Abstract

The process of freeze drying caused detrimental effect on plasma membrane and acrosome of the spermatozoa, even it potentially could alter the chromatin and DNA integrities. On the other hand, DNA integrity is essential for spermatozoa to participate in pronucleus formation during fertilization event. Therefore the evaluation of DNA integrity should be carried out to study the effect of freeze drying process. EDTA, EGTA, and PBS were used as dilution media of spermatozoa prior to freeze drying process to protect the DNA. Toluidine blue staining and comet assay methods were used to evaluate the alteration on chromatin and DNA integrities of spermatozoa, respectively. The results revealed that the highest compacted chromatin after 6 months storage of freeze-dried spermatozoa were observed from EGTA-3 (98%) and EGTA-1 (97%) treatments that had significant differences compared to all PBS treatments (90-92%), but not for fresh spermatozoa (100%). Whereas, the highest compacted DNA integrity of freeze-dried spermatozoa were observed from EGTA-2 (92%) and EGTA-3 (92%) but had no significant differences compared to other treatments including fresh spermatozoa (97%). These results demonstrate that EDTA and EGTA tend to be able to protect chromatin and DNA integrities of ram spermatozoa during freeze-drying and storage compared to PBS. Abstract

One of the factorthat influencethe process offertilization on Echymiperakalubu is the quality of the spermwhich produced by males. The ability of spermtofertilize anovumis not only owned by the spermatozoa which derived from the results of ejaculation, butis also owned by spermatozoa from the caudaepididymis.This study aimed to determine sperm concentration and motility from caudaepididymis.Twenty-one males E. kalubuwere used which was 370-1500g. Based onthis weight, it was divided into three groupings, they were prepubertal (370-640 g), young adult (>640-850g) and adult (>850g).The results showed that the sperm concentration of the prepubertal was1.75±1.14x106/ml,the young adult was 8.86±4.21x106/ml and increased to12.56±6.33x106/mlin the adult group.Sperm motility also increased with increasing maturity. The prepuberta lgroup showed that sperm motility was33.33±25.17%, increased to41.67±12.58% (young adult) and3.54±57.50% followed by the adult.

Abstract

The aim of this study was to evaluate the concentration, progressively motile, and percent live sperm in the various regions of epididymis and ductus deferens after preservation at 4oC. Epididymis and ductus deferens were collected from 21 epididymis and ductus deferens of domesticated cat (Felis catus) by castration. One testicle of pair (control testicle) was analyzed at the day of castration, while the other testicle of the pair was stored at 4oC up to 7 days. The sperm concentration, percentage of sperm motility and live sperm were examined daily until day-7 of preservation. The sperm concentration was higher (p<0.05) in cauda epididymis (23.99x106 sperm/ml) and ductus deferens (25.42x106 sperm/ml) than caput (11.51x106 sperm/ml) and corpus epididymis (14.82x106 sperm/ml). The percentage of sperm motility and live sperm decreased (p<0.05) during preservation period. However, the percentage of motile (11.33 to 16.00 %) and live (15.05 to 20.20 %) sperm could be found in preserved epididymis and ductus deferens up to day-7. These results show that motile and live sperm can be collected from cat’s epididymis and ductus deferens up to day 7 after preservation at 4oC.

Abstract

Since the discovery of cryopreservation method for bull semen, cryopreservation become an alternative method for maintaining gamet resources of certain animal which is threatened or near extinction. This technology was then applied to the preservation of embryo, oocyte, ovary and testis. The application of intracytoplasmic sperm injection (ICSI) for which sperm motility is unnecessary had supported the effort to create simplified method such as freeze-drying for sperm preservation. Due to the benefit of ICSI over the conventional in vitro fertilization (IVF) the spermatozoon could be mechanically driven to pass through the zona pellucida and entering the cytoplasm of oocytes prior to fertilization. The freeze-drying method is an alternative method in sperm preservation which ignored the motility of sperm. The sperm resulted from this technique is in drying state, therefore, it might be stored in room temperature or in refrigerator. Many reports have claimed that freeze-dried sperm which is not motile but has an intact DNA was able to fertilize oocytes, even produced offspring in mouse.

Abstract

Experiments were conducted on the morphology, fertilization and embryo development rate of vitrified ovine oocytes matured in vitro. Three vitrification solutions were used for vitrification. PBS supplemented with 1% BSA, 30% ethylene glycol was added by one of three different sucrose concentrations, 1.00 M (VS1), 0.50 M (VS2), and 0.25 M (VS3). The results showed that the percentages of normal vitrified oocytes after warming were 78 and 63% in VS1 and VS2, respectively, which was significantly higher as compared for VS3. The fertilization rates were 59 and 66% in VS1 and VS2, respectively, which were also significantly higher as compared with VS3 (35%). Zygote viability after 18 h was 57; 43; and 40%, for VS1,VS2, and VS3, respectively, which was not significantly different. The incidence of polyspermic penetration increased with increasing sucrose concentration, i.e 23, 11, and 9% in VS1, VS2, and VS3, respectively, as compared with unvitrified oocytes (4%). One therapy for cancer diseases is chemotherapy and radiation, that give a side effect of infertility in the gonad, therefore, it is necessary to preserve the gonad. Abstract

