Abstract

  Flavr Savr tomato is a transgenic tomato which is transformed using Polyglacturonase gene in antisense orientation. This is the first whole food product of biotechnology that reachs the market, but unfortunately it does not succeed.   Ethylene is thought to act as a natural triggering mechanism for fruit ripening and senescence.  Lowering the production of endogenous ethylene from fruit should delay fruit ripening and senescence. Several ways to control ethylene biosynthesis are: inactivation of the gene encoding ACC synthase, ACC oxidase, metabolism of SAM so that ACC is not produced, or metabolism of ACC before it can be converted to ethylene.  Effect of ethylene can also be blocked by blocking the perception of ethylene by specific tissues. Abstract

ABSTRACTResistance to yellow virus (YV) is an important breeding trait in melon. However information regarding genetic inheritance pattern of the character are limited. This study aimed to provide information on genetic control for resistance to YV in melon caused by Tomato Yellow Leaf Curl Virus (TYLCV). Twenty genotypes from three major melon groups (dudaim, cantaloupe, and inodorous) were evaluated using controlled inoculation method. The results revealed that one line, MEV1 from the dudaim group, showed high resistance to YV; while other lines belong to cantaloupe and indororus indicated as highly susceptible lines. Screening of the F1 from crossing between resistant and susceptible parents resulted in resistant F1 after inoculation and planted in endemic location. Subsequently, evaluation on F2 population revealed a non-normal distribution for disease severity score, indicating that resistance to YV in melon was controlled by major genes. Chi-square (χ2) test resulted in 13:3 ratio and indicated that the resistance to YV was controlled by 2 genes pair with dominant and epistasis recessive actions.Keywords: dominant and epstasis recessive action, major gene, Bemicia tabaci, Begomovirus

Abstract

Skinning injury in sweetpotato due to loss of skin or periderm which occurred during harvest is inevitable and account for financial loss due to dehydration, pests, and pathogens. Hence, studies on gene expression changed due to skinning injury can provide important information about this protective tissue and for improving the life of storage roots. New candidate genes involved in skinning injury were isolated with an Annealing Control Primer (ACP). Using 20 ACP primers, a total of 103 differentially expressed genes (DEGs) were retrieved. In this study, the functional annotation of these selected 15 up-regulated DEGs (10 contigs and 5 singletons) were characterized. The results showed that these 15 “DEG-unigenes” are mainly associated with defense and stress responses, regulation and signaling, protein synthesis and fate, and metabolism may play an important role in the primary responses to skinning injury in storage roots of sweetpotato. This study showed the importance of defense and stress responses genes to the formation of wound periderm. Furthermore, this results can be used for better understanding of the molecular mechanism of skinning/mechanical injury-related genes in the storage roots of sweetpotato as well as to all stems, fruits, and roots of all plants. Abstract

Gamboge is the main problem in mangosteen agribusiness because it is one of the major factors lowering fruit quality. Calcium is one of the important elements that strengthening cell wall; it is which was binding with the pectin as a middle lamella component. The objectives of the research were to study the effect of calcium application on the presence of gamboge spots, physical, and chemical properties of mangosteen fruit. Trial was conducted in a randomized complete block design with three replications in two consecutive years. The calcium source was dolomite, applied in four different dosages, i.e. 0, 18, 24, and 34 ton ha-1 for the fi rst year and 0, 12.5, 15, and 17.5 ton ha-1 for the second year. The results showed that calcium application raised soil pH and calcium content of the soil, exocarp and mangosteen leaves. Dolomite applications using 18 and 24 ton ha-1 in the fi rst year and 17.5 ton ha-1 in the second year were effective to reduce gamboge spots on the outer part of fruit, however they were not effective to reduce gamboge in aril. Dolomite applications did not increase transversal diameter, fruit weight, total soluble solids, total titratable acids, and ratio of total soluble solids with total titratable acids of the fruits.

