Abstract

The mammary gland contains adult stem cells that are capable of self-renewal and are likely target for neoplastic transformation leading to breast cancer. In this study, we developed a cell culture derived from the mammary glands of cynomolgus monkeys (Macaca fascicularis)(MfMC) and furthermore identified the expression of markers for stemness and estrogenreceptor-associated activities. We found that the primary culture can be successfully subcultured to at least 3 passages, primarily epithelial-like in morphology, the cultured cells remained heterogenous in phenotype as they expressed epithelial cell markers CD24, CK18, and marker for fibroblast S1004A. Importantly, the cell population also consistently expressed the markers of mammary stem cells (ITGB1 or CD29 and ITGA6 or CD49f), mesenchymal stem cells (CD73 and CD105) and pluripotency (NANOG, OCT4, SOX2). In addition to this, the cells were also positive for Estrogen Receptor (ER), and ER-activated marker Trefoil Factor 1, suggesting an estrogen responsiveness of the culture model. These results indicate that our cell culture model is a reliable model for acquiring a population of cells with mammary stem cell properties and that these cultures may also serve as a reservoir from which more purified populations of stem cell populations can be isolated in the future.

Abstract

Cynomolgus macaques (Macaca fascicularis) are an established animal model of asthma, which exhibit different responses to allergen exposure that are clinically relevant. The chemokine ligand gene (CCL7) encodes Monocyte Chemotactic Protein-3, which has an important role in asthma pathogenesis. While CCL7 polymorphism in humans is associated with asthma phenotype, very little is known about CCL7 in nonhuman primate models of respiratory disease. The objective of this study was to isolate and characterize CCL7 gene in cynomolgus macaques of Indonesian origin. In this study, we used sequencing and bioinformatics technique for gene isolation, characterization, and protein 3D structure prediction. We isolated a 2253 base-pair (bp) sequence of CCL7 in cynomolgus macaques, which exhibited 95% similarity in coding sequence to human CCL7. The amino acid sequence was more closely clustered with human CCL7 than with that of rodents. Importantly, the predictive protein structure of CCL7 was similar to that in humans. These similarities in CCL7 suggests the potential of cynomolgus macaque as a translational model to study asthma, particularly in the context of genetics and role of chemokines such as CCL7.

Abstract

Bone marrow mesenchymal stem cells have been widely used, because plasticity, specific surface markers, self-renewal to transform into various lineages including cardiomyocytes. Information about the connexin (Cx) cardiac conduction systems of the pigtail macaque (Macaca nemestrina) is limited. This study aimed to evaluate cardiomyocyte differentiation from bone marrow mesenchymal stem cells of pigtail macaques and to clarify the Cx cardiac conduction system. Bone marrow aspirates were obtained from the proximal humerus of four adult male pigtail macaques, collected into heparinized tubes, then centrifuged to obtain mononuclear cells that were isolated and cultured in an incubator. After these cells reached 70–80% monolayer confluency as homogeneous fibroblast-like cells, they were subcultured. On the second subculture passage, the cells were pelleted to extract the mRNA, which was analysed by reverse transcription–polymerase chain reaction, and then cultured for a third passage. Cells were positive for CD73 and CD105 and the reference gene glyceraldehyde-3-phosphate dehydrogenase, and negative for CD34 and CD45. Osteogenic, chondrogenic, adipogenic, and cardiomyocyte differentiation was confirmed based on specific staining. The pigtail macaque bone marrow mesenchymal stem cells can be isolated and subcultured. The transcription of genes and translation of proteins of the connexin cardiac conduction systems was successfully identified.