Penelitian ini bertujuan untuk mempelajari morfologi kelenjar aksesori muncak jantan secara makroanatomi dan mikroanatomi. Seekor muncak jantan dewasa berumur 4-5 tahun dengan bobot badan 19 kg digunakan pada penelitian ini. Muncak terlebih dahulu di-exanguinasi untuk dikoleksi kelenjar aksesori kelaminnya. Untuk memperoleh gambaran mikroanatomi, sampel kelenjar aksesori diproses dengan teknik histologi dan diwarnai dengan pewarnaan hematoksilin-eosin (HE). Hasil pengamatan makroskopis menunjukkan bahwa kelenjar aksesori muncak jantan terdiri atas ampula, duktus deferens, kelenjar prostat, kelenjar vesikularis, dan kelenjar bulbouretralis. Karakteristik histologi kelenjar aksesori muncak adalah ditemukannya kelenjar prostat yang berbentuk pars diseminata dengan kelenjar-kelenjar sekretori tersebar di sekeliling lumen uretra pars pelvina dimana secara makroskopis kelenjar tersebut tidak dapat diamati. Tipe kelenjar sekresi pada ampula, kelenjar vesikularis, dan pars diseminata prostat adalah tubuloalveolar, sedangkan pada kelenjar bulbouretralis tipe tubular. Dapat disimpulkan bahwa morfologi kelenjar aksesori muncak jantan memperlihatkan kemiripan dengan kelenjar aksesori pada ruminansia kecil lainnya seperti kambing, domba, reeves muntjak, dan pampas deer.

Abstract

Carbohydrate is a diverse compound in the body and can be  as glycoconjugate which bound to protein in proteoglycan and glycoprotein shape, and bound to lipid in glycolipid shape as well. The study was conducted to get information image of pattern distribution of the glycoprotein at stomach gland from various age levels at Macaca fascicularis (Mf) after and before bearing. Gastric specimen was collected processed through microtechnique procedure, followed by histochemistry Lectin staining to detect glycoprotein from stomach. Observation result of given score subjectively into four categories: (-) none/negatif, (+) low, (++) medium,and (+++) height. The results obtained were tabulated and analyzed diskriptively. We found that glycoproteins complex exist in the glandullar stomach of long tailed macaques are mannose, glucose, galactose, N-acetil-D-glocosamine and N-acetil-D-galactose all of these glycoproteins complex was observed at age foetus 70 days. Abstract

Tujuan penelitian ini adalah untuk menjelaskan pengaruh pemberian ekstrak hipofisis ikan mas terhadap pematangan gonad ikan sidat tropis betina (Anguilla bicolor bicolor). Ikan sidat dengan bobot 600 gram telah digunakan sebagai materi biologis. Ekstrak hipofisis ikan mas dengan tingkat 0, ¼ dan ½ dosis dan tiga tipe frekuensi penyuntikan yakni setiap 1, 2 dan 3 minggu dijadikan sebagai perlakuan. Parameter yang diamati adalah Gonado somatic index (GSI), Hepatosomatic index (HSI), diameter folikel, kadar lemak dalam folikel, serta kadar glikokonjugat dalam folikel dan hati. Hasil percobaan menunjukkan bahwa penyuntikan ekstrak hipofisis ikan mas dengan ½ dosis yang dilakukan setiap minggu selama 10 minggu memberikan pengaruh yang terbaik.Kata kunci: gonad, sidat, Anguilla bicolor bicolor.

Abstract

Bats have the second largest diversity in mammalian classes after rodentia (Nurfitrianto et al., 2013). Bat population reaches approximately 1100 species (Teeling et.al 2005). In Indonesia there are 205 or 21% of known world bat species (Suyanto 2001). Bats are the only flying mammals. The ability bat to fly is supported by the shape of the extremities and also the adaptation of the cardiovascular organs. Based on empirical studies of cardiovascular was known that cardiac muscle mass is a good indicator for the degree of adaptive specialization of prolonged locomotor activity, in both birds and mammals. Data on cardiovascular muscle morphology and muscles that support flying ability will provide an overview of the behavior, ecology and physiology of certain animals (Bishop 1997).Several studies have been conducted to study the cardiovascular system of bats. Research on bat hematology profile has been done (Heard and Whittier 1997, Mclaughlin et.al 2007, Wawrocka and Bartonicka 2014, McMichael et al. 2015, Ratnasooriya 2016, Rashid et al, 2016, Rahma et al., 2018). Joseph (1908) says that every animal has a different size and heart shape to each other. This heart size difference is influenced by the size of the body and the high energy required by the animal (Joseph 1908).Bat's heart rate is known to increase during flight (Thomas and Suthers 1972). This is causes an increase in the rate of metabolism. Writing this paper will discuss about the uniqueness of the bat's heart so that it can support its ability to fly.
