Abstract

Smooth Cayenne is the largest pineapple type cultivated in Indonesia, but its vegetative planting materials for mass propagation are limited. Somatic embryogenesis is a potential method to be applied. The aim of this study was to investigate the somatic embryogenesis regeneration under the effect of picloram and light. Callus formation was induced by picloram (21, 41 and 62 μM) added with 9 μM thidiazuron. The calli were transferred onto MS or Bac medium  enriched with N-organic compounds with or without addition of 21 μM picloram under dark or light condition. The compact calli were subcultured onto MS medium supplemented with 4.65 μM kinetin, while the friable calli were  transferred onto BIG medium (modified MS + 1.1 μM benzyl adenine + 0.9 μM indole butyric acid + 0.09 μM giberelic acid) or B medium (MS + 0.018 mM benzyl adenine). The results showed that the events of somatic embryogenesis were started from cell polarization, asymmetrical division, proembryo formation as  embryogenic tissues and friable embryogenic tissues, and embryo development. The best treatment for callus induction was 21 μM picloram. The addition of 21 μM picloram on N-organic enriched medium and the use of light condition  proliferated embryogenic calli. The N-organic enriched Bac medium and light condition yielded the highest number of mature somatic embryos (17 embryos perexplant in 2 months). The B medium was better than BIG medium to develop  somatic embryos from friable embryogenic tissues. The somatic embryogenesis method presented is potential for pineapple mass propagation and artificial seedproduction.Abstrak Bahasa IndonesiaSmooth Cayenne merupakan kultivar nenas yang banyak dibudidayakan di  Indonesia, namun ketersediaan benih untuk perbanyakan massal masih terbatas. Embriogenesis somatikadalah metode yang potensial untuk produksi bibit secara massal. Tujuan penelitian adalah untuk mempelajari pengaruh pikloram dan pencahayaan terhadap regenerasi embriogenesis somatik nenas. Kalus diinduksi menggunakan pikloram (21, 41, dan 62 μM) dan penambahan thidiazuron 9 μM. Selanjutnya, kalus dipindahkan ke media MS atau Bac yang diperkaya dengansenyawa N-organik dengan atau tanpa penambahan pikloram 21 μM dalam kondisi gelap atau dengan pencahayaan. Kalus kompak disubkultur pada media MS yang mengandung kinetin 4,65 μM, sedangkan kalus remah dipindahkan ke media BIG (MS modifikasi + bensil adenin 1.1 μM + indole butyric acid 0,9 μM + giberelic acid 0,09 μM) atau media B (MS + bensil adenin 0,018 μM). Hasil penelitian  menunjukkan bahwa tahapan embriogenesis somatik diawali dengan polarisasi sel, pembelahan asimetris, pembentukan proembrio sebagai jaringan embriogenik danjaringan embriogenik remah, serta perkembangan embrio. Perlakuan terbaik untuk induksi kalus adalah pikloram 21 μM. Penambahan pikloram 21 μM pada media yang diperkaya dengan senyawa N-organik mampu meningkatkan jumlah kalusembriogenik. Media Bac yang diperkaya senyawa N-organik dan kondisi pencahayaan menghasilkan jumlah embrio somatik dewasa terbanyak (17 embrio per eksplan dalam 2 bulan). Media B lebih baik daripada media BIG untuk regenerasi embrio somatik dari jaringan embriogenik remah. Metode embriogenesis somatik yang dihasilkan dari penelitian ini berpotensi diterapkan untukperbanyakan massal dan produksi benih nenas.

Abstract

Plant genetic improvement can be done through classical breeding or genetic engineering. WRKY is a transcription factor involved in regulating plant defense responses. OsWRKY76 gene is located in a narrow segment of chromosome 9 which is identified previously to be related to wide spectrum resistance in rice. A sequence of OsWRKY76 (+1.200 bp) has available in the gene bank and it makes possible to isolate, clone, and construct the gene into over-expression vector. The aim of this research was to assemble an over-expression construct of OsWRKY76 candidate gene and introduce it into rice through Agrobacterium-mediated transformation. A construct of pCAMBIA-1301::35S::OsWRKY76 has been successfully assembled and transformed into embryogenic calli of rice cv. Nipponbare using A. tumefaciens strain Agl-1 and EHA 105. A number of 126 independent lines has been produced, in which Agl-1 showed 3.8 times more efficient than EHA 105. PCR analysis of randomly selected 25 independent lines showed that all of them positively contained hptII gene, a selectable marker used in the over-expression construct of the OsWRKY76 candidate gene. Based on the result, it could be concluded that the over-expression construct of OsWRKY76 candidate gene have been successfully introduced into the tissue of Nipponbare.

Abstract

Indonesia has high variety of pummelo [Citrus maxima (Burm.) Merr.], but only several cultivar are cultivated extensively. Others became vurnerable and the  germplasm has to be conserved to prevent their extinction. One of the conservation method is in vitro conservation  using a slow growth technique. Factorial experiment was used in designing the experiment, the first factor was MS medium concentration, i.e. 1/2MS and MS. The second factor was sucrose concentration, i.e. 0; 1; 2; and 3%. The results showed that low concentration of MS medium and sucrose reducing the leaf number and shoot length but increasing the root number and length.  Based on inhibition of growth,  the most reducing growth was planlet on MS without sucrose.

