Abstract

The present study aimed to isolate gram-negative bacteria infected metacarpal of Panthera tigris sumatrae trapped in Subulussalam, Aceh Province. Swab sample was obtained from metacarpal injury of P. tigris sumatrae. Swab sample was cultured to nutrient broth media using sterile cotton swabs or Pasteur pipettes, and incubated at 37°C temperature for 24 hours. Culture was spared on MacConkey media and incubated again at 37°C temperature for 24 hours. Determination of bacteria colony growth on the surface of MacConkey media based on shape, colour, surface, size, and viscosity (consistency). The bacteria colony stained with Gram staining, and tested biochemically.  The result showed that gram-negative bacteria such as Citrobacter sp., Proteus sp., Providencia sp., Pseudomonas sp.,and Salmonella sp. isolated from metacarpal injury of P. tigris sumatrae.

Abstract

This study aims to determine the effect of chitosan gel on the number of white mouse leukocytes (Rattus norvegicus) on incision wound healing. This study used 10 female white rats that were 2 months old and weighed ± 165 g, that was designed with a Split-plot design using 10 rats and was divided into 2 treatment groups with each treatment group consisting of 5 replications. Treatment I (control) incision wound smeared with gentamicin ointment, treatment II incision wound smeared with chitosan gel 2 times a day for 14 consecutive days. This study used chitosan gel with a 5% concentration. Blood sampling through the lateral tail vein as much as 0.5 ml and observation of the number of leukocytes was carried out on days 0, 3, 7 and 14 after treatment. Examination of the number of leukocytes is done with a hemocytometer and using the Neubauer method. The parameters of this study are the number of leukocytes in the incision wounds healing process in white rats. The research data were analyzed using analysis of variance based on the Split-plot Design with the SPSS program. The number of leukocytes in group I (gentamicin) vs group II (chitosan) on days 0; 3; 7; and 14 was 7.89×104/mm3 vs 7.30×104/mm3; 6.28×104/mm3 vs 10.23×104/mm3; 7.27×104/mm3 vs 10.28×104/mm3; and 7.02×104/mm3 vs 7.52×104/mm3, respectively. From the results of this study it can be concluded that the use of chitosan gel is better than gentamicin ointment in the healing process of incision wounds in white rats. The results of thisstudy can be followed by conducting research on other animals for wound treatment applications.

Abstract

The aim of this study is to determine the reproduction performance of local does in which theirestrous was induced by short synchronization system. In this study 10 healthy female, unpregnant andhave relatively homogenous body weights were used. All does were devided into 2 experiment groups.Group 1 consisted of 4 does as control and group II consisted of 6 does as treatment. Group I, were injectedwith 125 ?g cloprostenol intramuscular, twice in 11 days interval. Group II, was treated with shortsynchronization using 125 ?g cloprostenol intramuscular, and then followed by injection of 300 IU hCGand 0.5 mg estradiol benzoate after 12 hours later. The does were inseminated 10 hours after onset ofestrous and repeated 12 hours later. Parameters measured were oestrous percentage, pregnancy, andlitter size. Results showed that all does (100%) from both groups showed estrous. Percentage of pregnantdoes group I and II were 75.00% and 83.33% respectively, and the averages litter size were 2.0 ± 1.0 and 1.4± 0.3, respetively. Treatment with short synchronization can increase pregnancy and delivered percentagealthough litter size not affected.

Abstract

The aim of this study was  to determine the effect of pituitary extracts on the reproductive threeperformance of local does. This study used 15 local female does that were divided into three treatmentgroups.  Each group consisted of five does.  All does were injected with 7.5 mg Prostaglandin F2? /PGF2?(Prostavet C, Virbac S.A) intramuscularlly, twice in 10 days interval. In group 1 (K1), the does wereinseminated 36 hours after last PGF2? administration.  In group II (KII), does were injected with 500 IUpregnant mare serum gonadotropine/PMSG (Folligon ™, Intervet, Boxmeer, Holland) on day 9 cycles andfollowed with injection of 7.5 mg PGF2? two days after. In group III (KIII), goats injected with pituitaryextract on days 9, 10, and 11 cycles with decreasing dose :  2, 1, and; 0.5 ml intramuscular injection locatedat some point, followed with injection of 7.5 mg PGF2? 2 days after. The does on the KII and KIII wereinseminated 36 hours after PGF2? administration. Parameters measured were oestrous percentage,pregnancy, kidding rate, twinning, total of litter size, and averages litter size.  Results showed that alldoes (100%) from all groups showed estrous and kidding rate. Percentage of pregnant does group I, II, andIII were  60, 40, and 80%, respectively. Percentage of twinning group I, II, and III were 0, 50, and 25%.  Thetotal of litter size group I and II were 3 does, and group III were 5 does. The averages litter size group I, II,and III were 1; 1.5; and 1.25 respectively. In conclusion, treatment with pituitary extracts can increasereproductive performance of local does.

