Abstract

This study was aimed to obtain the method in the study of callus induction of in vitro.  The research activities were carried out at Laboratory of Biotechnology, Faculty of Agrotechnology, Andalas University. This research used decriptive and experimental methods. One-way ANOVA analysis followed by Duncan’s test was used to determine significant differences (P ≤ 0.05). All statistical analysis were performed using the SPSS Ver. 23 statistical software package. The results showed that medium of callus induction is 4,0 mg/L BAP + 0,5 mg/L IAA with formation of callus (days), and average of callus formed, namely 4,33 days,  and 9,67 callus.

Abstract

Abstract

The 3rd generation mutant brown rice from the experimental cultivation of the 3rd generation mutant has problems in plant characteristics and various genetic traits. In the results of the research of the 3rd generation of mutants, several mutants with production levels were obtained in accordance with the initial objectives of the study, namely early-mature mutants and significant plant height. However, some mutants have unstable plant characters and traits, so a multi-location test of cultivation of the 3rd generation mutant was carried out at two locations with different elevations in order to obtain a 4th generation mutant which is more stable and has uniform characters and properties. This research was conducted in Jorong Sungai Batang, Tanjung Raya District, Agam Regency with a medium elevation level of 637,032 m above sea level and in Surau Gadang Village, Nanggalo District, Padang City with a minimum elevation level of 10.363 m above sea level. This research was carried out from December 2019 to April 2020. The materials used were M3 harvested red rice seeds of the Sigah genotype, namely mutant numbers MG13, MG9, MG2-209, MG5-58,MG6 and control seeds of red rice genotype Sigah. The purpose of the study was to obtain diversity and determine the stability of agronomic characters in the production of local red rice yields of West Sumatran Sigah M4 variety at two different elevations of the cultivation environment. From the results of the research, it was found that the diversity and agronomic characters of plants were quite stable in mutants numbered MG13, MG2-209 and MG5-58 with early plant age, and stable growth in the generative and vegetative phases.

Abstract

Produksi beras dunia hilang 25% setiap tahun karena kerusakan yang disebabkan oleh hama serangga, sepeti wereng. Salah satu upaya untuk mengendalikan hama wereng batang coklat yaitu dengan menggunakan genotipe lokal yang tahan. Penelitian ini bertujuan untuk mengetahui intensitas serangan dan tingkat ketahanan dari dua puluh empat genotipe lokal Kabupaten Kuantan Singingi terhadap hama wereng batang coklat. Penelitian ini menggunakan metode Rancangan Acak Lengkap satu faktor dengan empat ulangan. Faktor perlakuan yang dicobakan adalah 24 genotipe padi local Kabupaten Kuantan Singingi ditambah satu varietas yang tahan (Inpari 13) dan satu varietas yang rentan (TN1). Hasil penelitian menunjukan bahwa seluruh genotipe yang diuji 100% terserang wereng batang coklat dengan tingkat intesitas serangan yang berbeda-beda. Berdasarkan tingkat intensitas serangan ditemukan katagori ringan dan berdasarkan SES IRRI 2013 ditemukan genotipe dengan kriteria tahan, yaitu pada genotipe Padi Sironda Putih (PL01), Padi Singgaro Merah (PL06), Padi Kuning Umur Panjang (PL07), Padi Ros (PL08), Padi Samo Putih (PL09), Padi Limbayang (PL10), Padi Sokan Umur Panjang (PL12), Padi Singgam Putih (PL14), Pulut Benai (PL17), Padi Kuning (PL21), Padi Gondok (PL22) dan Padi Katiok Putih (PL24). Tingkat intesitas serangan dengan katagori sedang terdapat pada genotipe Padi Saronda Merah (PL02), Pulut Hitam (PL04), Padi Ronda Putiah (PL05), Pulut Benai (PL13), Padi Singgam Kuriak (PL15), Pulut Kari (PL16), Padi Putih (PL19), Pulut Lupo Ka Laki (PL20) dan Padi Saronda Kuning (PL23). Tingkat intesitas serangan dengan katagori berat terdapat pada genotipe Padi Pandan Wangi (PL03), Pulut Karate (PL11) dan Padi Kuning (PL18).