Abstract

Reverse transcriptase (RT) enzyme is a key tool in molecular biology for the synthesis of complementary DNA (cDNA) from messenger RNA (mRNA). Combining RT activity with PCR amplification has been a gold standard as the first step in cloning the coding region of any gene of interest. Evidently, RTs have been critical in advancing molecular biology, genetics and medicine to their current stage. In this study, we were developing the RTDRNase H recombinant enzyme isolated from serotype-2 simian betaretrovirus-2 (SRV-2) infected Indonesian Macaca fascicularis using Escherichia coli expression system. The study was conducted using RT SRV-2 gene expression using E. coli expression system, proteins purification, and application to RT PCR technique. The SDS PAGE expression analysis showed a specific band size of 32.7 kDa assumed as RT protein enzyme. Application of RT SRV-2 enzyme generated to the RT PCR technique of β-globin CDV and SRV-2 env gene target showed that the RT SRV-2 enzyme was capable to reverse transcribed mRNA into cDNA as indicated by the presence of specific DNA band compared with commercial RT enzymes. This RT SRV-2 enzyme showed its activity similar to that of commercial one, although the activity was lower.

Abstract

The aim of this study was to determine the content of total monomeric anthocyanins and to identify the anthocyanin composition of jambolan fruit (Syzygium cumini) growing in Indonesia. Anthocyanins were extracted with methanol containing 0.1% HCl, removed from non-polar impurities by partition with chloroform and purified in a C18 solid phase cartridge. Anthocyanins were identified by high-performance liquid chromatography with diode array detection (HPLC-DAD), UV-visible spectral analysis, and physico-chemical reactions. Using pH-differential method, it was known that the content of total monomeric anthocyanin in the ripe fruit was 161 mg/100 g fresh fruit (731 mg/100 g skin on wet basis or 3430 mg/100 g skin on dry basis), based on cyanidin-3-glucoside. Furthermore, five anthocyanins without acylating groups were identified as 3,5-diglucoside derivatives of delphinidin (41.29%), petunidin (27.79%), malvidin (25.60%), cyanidin (4.19%), and peonidin (1.13%). These results demonstrate that the skin of jambolan fruit especially the ripe one is rich in anthocyanin, with wide range in anthocyanidin backbone (five aglycons).

Abstract

The effect of two fractions of oleoresin compounds from ginger root on lymphocyte function were studied by observing B cell proliferations, which were  measured by the incorporation of 3H-thymidine during cell incubation. The doses of oleoresin compounds and its two fractions tested were 50,100, 150, and 200 mg per ml. Lymphocyte from human peripheral blood were isolated using ficoll density gradient technique, and cultured  in the presence of the compounds in RPMI-1640 medium for 4 days, with or without the addition of 3 mM paraquat as the oxidizing agent.  The results showed that the effects of oleoresin and its two fractions depended on its doses. Oleoresin and its two fractions increased B cell proliferation with  the highest activity of 456 percent at low dose of 50 mg per ml. These result showed a positive effect of oleoresin compound and its two fractions on B cell functions  at low doses. It indicated that bioactive compounds of ginger root at low  concentration have  positive effects on humoral immune response and this supports the traditional belief that ginger increases body resistance to common cold.

Abstract

Nikotin dengan dosis tertentu telah dilaporkan memiliki pengaruh terhadap penurunan nafsu makan pada tikus, tetapi belum pernah dilaporkan pada hewan model monyet ekor panjang. Oleh karena itu penelitian ini bertujuan untuk mengetahui pengaruh nikotin terhadap bobot badan, indeks massa tubuh (IMT) dan kadar glukosa darah monyet ekor panjang (Macaca fascicularis) yang diberi diet berenergi tinggi selama tiga bulan. Sepuluh monyet jantan dewasa secara acak dibagi menjadi dua kelompok. Kelompok pertama diberi pakan dengan bahan utama dari lemak sapi dan kelompok kedua dengan pakan monyet komersial (monkey chow). Cairan nikotin ditambahkan kedua pakan dengan dosis 0,75 mg/kg berat badan. Rancangan yang digunakan dalam penelitian ini adalah rancangan acak lengkap (tersarang) dalam waktu. Data yang dikumpulkan dianalisis untuk mengetahui hubungan antara waktu dan perlakuan. Pengukuran dilakukan setiap empat minggu terhadap berat badan, indeks massa tubuh dan kadar glukosa darah. Abstract