Abstract

Salah satu upaya untuk meningkatkan konsentrasi testosteron adalah dengan pemberian gonadotropin releasing hormone (GnRH). Peningkatan testosteron menyebabkan peningkatan kualitas spermatozoa. Penelitian ini bertujuan mengetahui pengaruh pemberian gonadotropin releasing hormone (GnRH) terhadap peningkatan kualitas semen dan level testosteron domba waringin. Dalam penelitian ini digunakan tiga ekor domba waringin dengan rancangan pola bujur sangkar latin 3 x 3 sehingga hewan percobaan menerima suntikan NaCl fisiologis sebagai kontrol (K0), 50 ìg GnRH (K1), dan 100 ìg GnRH (K2). Penampungan semen dilakukan satu kali ejakulasi/minggu, selama tiga minggu. Sampel semen dikoleksi menggunakan elektroejakulator 24 jam setelah perlakuan dan diamati warna, konsistensi, volume, motilitas, konsentrasi, viabilitas, dan abnormalitas spermatozoa. Koleksi darah untuk pemeriksaan konsentrasi hormon testosteron dilakukan 60 menit setelah penyuntikan GnRH. Analisis konsentrasi testosteron dilakukan menggunakan metode enzyme linked immunosorbent assay (ELISA). Data mengenai warna dan konsistensi semen dilaporkan secara deskriptif, sedangkan level testosteron, volume semen, motilitas, konsentrasi, viabilitas dan abnormalitas spermatozoa dianalisis dengan analisis varian. Hasil pengamatan menunjukkan bahwa warna dan konsistensi semen yang dikoleksi pada semua kelompok perlakuan adalah krem dengan konsistensi kental. Hasil analisis statistika menunjukkan bahwa volume semen, konsentrasi spermatozoa, motilitas spermatozoa, viabilitas spermatozoa dan abnormalitas spermatozoa setelah pemberian GnRH menunjukkan perbedaan yang tidak signifikan (P>0,05). Rata-rata (±SD) konsentrasi testosteron pada kelompok K0, K1, dan K2 masing-masing adalah 1,82±1,08; 8,05+2,24; dan 8,81±1,09 ng/mL (P<0,05). Disimpulkan bahwa pemberian GnRH tidak memengaruhi kualitas semen namun dapat meningkatkan konsentrasi hormon testosteron pada domba waringin.

Abstract

Telah dilakukan penelitian tentang struktur histologi lambung (rumen, retikulum, omasum, dan abomasum) sapi aceh. Penelitian ini bertujuan mengetahui struktur histologi lambung sapi aceh. Sampel penelitian diambil dari 3 ekor sapi aceh berjenis kelamin jantan, telah dewasa kelamin yang dipotong di Rumah Potong Hewan di Aceh Besar. Terhadap sampel penelitian dilakukan proses mikroteknik untuk selanjutnya dilakukan pewarnaan Hematoksilin-eosin (HE). Pengamatan terhadap struktur histologi menggunakan mikroskop cahaya binokuler. Berdasarkan hasil penelitian, dapat disimpulkan bahwa struktur histologi lambung sapi aceh tidak berbeda dengan struktur histologi lambung ruminansia lainnya (sapi, domba, dan kambing), yaitu terdiri dari empat lapisan, tunika mukosa, tunika submukosa, tunika muskularis, dan tunika serosa. Lamina propria rumen menyatu dengan submukosa membentuk propria-submukosa dan terdapatnya kapiler fenestrated dibawah membran basal. Ciri khas dari retikulum sapi aceh terdapat pada lamina muskularis mukosa yang membentuk lipatan memanjang. Tunika mukosa abomasum terdapatnya kelenjar yaitu kelenjar kardia, fundus, dan pilorus.(A Study to detect the microscopic structure of gastric (rumen, reticulum, omasum, and abomasum) in aceh cattle. The purpose of this research was to know the histological structure of the gastric in aceh cattle. The samples were collected from 3 of male aceh cattle in Aceh Besar abattoir. The tissue samples were processed by microtechnique and Hematoksilin-eosin (HE). Microscopic analysis was performed using binocular light microscope. The study showed that the wall of gastric aceh cattle not different with another ruminasia (cow, sheep, and goat), they are made up of four layers, that was tunica mucosa, submucosa, muscularis, and serosa. The mucosa segment there are lamina epithelia, lamina propria, and lamina mucosa muscularis. At rumen, lamina propria merges to submucosa created submucosa-propria and founded fenestrated capiler under epithelium basal membran. The characterstic of reticulum based on mucosa muscularis which is create fold lengthwise. Mucosa of abomasum founded some glands, there are cardiac, fundic, and pyloric).

