Abstract

---------------------------This paper proposes an idea of revitalization. This idea is based on by looking at the lawsuits occurred within the Islamic laws related to family, especially poligamy. This is proposed because polygamy is believed no longer relevant with the changing times which now requires equality of between man and woman. To address this problem, some ideas put forward to reform and re-interpretate some Islamic laws which lays into two major epistemologies: first, contextual interpretation over the texts which are the basis of the implementation of poligamy which in turn was not the core mission of Islam. Meanwhile, it is believed that the fundamental goal of marriage in Islam monogamous. The justification of poligamy by putting forward the texts do not eliminate but gradually endorse the practices of polygamy itself. Therefore, there is still a room to critique the context of polygamy by contextualizing the polygamy laws in which most previous studies were not objective and tend to justify the dominant discoureses on gender relation.

Abstract

Endophytic fungus IP-2 is the endophytic fungus isolated from the plant Artemisia annua L. This fungus is known to have activity as an inhibitor of heme polymerization. The aim of this study is to examine the suitable medium for the growth and production of the active metabolite and to determine the appropriate time for harvesting the active metabolite. Endophytic fungus was fermented in 4 types of media for 14 days. Sampling was taken every day to obtain the weight of the dry cells which were used for analysis of growth. The media obtained is used to determine their activities as heme polymerization inhibitor. Tests were carried out using the method developed by Bassilico et al (1998). Data of dry cell weight and the percentage inhibition of heme polymerization were analyzed in order to obtain the curve of growth and metabolite production. Both curves will be used to determine the proper time to harvest the active metabolite. The results showed that the PDB was the best medium for endophytic fungus IP-2 growth and was able to produce the active metabolite higher than most other media. In the PDB media, at day-7 showed that active metabolite produced at most so that proper harvest time metabolite is on the 7th day of fermentation.

Abstract

Background: Previous study showed that 1,10-phenanthroline skeleton were active in vitro on both Plasmodium falciparum chloroquine-resistant and -sensitive strains. Based on the skeleton, a series of 1,10-phenanthroline derivatives have been synthesized. However the antiplasmodial activity of those molecules has not been reported. Objective: To know the in vitro antiplasmodial activity of thirteen 1,10-phenantroline derivatives and its quantitative structure-activity relationship. Methods: The in vitro antiplasmodial activity was tested on two strains of Plasmodium falciparum, FcB1Columbia (chloroquine-resistant strain) and a Nigerian (chloroquine-sensitive strain) using a radioactive micromethod. The parasite growth was estimated by [3H1-hipoxanthine incorporation after 24 and 48 hours incubation with each molecule tested. The control parasite free from any molecules was referred to as 100% growth. For this radioactive method IC5o value showing concentration inhibiting 50% of the parasite was determined graphically in concentration versus percent inhibition curves. The quantitative structure-activity relationships (QSAR) of 1,10-phenanthroline derivatives were investigated using atomic net charges as predictors of their activity. Data of predictors were obtained using semi-empirical Austin Model 1 (AM1) calculation method. The possible linear relationships of in vitro antiplasmodial activity with atomic net charge parameters of those compounds were studied. The best model QSAR was evaluated by multiple linear regression method. Results: The results showed that the IC5o values of 1,10-phenanthroline derivatives range from 0.02 to 11.05 uM for the FcB1 strain and from 0.14 to 19.84 pM for the Nigerian strain. The molecure (4), 2,10- methy1-3-12-chloroethyl)-4-chloropirydo [2,3-i] quinolineium iodide exhibited the best in vitro antiplasmodial activity with an IC50 value ranging 0.02 to 0.16 1.1M. The best model QSAR was expressed by log IC5o = -3.4398 - 14.9050 qN1 - 8.5589 qC10 - 14.7565 qC7 + 5.0457 qC11 (n = 13; r = 0.96275; r2 = 0.92689; SE = 0.61578 and F (4,8) = 25.3556).Conclusion: The molecule (4) yield the best in vitro antiplasmodial activity on both P. falciparum strains. In addition, there is correlation between antiplasmodial activity and atomic net charges of atoms on the 1,10-phenanthroline skeleton.Key words : 1,10-phenanthroline, antiplasmodial activity, quantitative structure-activity relationships