Abstract

Papaya is one of tropical fruits native to Southern Mexico and which have been cultivated in Indonesia for a longtime. Papaya is usually propagated by seeds. Therefore, the offsprings are not true-to-type. This study was conducted todevelop a protocol of in vitro propagation of papaya ‘Sukma’ from shoot tips of in vitro germinated seeds as explants. Seedswere extracted from fruit that physiologically ripe and it germinated on MS basal medium. The experiment was set up in arandomized block design with culturing day as blocks (four blocks) and various concentrations of BA (0.0, 0.5, 1.0, 1.5, and2.0 mg L-1) in MS medium enriched with NAA 0.5 mg L-1 as treatment. Shoot tips in the MS medium without plant growthregulator as control so there was six treatments and 24 observation units. Each observation units contained five culturevessels and each culture vessels contained four explants. The results showed that the highest number of shoots, number ofleaves, number of nodes, and percentage of explant forming callus were obtained by BA 1.0-1.5, 1.0-2.0, 0.5-2.0, and 1.0-1.5 mg L-1, respectively. The highest percentage of explant forming roots were obtained in medium without BA. Analysis ofregression showed that the optimum concentration to get the highest number of shoots and leaves were BA 1.31 and 1.35 mgL-1, while explants will be rooted in medium without BA. Thus, in vitro propagation for papaya ‘Sukma’ should be conductedin two steps, i.e. for shooting and rooting growth.

Abstract

Increasingly limited land in the high elevation and increasing of global temperature needed breeding of cauliflower plants originating from temperate regions to be adaptively planted in low elevation. This study aimed to identify the morphological variability and yield component of cauliflower planted in the high elevation and low elevation. The experiment was conducted in Pasir Sarongge (1,117 m asl) and Tajur II (340 m asl) from June to December 2016. This experiment was conducted with a randomized complete block design single-factor (12 genotypes of cauliflower) with three replications. The results showed that BOB 021 genotype was not flowering in Tajur. Qualitative characteristics that are stable in two locations include leaf attitude, leaf color, curd color, leaf lobing, and curd covering by inner leaves. Vegetative characters and yields in low elevation were higher than high elevation, time of flowering and harvesting in high elevation was earlier than in low elevation. Based on the correlation analysis, an increase in the yield characters can be seen from the increase in vegetative character and increase in longevity time of flowering and harvesting. Abstract

Pepaya tipe sedang dan kecil sudah digemari di Indonesia sejak satu dekade lalu. Sebagian besar tanaman pepaya yang diperbanyak dari biji menghasilkan profit yang rendah karena heterogenitas dan variasi genetik yang diakibatkan oleh penyerbukan silang, sehingga diperlukan kegiatan secara kultur in vitro. Penelitian dilakukan di Laboratorium Kultur Jaringan Pusat Kajian Hortikultura Tropika (PKHT) IPB, Bogor, dari November 2018 hingga Januari 2020. Eksplan merupakan tunas aksilar pepaya kultivar Callina dan Caliso umur 2 minggu. Penelitian dirancang secara faktorial dalam lingkungan Rancangan Acak Kelompok (RAK). Penelitian ini dibagi menjadi dua percobaan: a) Percobaan pertama bertujuan untuk mengetahui pengaruh metode sterilisasi dengan faktor sumber eksplan (rumah kaca dan lapangan) dan metode sterilisasi (M1, M2, M3, dan M4), dan b) Percobaan kedua bertujuan untuk optimalisasi konsentrasi BAP dan GA3 pada pertumbuhan tunas dengan konsentrasi BAP 0.0, 0.5, 1.0, 1.5, dan 2.0 mg L-1, dan konsentrasi GA3 0.0, 0.3, dan 0.4 mg L-1. Hasil penelitian menunjukkan bahwa eksplan dari rumah kaca pada sterilisasi metode M4 menghasilkan tingkat kontaminasi terendah yaitu 19.9% untuk Caliso dan 29.9% untuk Callina. Media terbaik untuk menghasilkan tinggi tunas optimum adalah BAP 0.5 mg L-1 dan GA3 0.3 mg L-1 untuk Caliso dan BAP 0.5 mg L-1 dan GA3 0.4 mg L-1 untuk Callina dengan tinggi rata-rata 1.6 dan 1.8 cm, sehingga tunas dapat dipindahkan ke tahap pengakaran.
