Abstract

Groupers are bio-indicators of the health of coral ecosystems and high economic value. Various speciesof grouper fish sold in markets in Indonesia is not known. The purpose of this study was to identify thespecies of grouper fish market located in Karangasem and Kedonganan. The method used in this studywas polymerase chain reaction(PCR) techniqueand mitochondrial DNA 16S rRNA gene as a molecularmarker. Grouper samples were collected from the fish market Karangasem total of 11 samples and 42samples collected from Kedonganan. Sequences from each sample was obtained from the analysis inBerkeley Sequencing Facility, California. Sequencing samples were then analyzed by BLAST and matchedwith the sequences found in GenBank. The analysis showed that six species can be identified from the fishmarket Karangasem namely Cephalopholis leopardus, C.cyanostigma, C. miniata, C. sonnerati, Variolaalbimarginata, and Epinephelus fasciatus. Seven species can be identified in the fish market Kedonganannamely E. fuscoguttatus, E. coeruleopunctatus, E. merra, E. polyphekadion, C. cyanostigma, C. miniata andV.albimarginata.

Abstract

Newcastle disease (ND) is a very harmful avian disease, endemic in Indonesia and various parts of the world. The causative agent is ND virus or Avian orthoavulavirus 1 (AOAV-1). This virus is an RNA virus with wide genetic variation. Based on the genome length, it can be classified into AOAV-1 Class I and II. Class I are generally avirulent whereas Class II are consist of both virulent and avirulent viruses, currently there are 18 genotypes of the class II. To find out the molecular characteristics of AOAV-1 currently circulating in the field, isolation and identification of viruses from laying hens that was suspected ND from Tabanan Bali in 2017, was performed. The isolated viruses hereafter named as Tabanan1/ARP / 2017. A one-step RT-PCR reaction was carried out to amplify NP, F and HN gene fragments from the virus using three specific pairs of AOAV-1 primers. The obtained nucleotide sequences are then used in phylogenetic analysis. For phylogenetic analysis several strains of AOAV-1 from class II representing genotype I-VII as well as one strain from Class I were accessed from GenBank. From the analysis of the F gene nucleotide sequences, it was found that Tabanan 1 / ARP / 2017 is a genotype VII virus with an amino acid sequence at the F protein cleavage site is 112 R-R-Q-K-R-F117, a typical virulent strain. Phylogenetic analysis using nucleotide sequences NP and HN genes also positioned this isolate in genotype VII. At the nucleotide level, genetic distance with virulent isolates that was isolated in 2007 and 2010 were 8.26% and 1.08% while at the amino acid level were 5.26% and 0.64%. There were found mutations in amino acids at positions 107 and 108 of F protein.

Abstract

The purpose of this study is to discover about heterozygosity, migration between populations, and mating habits of Macaca fascicularis among the population in Nusa Penida Island, Klungkung, Bali. Fourteen blood samples from Macaca fascicularis were taken from Nusa Penida Island that consist of eleven samples from Puncak Mundi Temple and three samples from Paluang Temple were used in the study. Samples were extracted using QIAamp DNA Blood Mini Kits produced by Qiagen. Five loci microsatellite (D10S611, D11S1366, D13S765, D17S1290, and D18S536) were amplified using the Polymerase Chain Reaction (PCR) technique. Thirty cycles of PCR were carried out with an annealing temperature of 500C. Alleles were separated by electrophoresis on eight percent polyacrylamide gel and presented with silver staining. The results showed high heterozygosity is 0.762 of Macaca fascicularis populations on Nusa Penida Island with random mating habits. Low genetic differentiation is 0.05. High the number of migrations between populations (genetic flow) of Macaca fascicularis populations on Nusa Penida Island is 4.75 with migration between populations of five individual/generation. Based on the results of the study it was concluded that the genetics of the Macaca fascicularis population on Nusa Penida Island varies.

Abstract

Sumatran elephant (Elephas maximus sumatranus) is the biggest land mammals in Indonesia in criteria A2c category Critically Endangered (CR) dan Appendix I in Convention on International Trade in Endangered Species of Wild Flora and Fauna (CITES). Considering the importance of conservation and health maintenance of Sumatran elephants, various diagnostic tests must be carried out to determine the health status of elephants ranging from physical examination to blood chemistry test and urinalysis.The aim of this study was to obtain the chemical profile of the Sumatran elephant (Elephas maximus sumatranus) which is captive in the Bali Elephant Camp, Carangsari, Petang, Badung Regency, Bali. The urine sample taken is mid stream (middle emission) urine. Sampling was carried out 5 times for 2 weeks with 2-3 days distance between samples taken and the dipstick test was carried out twice in one sample. Based on the results of this study, it can be concluded that of the 8 Sumatran elephants maintained at the Bali Elephant Camp all are 100% negative for glucose. Meanwhile, protein levels were found in 5 individuals (62.5%) in the range 0-2000. Positive bilirubin and hematuria were found in one individual (12.5%). Ketone levels are found in the range 0-160 in 7 individuals. A total of 84.7% of urobilinogen data (mg / dL) is 0.2. The pH value is in the range of 6-9, while the value of density is in the range 1-1,020. Leukocyte level (Leu / µL) is worth 70 (91.3%).

Abstract

Tuna is an export commodity which has very high economic value. However, some tuna speciesare threatened with extinction. The purpose of this study was to identify the tuna species that aresold in Kedonganan Fish Market. The research method was polymerase chain reaction technique(PCR) using the marker sequence mitochondrial DNA control region. Samples were obtained fromthe Fish Market tuna Kedonganan, Kuta, Badung, Bali. The total number of samples are 28specimens. Sequence from each sample was obtained through sequencing techniques. Sequencesobtained were run in BLAST (Basic Local Alignment Search Tool) and subsequently analyzed withMEGA 5 for species confirmation. Three species of tuna that are identified in the Kedonganan FishMarket is: Thunnus albacares, T. obesus, and Katsuwonus pelamis. All three species have highgenetic variation HD = 1. This study needed to be continued with more number of samples todetermine the species of tuna sold in Kedonganan Fish Market.