Abstract

 The study was carried out for investigation of the adaptability of white jabon seedlings from 12 populations to drought and water logging stresses in a controlled green house. The results showed that the adaptive responses of white jabon seedling to drought and waterlogging stresses were affected by genotype (population). The drought and waterlogging stresses significantly inhibited plant growth, biomass accumulation and allocation, leaf area, also decreased chlorophyll content, increased carotenoids contents, and accumulated free proline. Relative water content and specific leaf area tended to be higher in waterlogging and declined in drought stresses.  The result clearly indicated that white jabon seedlings were more adaptive to waterlogging than to drought stresses. Moreover, there were different responses to drought and waterlogging stresses between the twelve populations. Kampar, Gowa, Kuala Kencana and OKI populations exhibited higher growth performance and stress tolerance index to be adapted to waterlogging stress, while Gowa, Pomalaa and Kampar populations had relatively better growth performance in the drought stress. Keywords: Anthocephalus cadamba, genotype, growth, population, stress tolerance index

ABSTRACTIndirect Organogenesis and Somatic Embryogenesis ofPineapple Induced by Dichlorophenoxy Acetic Acid. IkaRoostika, Ika Mariska, Nurul Khumaida, and Gustaaf A.Wattimena. This research aimed to study the effect of 2,4-D,AdS, and basal media to the regeneration of pineapplethrough indirect organogenesis and somatic embryogenesis,and to study the complete event of somatic embryogenesis.Callus formation was induced by 21, 41, and 62 μM 2,4-Dwith addition of 9 μM TDZ. The non embryogenic calli weretransferred onto 4.65 μM Kn containing medium.Embryogenic callus formation was induced on MS or Bacbasal media consisted of N-organic compounds withaddition of AdS (0, 0.05 and 0.1 μM). The embryogenic calliwere regenerated on modified MS medium with addition of0.9 μM IBA, 1.1 μM BA, 0.09 μM GA3 or MS mediumsupplemented with 0.018 mM BA. The result proved that thesingle auxin of 2,4-D was not enough to induce embryogeniccells. Therefore the non embryogenic calli were regeneratedthrough organogenesis. The 21 μM 2,4-D yielded high level ofcallus formation (80%), higher fresh weight (0.2 g/explant)and higher number of shoot (25 shoots/explant in twomonths). Embryogenic calli were produced on N-organiccompounds enriched media. The regeneration mediumsignificantly affected the level of browning, where the MSmedium with addition of 0.018 mM BA yielded lower level ofbrowning. There was an interaction of embryogenic callusinduction medium and regeneration medium to the numberof mature somatic embryos. The embryogenic callusinduction on MS medium enriched with N-organiccompounds and 0.05 μM AdS followed by the regenerationof somatic embryos on MS medium with addition of 0.018mM BA was the best treatment which yielded 17 maturesomatic embryos/explant

Abstract

Tujuan dari penelitian ini adalah mengembangkan model biomekanika untuk aktivitas mendorong manual berdasarkan kemampuan gaya dorong tubuh. Model yang dikembangkan merupakan model statik yang dianalisis dalam pandangan 3 dimensi. Proses pemodelan dimulai dengan penentuan koordinat persendian dan koordinat titik pusat massa segmen. Langkah selanjutnya adalah menentukan formulasi model kapasitas gaya dorong tubuh berdasarkan keseimbangan momen di sendi L5/S1, dengan data input massa tubuh, tinggi tubuh, dan sudut antar segmen. Meskipun analisis dilakukan secara statik dalam pandangan 3 dimensi, tetapi model ini cukup memberikan informasi berkaitan dengan kapasitas gaya dorong tubuh pada posisi pendorongan tertentu. Model tersebut telah divalidasi secara internal dengan menjelaskan persamaan-persamaan kinetika yang dibuat secara logis. Hasilnya model dapat disimpulkan valid, karena benar dalam penentuan persamaan kinetikanya

