Abstract

During a visit to Bali Island in September 2011, we found chilipepper plants exhibiting yellowing symptoms. The infected plants showed quite similar symptom to those of the recently reported Pepper yellow leaf curl virus from Israel and Japan, but there was no shortening of internodes or leaf rolling. We have amplified part of the genome of a virus associated with this disease using degenerate primers for members of the genus Polerovirus. The reverse transcription-polymerase chain reaction (RT-PCR) from symptom-bearing samples resulted in the amplification of a 650 bp band which is the expected size. RT-PCR from healthy samples did not produce an amplicon. Based on our results, we concluded that yellowing disease on chilipepper observed in Payangan, Gianyar, Bali is associated with a virus belonging to the genus Polerovirus. This is the first report on Polerovirus infection in chilipepper in Indonesia. Some researches for elucidation of the virus species and biological/molecular characters are being conducted.Key word: chilipepper, Indonesia, Polerovirus

Abstract

Budok caused by Synchytrium pogostemonis is the important and destructive disease on patchouli plant (Pogostemon cablin). Chemical control must be done in incidence with high disease intensity and prediction of outbreak. The experiment was done using benomyl (Benlate 50 WP) and cuprous oxide (Kocide 77WP) 0.1%, 0.2% and mixture of both ( 1 g cuprous oxide and 1 g L-1 benomyl). Patchouli plant was sprayed eight times with one week interval. Harvesting was done two weeks after the end of spraying. Residue analysis of Cu was done with AAS and benomyl residue with HPLC. The result showed that 0.1% benomyl significantly reduced the disease intensity. The other treatments showed lower result. Harvested patchouli plants treated with 0.1% benomyl showed higher yield compare with 0.2% benomyl and cuprous oxyde. Cu residue in dried leaves was between 460 ppm and 950 ppm. Residue of benomyl was 54–100 ppb. Diseased seedling still can be used after spraying by benomyl four times with 2 weeks interval.

Abstract

Tobamovirus is a group of virus with a wide host range, including orchid plant which considered as an economically important plant. This research aimed to identify Tobamovirus infecting orchids. Virus isolates were collected from orchid nursery in Sleman, Yogyakarta. Plant extract from orchid showing necrotic flex symptom was inoculated to indicator plants Chenopodium amaranticolor. Chlorotic local lesion symptoms occurred within 3 days after inoculation. RNA total from symptomatic C. amaranticolor was extracted by using a commercial kit. cDNA was synthesized using oligo d(T) primer. Amplification of cDNA using partial movement protein specific primers TMV-1F and TMV-2R was successfully amplified the amplicon with size ± 422 bp. The nucleotide sequences of this amplicon  showed highest DNA homology (98%) with Tobacco mosaic virus Yongren-2 isolat from China.

Abstract

Soybean (Glycine max) is an important  food crops in Indonesia, but the production is considered low. Infection of Cowpea mild mottle virus (CPMMV) may affect to the productivity of the crops.  The virus cause a variety of symptoms on soybean such as mosaic, chlorosis, necrosis, distortion, and leaf dwarf. The aims of this study were to detect and identify CPMMV by molecular technique from some varieties of soybeans, i.e.  ‘Anjasmoro’, ‘Argomulyo’, ‘Grobogan’, and two local Edamame varieties. Symptomatic samples was obtained randomly from main soybean growing areas in Java.  DNA amplification using specific primer for coat protein of CPMMV was only successfully gained from leaf samples of var. Argomulyo from Malang which showing mosaic on the leaf surface, vein clearing, and chlorosis. Further amino acid and nucleotide sequence analysis showed that this CPMMV isolate has highest homology (99%) to CPMMV isolate of French bean from China.

Abstract

Cucumber mosaic virus on Black Pepper in Yogyakarta and Bangka BelitungPepper  (Piper  nigrum)  is  spice  crop  which  has  been  cultivated  a  long  time  ago  in  Indonesia. Stunting is one of disease on pepper caused by cucumber mosaic virus (CMV). The research aimed to diagnose the biological, morphological and nucleaic acid characters of CMV on pepper in Yogyakarta and Bangka Belitung. CMV infection on pepper in both area (Putat dan Kleben village, Yogyakarta and Air Buluh village, Bangka Belitung) showed typical symptoms such as mosaic, narrow leaves and stunting. The disease incidence and disease severity of stunting disease are varies. The virus able to transmitted by cutting, grafting and mechanically on Nicotiana tabacum and Chenopodium amaranticolor. However, it was unable to transmitted mechanically on pepper and by Aphis gossypii. The virus particles were isometric with diameter size 28-30 nm. RT-PCR using coat protein partial gene primer successfully amplified a DNA with size ± 500 bp from all three samples. The homology of nucleotide between three isolates was 98-97%, while the highest homology of those three strains CMV  from Yogyakarta and Bangka Belitung was 98% against strains from China in Brassica chinensis. Three strains CMV from pepper were in the same group, and separated from CMV pepper lines from Indonesia and other CMV isolates.

