Abstract

The purpose of this study was to find out whether the refill water (AMIU) was contaminated with coliform or not. The methods used were presumptive, confirmed, and completed test. Sample of this study was refill water (AMIU) from five refill water shops located in Ilie village, sub-district of Ulee Kareng, Banda Aceh. The sample was taken 3 times with interval period of one week. Sample was culture in lactose broth and incubated at 37° C for 48 hours. If it did not form gas in Durham tube, presumptive test result was considered negative and the samples were not proceed to the other test. The data was presented using descriptive analysis. It could be concluded that AMIU from 5 shops in Ilie Ulee Kareng, Banda Aceh is negatively contaminated with coliform bacteria.

Abstract

The research aimed to isolate Salmonella sp.in feces of broiler chicks. Fifteen feces samples of broiler chick were obtained from Ulee Kareng Market in Banda Aceh. Chick cloaca was swabbed using a sterile cotton swab, put into the microtube, and brought to the Microbiology Laboratory of Veterinary Medicine Faculty, Syiah Kuala University. The samples were incubated in selenite cystine broth (SCB) medium, cultured in salmonella shigella agar (SSA) and separated colonies were inoculated in nutrien agar (NA) at 37° C for 24 hours. Bacteria stained with Gram staining then tested biochemically. The result showed that Salmonella sp. were identified in 9 out of 15 feces chicks samples. It ca be concluded that broiler chick in Ulee Kareng Market Banda Aceh contaminated with bacteria Salmonella sp.

Abstract

This study aims to determine the effect of storage time on the total number of bacteria in nile tilapia (Oreochromis niloticus) flesh stored at a temperature of 20 C and at different retention time. This study used split plot completely randomized design (CRD) with 2 treatment maintenance temperature of 29 C and 35 C. Experimental animals used were 30 nile tilapia fish grouped into five storage group namely 0 (P0), 7 (P1), 14 (P2), 21 (P3), and 28 (P4) days. The results showed that total number of bacteria found in fish flesh was higher than normal standard based on ISO 7388:2009 which was 5x105 cfu/g. The average of total numbers of bacteria in flesh of nile tilapia fish reared in 29 C on P0, PI, PII, PIII, and PIV were 6.55±0.19, 6.99±0.07, 6.96±0.08, 6.96±0.28, and 7.18±0.15, respectively. While the average of total numbers of bacteria in flesh of nile tilapia fish maintained in temperature of 35 C were 6.68 ± 0.10, 7.00±0.05, 7.19±0.12, 7.24±0.12, and 7.35±0.09, respectively. Total number of bacteria in nile tilapia flesh was affect significantly (P0.05) by temperature and storage temperature maintenance. In conclusion, storage time and different tempera ure maintenance of nile tilapia can increase total amount of bacteria on fish flesh.

Abstract

This research aims to identify the Staphylococcus aureus (S. aureus) bacteria from preputium and vagina of horses. Five swabs preputium and 5 swabs vagina samples from 3-9 years olds horse in the village of Bebangka, Gelelungi, Reloop, and Kelapeng, Pegasing Aceh Tengah District were used in this study. Preputium and vagina swabs of horses were dipped into reaction tube containing Nutrient Broth (NB). Staphylococcus aureus was identified on manitol salt agar (MSA). Separated colony colored bright yellow on the media were tested for Gram staining, hemolysis, catalase, and sugar test (glucose and manitol). The result showed that the isolates growth on MSA were colored bright yellow, Gram positive, β hemolisis, and catalase positive. The isolates fermented manitol and glucose. Isolates identified from horses preputium and vagina showed the characteristic of S. aureus. In conclusion S. aureus bacteria were positively identified from all of swabs samples, both in preputium and vagina of horses.Key words: Staphylococcus aureus, horse, preputium, vagina

Abstract

This study was aimed to determine ratio of bursa body weight in ALPU chickens. The samples used were 72 ALPU chickens consisting of 36 males and 36 females aged 1-18 weeks. This study was observed in every week by weighing the weight of ALPU chickens as much as 2 males and 2 females, then chickens slaughtered. After that, Fabricius's bursa on each chicken was separated from the chicken body part and weighed using a digital balance sheet. The results of this study showed that the maximal ratio in male ALPU chickens was 0.0032 at the age 2 weeks, while the maximal ratio in female ALPU chickens was 0.0036 at the age 4 weeks. From the results of the analysis using t-test was concluded that bursa body weight ratio in male and female ALPU chickens during starter to grower group was significantly different (p0.05). The grower to finisher group was not significantly different (p0.05).