Abstract

One of the causes of the low productivity of rice grown on peat soils was the high levels of of toxic phenolic compounds in the soil. Development of tolerant rice varieties is one of strategies to grow rice on peat soils. The aims of this research were to select rice varieties tolerant to peat soils and to study the correlation between the plant phenolic acid content with plant tolerance to peat soil. This research was conducted at the Seed Farm Centre Lubuk Minturun, Padang from February to July 2009. The research consisted of two experiments i.e. in peat soil and in mineral soil. The experiments used a completely randomized design using 21 varieties of rice as treatments which were replicated four times. The results showed that there were nine rice varieties categorized as tolerant to peat soil. These tolerant varieties showed higher stress tolerance index (STI), demonstrated by a high level of phenolic acids (coumaric, syringic, and ferulic) in the plant tissues. The STI value had positive correlation with the content of plant phenolic acids, thus the level of plant phenolic acids can be used as an indicator to determine tolerant rice varieties in peat soils. Abstract

Padi lokal masih banyak ditemukan dan merupakan aset sumber daya genetik dalam penyediaan varietas unggul yang adaptif, sehingga pengembangannya masih terus diupayakan. Salah satu jenis padi gogo lokal di Sumatera Utara yang banyak ditanam masyarakat adalah padi gogo beras merah, selain memiliki keunggulan baik sebagai makanan pokok maupun fungsi kesehatan bagi tubuh. Varietas lokal biasanya beradaptasi baik pada daerah asalnya dengan rasa nasi dan aroma sesuai selera masyarakat setempat. Namun demikian padi lokal memiliki kekurangan seperti umur dalam, batang tinggi sehingga mudah rebah, tidak responsif terhadap pemupukan dan produksi rendah. Penelitian ini dilaksanakan sejak April 2016 sampai Juni 2017 bertujuan untuk memperbaiki genetik padi beras merah lokal Sumatera Utara (Sigambiri merah) khususnya terkait umur tanaman agar lebih genjah dan postur pendek/semi pendek melalui pemuliaan mutasi (mutasi induksi). Untuk mendapatkan dosis optimum, benih padi diiradiasi dengan sinar gamma Co-60 dosis 0, 100, 200, 300, 400, 500, 600, 700, 800, 900, dan 1000 Gy di Pusat Aplikasi Isotop dan Radiasi-Badan Tenaga Nuklir Nasional (PAIR - BATAN), Jakarta. Penanaman M1 dan M2 dilaksanakan di BPTP Sumatera Utara. Dari hasil pengamatan persentase tumbuh bibit, tinggi tanaman dan panjang akar pada fase pembibitan, dan persentase kehampaan gabah pada tanaman M1 diperoleh dosis iradiasi 200 - 300 Gy merupakan dosis yang efektif dalam menghasilkan keragaman genetik. Hal ini juga terlihat pada populasi tanaman M2 hasil iradiasi 200 Gy menghasilkan jumlah mutasi klorofil yang tergolong luas dengan 8 macam tipe mutasi dari 8 tipe, yaitu albina, xhanta, viridis, tigrina, spotting leaf, alboviridis, marginata, dan striata. Juga menghasilkan keragaman genetik yang luas pada variabel karakter tinggi tanaman, jumlah anakan produktif dan umur panen. Hasil seleksi yang dilakukan pada populasi M2 diperoleh kandidat mutan genjah sebanyak 69 kandidat dengan frekuensi mutasi sebesar 1,09%. Tanaman genjah dan postur pendek (dwarf)/semi-pendek (semidwarf)  terseleksi tentu akan sangat berguna sebagai bahan tanaman awal dalam perbaikan varietas padi beras merah dalam program pemuliaan tanaman ke depan.