Abstract

Male and female, basically, have the same chance as idol muttaqin person, as well as a leader in the earth, including becoming a teacher of tarekat (mursyid). Assigning female as mursyid will not reduce or break the identity of the Sufism group, on the other hand, it is a kind of glorifying and respecting to the God. However, in tasawuf culture, most of the rituals conducted by the followers often reflecting the God in the male form. This conception, then, influences the way they lead the group. The common way of practicing the leadership model finally drives into legalizing the male superiority as the mursyid rather than the female one. Because of the phenomenon, the paper is written, to explore the issues on male and female role in the Sufism, specifically, for becoming the mursyid. It is expected to be powerful and meaningful cultural analysis which is viewed from tasawuf paradigm.

Abstract

Dualism dilemma between zakat and tax in Indonesia can be relatively mitigated by ratification of Act No. No. 38/1999 on Management of Zakat. In the regulation, zakat has been synergized with tax by placing zakat as a deduction from taxable income element (PKP). But so far it has not been given the significant impact on the acceptance of zakat and awareness of Muslims to pay zakat. There are also some problems in practical level that contribute to that fact. This article explores the zakat and tax synergy that have been achieved through Act No. 38 of 1999, the problems found in its execution, and of course an offer for a solution to optimize the role of zakat and tax for the people welfare. By examining same practice in some countries, this paper recommends zakat as a direct tax deduction (tax credit) as a strategic step in the effort to optimize the role of zakat.-----Dilema dualisme zakat dan pajak di Indonesia relatif bisa diredakan melalui pengesahan Undang-undang No No 38/1999 tentang Pengelolaan Zakat.

Abstract

The rapidly increasing resistance of Plasmodium falciparum to the most commonly used antimalarial drug indicates the urgent need for new antimalarial compounds. Previous study showed that among thirteen derivatives of 1,10-phenanthroline, four compounds were active in vitro and potential for further development. The study was conducted to provide the in vivo antiplasmodial activity of four 1,10-phenanthroline derivatives and to evaluate the possibility of their mechanism of action. The in vivo antiplasmodial activity of the four compounds was evaluated by a standard 4-day test on Plasmodium venckei petteri infected mice. The study of haem polymerization inhibitory activity (HPIA) and the potentiating of those compounds in combination with halofantrine and inhibitor protease (E64) were carried out to evaluate its mechanism of actions. The results showed that among four compounds tested, compound 1 (2,10-methyl-3-(2-chloroethyl)-4-chloropirydo [2,3-i] quinolineium iodide) was the most active compound with DE50 was 0.22 ± 0.03 mg/kg BW. This molecule less active than halofantrine, but more active than chloroquine. The HPIA of 1,10-phenanthroline derivatives tested (IC50 HPIA was 1.57-1.95 mmol/well) did not vary significantly. These IC50 HPIA values were significantly lower than halofantrine (CI50 HPIA was 0.12 ± 0.04 mmol/well) and than chloroquine (CI50 HPIA was 0.75 ± 0.03 mmol/well). Combination the 1,10-phenanthroline derivatives and halofantrine produced simple additive effect that implies that each drug posses a different mechanism of action. Conversely, there was antagonist interaction between 1,10-phenanthroline derivatives and E64 showing that the drugs have a similar mechanism of action. It can be concluded that the compound 1 appears as a potential antimalarial compound. In addition, the possible mechanism of action of this compound is inhibitation of protease involved in haemoglobine degradation within the malaria parasite.Key words : 1,10-phenanthroline – in vivo antiplasmodial – haem polymerization inhibitoryactivity – protease inhibitor .