Abstract

AbstrakPerkembangan internet dan perkembangan teknologi gadget yang cukup pesat telah merevolusi proses jual beli akhir-akhir ini. Dikarenakan kondisi jalan yang semakin macet, dan kesibukan orang-orang dalam bekerja sehingga fenomena jual beli melalui toko online menjadi semakin marak. Melihat paradigma tersebut,  maka kemampuan membuat toko online akan menjadi nilai tambah bagi setiap orang yang ingin memulai usaha tapi belum bisa memiliki toko secara fisik. Disamping itu, kemampuan membuat toko online itu dapat menjadi modal untuk ditawarkan kepada pemilik toko fisik agar bisa juga memanfaatkan toko online untuk meningkatkan pangsa pasar mereka. Permasalahan yang terdapat di SMK Pasundan adalah siswa nya belum memiliki keterampilan dalam pembuatan toko online,  sehingga Universitas Esa Unggul melalui kegiatan pengabdian masyarakat memberikan Penyuluhan Teknologi Informasi, Internet dan Pelatihan Aplikasi Komputer untuk Perkantoran bagi guru-guru dan siswa-siswi SMK Pasundan Tangerang. Agar para siswa SMK Pasundan Tangerang yang terdiri  dari jurusan Teknologi Komputer dan Jaringan (TKJ) serta jurusan Administrasi Perkantoran (Adm-Bis) ini dapat memiliki keterampilan dalam pembuatan toko online, maka materi pelatihan yang diberikan adalah tentang pembuatan toko online dengan aplikasi OpenCart. Dengan diadakannya pelatihan ini, para siswa-siswi SMK Pasundan Tangerang memiliki antusias yang cukup tinggi dengan pelatihan ini memiliki keterampilan dan wawasan dalam pembuatan toko online serta memiliki minat untuk mulai berwirausaha. Lebih jauh para siswa-siswi SMK Pasundan ini bisa menjual keterampilannya kepada orang-orang yang ingin memulai usaha secara online. Kemampuan yang diperoleh oleh siswa SMK Pasundan Tangerang ini dirasakan sangat bermanfaat dan mereka sangat berharap untuk dikembangkan lebih lanjut untuk dikembangkan lebih lanjut. Hasil dari pelatihan pembuatan toko online ini masih dalam bentuk prototyping dan masih perlu tindak lanjut pengembangan pelatihan agar ditindaklanjuti dengan pelatihan tambahan dari sisi marketing dan pengetahuan produk. Kata Kunci : e-Commerce,toko online,OpenCart.

Abstract

Universitas Terbuka (UT) sebagai perguruan tinggi yang menerapkan sistem belajar jarak jauh menyusun kurikulum program pendidikannya berbeda dengan perguruan tinggi yang mempunyai sistem belajar tatap muka. Mahasiswa UT diberikan kebebasan dalam memilih dan menentukan mata kuliah untuk diregistrasikan pada satu semester. Dalam kurikulum program studi di UT terlihat bahwa terdapat mata kuliah yang mempunyai hubungan keterkaitan materi antarmata kuliah. Hubungan tersebut digambarkan sebagai hubungan prasyarat antarmata kuliah. Untuk membuat rencana studi, mahasiswa perlu mengetahui hubungan keterkaitan materi antarmata kuliah tersebut sehingga dapat menentukan mata kuliah yang akan diregistrasi pada suatu semester. Artikel ini mengidentifikasi keterkaitan materi antarmata kuliah untuk dipetakan dalam rencana studi. Untuk memetakan mata kuliah berprasyarat tersebut perlu dilakukan optimasi pemetaan mata kuliah. Tujuan pengembangan model optimasi ini adalah untuk memaksimalkan hubungan keterkaitan materi dengan memetakan mata kuliah berprasyarat secara berurutan. Hal ini juga berarti meminimalkan jarak antarsemester urutan mata kuliah berprasyarat. Model optimasi yang dikembangkan berupa Integer Linear Programming. Model tersebut kemudian diimplementasikan pada kurikulum Program Studi Matematika FMIPA UT. Hasil simulasinya berupa rencana studi bagi mahasiswa Program Studi tersebut. Dari analisis hasil simulasi diperoleh rencana studi dengan jumlah maksimal 21 dan 23 SKS per semester merupakan pilihan terbaik bagi mahasiswa.   Universitas Terbuka (UT) as an open and distance learning institution has curriculum that different from face to face universities. UTs students can take any course since UT offers all courses in any semester. Some courses are prerequisite for some other courses. Students need to be careful in taking the courses because some courses also have same schedule in examination. Besides, they also need to consider the courses that have prerequisite. This article identifies the relationship between some courses that need to be considered by the students when they register the courses. A map of courses was developed by using Integer Linear Programming. This map was intended to maximize the information of interrelationship between courses in rows. The distance between prerequisite courses was minimized for some semesters. This model was implemented on Mathematics study program at Faculty of Mathematics and Science in UT. The result of simulation was a study planning for Mathematics students in taking courses in every semester. The optimal number of credits semester that need to be registered was 21 or 23 credits.

Abstract

Universitas Terbuka (UT) was implementing the centralized distribution of learning material, it is not considered efficient. Therefore, the decentralized distribution system which requires warehouses at certain regional offices will be proposed. The aims of this research are (1) investigating the decentralized learning material distribution model, (2) choosing the optimal location of warehouses to minimize the cost of learning material distribution, (3) choosing the regional offices which will be served optimally by each warehouse, (4) comparing efficiency of the centralized and decentralized learning material distribution system. The distribution problem is modeled as a linear mixed integer programming problem. The model will be solved using Branch and Bound method. This study considers centralized learning material distribution system and two alternative of decentralized learning material distribution systems, which are alternative 1 and alternative 2. It is found that in 2008 the cost of the alternative 2 is lower than the centralized learning distribution system. Besides, if the demand of learning material is increasing, the alternative 2 is still less costly than the other systems.