Abstract

Kultivar padi beras hitam merupakan padi lokal yang berasal dari Pasaman Timur yang memiliki umur panjang dan tinggi tanamannya terlalu tinggi.  Penelitian ini dilaksanakan sejak Maret 2012 sampai Maret 2013 bertujuan untuk memperbaiki genetik padi beras hitam lokal Sumatera Barat khususnya terkait umur tanaman agar lebih genjah melalui pemuliaan mutasi.  Untuk mendapatkan dosis optimum, benih padi beras hitam diiradiasi dengan sinar gamma 60Co dosis 0, 100, 200, 300, 400 dan 500 Gy di Pusat Aplikasi Isotop dan Radiasi-Badan Tenaga Nuklir Nasional (PAIR – BATAN), Jakarta. Dari hasil pengamatan persentase kecambah, tinggi tanaman dan panjang akar pada fase pembibitan, dan persentase kehampaan gabah M1 diperoleh dosis iradiasi 200 - 300 Gy merupakan dosis yang efektif dalam menghasilkan keragaman genetik.  Hal ini juga terlihat pada populasi M2 hasil iradiasi 200 Gy menghasilkan keragaman genetik yang luas pada variabel karakter tinggi tanaman, jumlah anakan produktif dan umur tanaman. Dari hasil seleksi yang dilakukan pada populasi M2 diperoleh kandidat mutan genjah sebanyak 81 kandidat dengan frekuensi mutasi sebesar 0.08 %.  Tanaman genjah terseleksi tentu akan sangat berguna sebagai bahan tanaman awal dalam perbaikan varietas padi beras hitam di masa yang akan datang. Abstract

This Research aimed to select wheat genotypes which potential to be developed in a high rainfall area. It was conducted at September 2012 to January 2013 in Alahan Panjang, Solok Regency, West Sumatera. Seventeen wheat genotypes were planted at Alahan Panjang, Solok, West Sumatera from September 2012 – January 2013, including local varieties (Nias, Selayar, Dewata). It used random design block with 3 replications and 10 samples each block. If genotypes were significant according to test f then continued by test duncan on standard 5 %. Observed variables were amount of spikelet per spike, amount grain of spikelet, amount of floret per spike, spikelet length and width, grain length and width, amount of full grain, amount of unfull grain and a thousand grain weight. The result showed that UAGDM 13 had a good amount of spikelet per spike. All genotypes had high amount of floret per spike except UAGDM16. A good spikelet length was belonged to all genotypes except UAGDM11 and UAGDM13. Grain length and width were not significant among all genotypes. Three genotypes which had a high amount of full grain were UAGDM5, UAGDM6 and UAGDM17. Genotype UAGDM1 was the best of a thousand grain weight.

Abstract

Karakterisasi dan evaluasi beberapa genotipe sorgum koleksi dari fakultas pertanian Universitas Andalas telah dilakukan mulai Agustus 2012 sampai dengan Desember 2012 di Sukarami Kabupaten Solok. Tujuan penelitian ini adalah untuk melihat keragaman fenotipe sorgum di daerah dataran tinggi. Genotipe yang di uji adalah L1, L2, L3, L4 dan L5. Metoda yang digunakan adalah menanam genotipe sorgum dalam petakan berukuran 2x3 m, dengan sampel yang diamati masing-masing genotipe adalah 6 sampel. Kegiatan perekaman data dilakukan terhadap keragaan karakteristik tanaman sorgum secara kualitatif. Pada masa vegetatif pengamatan seperti (Bentuk Ujung Daun, Kehadiran Lidah daun, Permukaan Daun, Warna Batang Kecambah), dan pada masa generatif pengamatan berupa (warna biji, bentuk biji, ukuran biji, warna sekam dan sifat sekam). Pertumbuhan genotipe sorgum yang diamati pada masa vegetatif, tidak menunjukkan perbedaan yang signifikan secara morfologi antara genotipe L1, L2, L3, L4, dan L5. Morfologi daun seperti ujung daun, bentuk lidah daun, keadaan permukan daun, pertulangan daun kelima genotipe sorgum memiliki sifat yang seragam. Perbedaan genotipe sorgum dapat dikenali lebih jelas pada fase generatif dibandingkan fase vegetatif.

Abstract

One of the important things in determining the quality of livestock products is feed ingredients. The quality of Poultry products will be influenced by the nutrient intake provided. Tomato reject is one of the special ingredients of a feed because it contains high lycopene. Lycopene is well known as an antioxidant, also it can lower cholesterol content. However, lycopene still in the matrix of tomato tissue and trans-form lycopene what difficult to digest and absorb in the body. So, in this paper presents the extent to which the development of research utilization of tomatoes high lycopene as poultry feed. As well as opening more widely how to increase the value of lycopene tomatoes in poultry products.