Abstract

ABSTRAK                Penelitian ini bertujuan untuk mengetahui keberadaan dan jenis bakteri enterik pada feses gajah sumatera (Elephas maximus sumatranus) di Pusat Konservasi Gajah (PKG) Saree, Aceh Besar. Sampel feses segar dikoleksi pada pagi hari dari 6 ekor gajah sumatera dengan cara feses langsung diambil dari anus menggunakan swab dan dimasukan dalam Nutrient broth lalu disimpan dalam Cooler box steril. Penelitian ini menggunakan metode Carter (1987) yang sudah dimodifikasi. Untuk isolasi dan identifikasi bakteri enterik pada feses gajah sumatera. Semua sampel feses dikultur pada media MacConkey, Salmonella Shigella Agar (SSA), IMViC (Indol, Methyl red, Voges-Proskauer, sulfid indol motility, Simmons citrate), media Triple Sugar Iron Agar (TSIA), dan uji fermentasi gula-gula (glukosa, laktosa, sukrosa, maltosa). Data hasil penelitian ini dianalisis secara deskriptif berdasarkan keberadaan bakteri enterik yang terdapat dalam feses. Dari hasil penelitian diketahui bahwa di dalam feses lima ekor gajah sumatera yang berumur di atas enam tahun terdapat lebih dari satu bakteri enterik, sedangkan satu ekor gajah yang berumur satu tahun hanya terdapat satu jenis bakteri enterik. Pada gajah bernama Midok ditemukan bakteri E. Coli dan Salmonella sp, pada gajah Amoy bakteri E. Coli dan Salmonella sp, gajah bernama Ella ditemukan bakteri Enterobacter sp dan Salmonella sp, gajah bernama Senna bakteri E. Coli dan Citrobacter sp, gajah bernama Osin ditemukan bakteri E. coli dan Salmonella sp, dan gajah bernama Junaidi ditemukan bakteri E. coli.  Dari penelitian ini dapat disimpulkan bahwa bakteri enterik yang terdapat pada feses gajah sumatera adalah Eschericha coli, Salmonella sp, Enterobacter sp, dan Citrobacter sp.

Abstract

ABSTRAKPenelitian ini dilakukan untuk mengetahui keberadaan telur dan larva nematoda pasca pemberian anthelmintik pada gajah sumatera (Elephas maximus sumatranus) di Conservation Response Unit (CRU) Sampoiniet Aceh Jaya dan Pusat Konservasi Gajah (PKG) Saree. Sampel feses gajah sumatera diambil dari 4 ekor gajah yang ada di CRU Sampoiniet Aceh Jaya dan 1 ekor gajah yang ada PKG Saree. Pengambilan sampel feses gajah Sumatera dilaksanakan sebanyak 3 kali seminggu. Hasil penelitian menunjukkan tidak ditemukan telur maupun larva pada gajah-gajah yang berada di CRU Sampoiniet. Ditemukan telur cacing Ascaris sp dan Toxocara spp serta ditemukan juga larva dari cacing Rhabiditiform dan Ascaris sp pada gajah di PKG Saree. Efikasi obat cacing masih optimum selama 3 minggu pasca pemberian obat cacing.Kata kunci: Telur cacing nematoda Ascaris sp, Toxocara spp, Larva Rhabiditiform, Ascaris sp, Gajah sumatera (Elephas maximus sumatranus)ABSTRACT This research was conducted to determine the eggs and larvae of namatode after anthelmintic administation on sumatran elephant (Elephas maximus sumatranus) in Conservation Response Unit (CRU) Sampoinet Aceh Jaya and Conservation Center of Elephant (CCE) Saree. The  Sample feces of sumatran elephant takes from 4 elephant in CRU Sampoiniet Aceh Jaya and 1 elephant in CCE Saree. The results showed no eggs or larvae are found on the elephants residing in CRU Sampoiniet. Ascaris worm eggs found sp and Toxocara spp and found larvae of Ascaris sp and Rhabiditiform worms in CCE Saree. The feces sample of sumatran elephant was taken three times, conducted every week. It was conducted, the elephant in Sampoiniet could not be found the eggs of parasite in feces due to anthelmintic treatment.Keywords: Egg worm nematodes Ascaris sp, Toxocara spp, Rhabiditiform larvae, Ascaris sp, Sumatran elephant (Elephas maximus sumatranus)

