mg/kgBB sebesar 49,74 % , dan 48,21 %. Penurunan ini kemungkinan diakibatkan oleh rusaknya tubulus seminiferus testis yang mengalami nekrosis

Abstract

Kembang sepatu flower (Hibiscus rosa-sinensis L.) was fractionally used as expectorant. Based on Bioassay Guided fractionation, an active fraction was separated, and the fraction was identified is Alkaloid was the major compound based on TLC analysis. Viscosity value measured by viscometer was used as a Bioassay model of expectorant activity in vitro and acetyl cysteine was used as positive control. Alkaloid content determination of the ethanolic extract was measured by TLC-Densitometric compared with standard curve of isolated alkaloid as the selected marker (Y=12,1360X+2901,4474). The alkaloid content in the ethanolic extract was determined as 2.35±0,67%.Keywords : alkaloid, ethanolic extract, Hibiscus rosa-sinensis L.

Abstract

Eupatorium riparium Reg. adalah tumbuhan obat penting asli dari Mexico dan India Barat, yang masuk ke tanah Jawa sejak tahun 1800. Tumbuhan ini mempunyai catatan sejarah digunakan untuk obat tradisional dalam berbagai kultur budaya bangsa secara luas di seluruh dunia dan biasa digunakan untuk obat hipertensi, gagal jantung, diuretik, antikanker, antifungi, dan penyakit yang disebabkan oleh bakteri. Studi pada HeLa cell line ini dilakukan selama satu bulan. Selanjutnya, studi ini bertujuan meneliti aktivitas antiproliferatif ekstrak wasbensin daun E. riparium terhadap kanker servik manusia Hela cell line. Aktivitas antiproliferatif diuji menggunakan reagen proliferatif sel WST-1 dengan waktu 1, 2, dan 4 jam setelah diinkubasi selama 72 jam pada suhu 37oC dan 5%CO2. Hasil penelitian menunjukkan bahwa ekstrak wasbensin daun E. riparium mempunyai aktivitas proliferatif yang potensial terhadap HeLa cell line dengan nilai IC50 berikut 102.69 𝜇g/ml (1 jam), 198.67 𝜇g/ml (2 jam). Saran selanjutnya, penelitian lanjutan perlu dilakukan untuk mengetahui mekanisme antikanker HeLa cell line.Kata kunci: Eupatorium riparium Reg, antiproliferatif, HeLa, WST-1