Abstract

Gejala mosaik kuning dan keriting daun ditemukan pada pertanaman kacang panjang di Sleman, Yogyakarta. Begomovirus diketahui sebagai salah satu penyebab penyakit tersebut. Penelitian dilakukan untuk mengidentifikasi spesies Begomovirus dan DNA satelit yang berasosiasi dengan penyakit mosaik kuning kacang panjang. Ekstraksi DNA total dari tanaman bergejala dilanjutkan dengan amplifikasi fragmen DNA spesifik Begomovirus dan Betasatelit. Amplikon DNA berukuran ±1500 pb dan ±1300 pb berhasil diperoleh menggunakan berturut-turut primer universal Begomovirus dan primer spesifik Betasatelit. Analisis sekuen nukleotida mengonfirmasi identitas Begomovirus yang menginfeksi tanaman kacang panjang ialah Mungbean yellow mosaic India virus (MYMIV) dengan homologi 99% terhadap isolat MYMIV asal Indonesia. DNA satelit yang berasosiasi dengan MYMIV menunjukkan karakteristik Betasatelit, yaitu memiliki satellite common region (SCR) dengan struktur stem-loop dan sekuen TAATATTAC pada bagian loop, adenine rich region sebesar 54.96%, dan ORF (open reading frame) non-coding. Lebih lanjut, analisis rekombinasi menggunakan SimPlot mengindikasikan bahwa satelit non-coding MYMIV merupakan satelit rekombinan antara Betasatelit dan DNA-B Pepper yellow leaf curl Indonesia virus (PepYLCIV). Artikel ini merupakan laporan pertama asosiasi betasatelit DNA non-coding dengan MYMIV di Indonesia.

Abstract

Salah satu kendala utama dalam budi daya tanaman stroberi di Indonesia adalah banyaknya penyakit tanaman. Penelitian ini merupakan lanjutan dari laporan tentang penyakit penting pada buah stroberi (Fragaria ananassa) di Kabupaten Purbalingga, Jawa Tengah untuk mengidentifikasi penyakit oleh cendawan pada bagian tanaman selain buah. Sampel berupa daun atau tanaman bergejala, diidentifikasi secara morfologi atau molekuler apabila identifikasi morfologi tidak memungkinkan. Konfirmasi gejala penyakit yang disebabkan oleh cendawan dilakukan dengan mengikuti postulat Koch, yaitu menginokulasikan cendawan patogen hasil isolasi pada tanaman stroberi sehat. Hasil penelitian menunjukkan bahwa penyakit penting yang disebabkan oleh cendawan pada tanaman stroberi adalah penyakit hawar daun yang disebabkan oleh Phomopsis sp. dan Pestalotia sp.; penyakit gosong daun yang disebabkan oleh Curvularia sp. dan Diplocarpon sp.; penyakit bercak daun yang disebabkan oleh Cercospora sp., Phoma sp., dan Gnomonia sp.; serta penyakit layu yang disebabkan oleh Verticillium sp. dan Fusarium sp. Selain laporan tentang penyakit penting tanaman stroberi, ini juga merupakan laporan pertama tentang penyakit bercak Gnomonia pada tanaman stroberi di Indonesia.

Abstract

Penyakit daun keriting kuning pada pertanaman cabai di Sulawesi Tenggara telah dilaporkan sejak tahun 2018, yaitu di Kabupaten Kolaka Timur dan Kota Kendari. Gejala penyakit di lapangan semakin meluas seiring dengan perluasan penanaman cabai. Penelitian bertujuan menghitung kembali insidensi penyakit daun keriting kuning pada pertanaman cabai di Sulawesi Tenggara, mengidentifikasi serangga yang berasosiasi dengan tanaman cabai, dan mengidentifikasi penyebab penyakitnya. Pengamatan insidensi penyakit dilakukan di pertanaman cabai yang berada di Kabupaten Bombana, Konawe Selatan, Konawe, Kendari, Kolaka Timur, Kolaka, dan Kolaka Utara. Identifikasi kutukebul dilakukan berdasarkan karakter morfologi. Deteksi dan identifikasi begomovirus menggunakan metode polymerase chain reaction, yang dilanjutkan dengan analisis sikuensing. Rata-rata insidensi penyakit daun keriting kuning di tujuh kabupaten ialah 36%–90%. Spesies kutukebul yang ditemukan adalah Aleurotrachelus trachoides. Fragmen DNA spesifik begomovirus berukuran 580 pb berhasil diamplifikasi dari sampel tanaman cabai asal tujuh kabupaten di Sulawesi Tenggara. Analisis sikuen mengonfirmasi infeksi Pepper yellow leaf curl Indonesia virus pada pertanaman cabai di Kabupaten Kolaka, Kolaka Utara, Bombana, Konawe, dan Konawe Selatan.