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui apakah harimau sumatera (Panthera tigris sumatrae) dan harimau benggala (Panthera tigris tigris) di Taman Margasatwa Medan terinfestasi cacing parasit gastrointestinal dan apa saja jenis dari cacing parasit tersebut. Sampel pada penelitian ini menggunakan feses yang dikoleksi untuk pemeriksaan telur cacing yang didapat dari 9 ekor harimau sumatera dan 6 ekor harimau benggala. Sampel diambil tiga kali dalam waktu yang berbeda dengan interval 2 minggu. Sampel feses diambil pada pagi hari dan dimasukkan ke dalam botol sampel dan ditambahkan dengan formalin 10% dengan perbandingan 1:1. Sampel dibawa dengan menggunakan ice box yang telah diisi es ke Laboratorium Parasitologi Fakultas Kedokteran Hewan Univesitas Syiah Kuala untuk dilakukan pemeriksaaan. Pemeriksaan sampel dengan menggunakan metode sentrifugasi,  metode McMaster, dan metode sedimentasi Boray. Hasil penelitian ini menunjukkan harimau sumatera (Panthera tigris sumatrae) dan harimau benggala (Panthera tigris tigris) di Taman Margasatwa Medan terinfestasi Toxocara cati dan Ancylostoma tubaeformae.  Kata Kunci: Harimau Sumatera, Harimau Benggala, Taman Margasatwa Medan, Toxocara cati, Ancylostoma tubaeformae. ABSTRACTThis study aims to determine whether Sumatran tigers (Panthera tigris sumatrae) and Bengal tigers (Panthera tigris tigris) in Medan Wildlife Park are infected with gastrointestinal parasitic worms and what are the types of parasitic worms. The samples used in this study were faeces from 9 Sumatran tigers and Bengal Bengal as many as 6 in Medan zoo. Samples were taken three times at different times and at intervals of 2 weeks. Samples were taken in the morning and put into bottle samples and added with equal volume of 10% formalin. Samples were transported using ice boxes that were filled with ice to the Laboratory of Parasitology, Faculty of Veterinary Medicine, Syiah Kuala University, to be examined. Sample examination using centrifugation, McMaster, and Boray sedimentation methods. The results of this study indicate that there are two nematode worms infesting Sumatran tigers (Panthera tigris sumatrae) and Bengal tigers (Panthera tigris tigris) in Medan zoo namely Toxocara cati and Ancylostoma tubaeformae. Keyword: Panthera tigris sumatrae, Panthera tigris tigris, Medan Wildlife Park, Toxocara cati, Ancylostoma tubaeformae.

Abstract

Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri asam laktat (BAL) genus Lactobacillus yang terdapat pada feses orangutan Sumatera (Pongo abelii) di Kebun Binatang Kasang Kulim Bangkinang, Riau. Sampel yang digunakan adalah feses segar dari empat ekor orangutan Sumatera (Pongo abelii). Bakteri ini diidentifikasi dengan metode Carter dan Cole yang dimodifikasi. Media selektif yang digunakan yaitu de Man Rogosa Sharpe Agar (MRSA) dengan metode streak plate. Koloni yang tumbuh di media MRSA diamati morfologinya dan dilakukan pewarnaan Gram serta dilanjutkan dengan uji biokimia yaitu uji katalase, oksidase, Voges Proskauer (VP), Sulfit Indol Motility (SIM), Triple Sugar Iron Agar (TSIA), oksidatif/fermentatif (O/F), dan uji gula-gula yaitu glukosa, laktosa, mannitol, maltosa. Hasil identifikasi terhadap empat sampel feses orangutan ditemukan genus Lactobacillus pada tiga sampel orangutan. Oleh sebab itu, dapat disimpulkan bahwa bakteri asam laktat genus Lactobacillus dapat diidentifikasi pada feses orangutan Sumatera (Pongo abelii) di Kebun Binatang Kasang Kulim Bangkinang Riau.