Tikus putih (Rattus norvegicus) merupakan hewan coba yang digunakan untuk penelitian biomedik. Penelitian ini bertujuan untuk memberikan nilai tekanan darah normal tikus putih dengan galur (Wistar dan Sprague-Dawley) dan jenis kelamin yang berbeda melalui pengukuran tekanan darah non-invasive. Pengukuran tekanan darah dengan menggunakan instrumen CODA® pada 20 ekor tikus putih. Profil tekanan darah yang diukur meliputi tekanan darah sistolik, diastolik, tekanan arteri rata-rata, denyut jantung, dan aliran darah pada ekor. Nilai tekanan darah sistolik, diastolik, dan tekanan arteri rata-rata pada galur Sprague-Dawley lebih tinggi secara signifikan (P<0.05) dibandingkan Wistar. Tikus putih galur Sprague-Dawley jantan memiliki profil tekanan darah diastolik dan tekanan arteri rata-rata yang lebih tinggi secara signifikan (P<0.05) dibandingkan betina. Galur Wistar jantan menunjukkan profil tekanan darah yang lebih tinggi dibandingkan betina namun tidak berbeda secara signifikan (P>0.05). Sprague-Dawley jantan menunjukkan hasil tekanan darah diastolik dan tekanan arteri rata-rata yang lebih tinggi secara signifikan (P<0.05) dibandingkan Wistar jantan. Studi ini menekankan pentingnya memilih ras dan jenis kelamin hewan coba tikus dalam studi kardiovaskuler.

Abstract

Penentuan status reproduksi pada satwa liar atau satwa yang ditangkarkan merupakan faktor yang sangat penting dalam manajemen pengembangbiakan satwa. Evaluasi metabolit hormon estrogen dan progesteron secara non-invasive dari sampel feses untuk memonitor fungsi reproduksi telah dilakukan sejak lama pada beberapa spesies mamalia. Validasi asai pada Tarsius belum pernah dilaporkan sehingga validasi asai merupakan hal yang sangat penting sebelum digunakan dalam studi karena metabolit steroid bersifat spesifik spesies. Tujuan penelitian ini adalah melakukan validasi analitik kit enzyme linked immunoassay (ELISA) komersial untuk menganalisis metabolit hormon estrogen dan progesteron pada feses T. spectrum. Uji paralelisme dilakukan pada asai DRG® estradiol (E2), estron (E1), dan progesteron (P4) dengan pengenceran bertingkat (1:2–1:128) ekstrak feses dari beberapa status reproduksi yang berbeda  pada Tarsius yang dibandingkan dengan kurva standar dari masing-masing asai.  Hasil uji paralelisme terhadap kit DRG® estron menunjukkan hasil yang tidak paralel. Dari uji paralelisme DRG® estradiol dan progesteron, didapatkan hasil kurva sampel dengan standar yang tidak konsisten. Hanya ditemukan satu dari lima kurva sampel yang diuji yang paralel dengan kurva standar asai DRG® estrogen dan progesteron. Berdasarkan hasil tes paralelisme tersebut, kit komersial ELISA DRG® estron, estradiol, dan progesteron tidak dapat digunakan untuk mengukur metabolit  estrogen dan progesteron pada feses T. spectrum.

Abstract

Aterosklerosis pada arteri iliak berkaitan dengan kejadian peripheral arterial disease (PAD) pada manusia. Penelitian ini bertujuan untuk menganalisis lesio aterosklerosis pada arteri iliak sebagai salah satu faktor penyebab PAD. Penelitian ini menggunakan sembilan monyet ekor panjang jantan dewasa dari 3 kelompok respons yang diberi pakan aterogenik IPB-1 selama 2 tahun. Arteri common iliak diambil pada saat nekropsi, diproses menjadi slaid histologi dan diwarnai dengan Verhoeff van Giesson (VVG). Keparahan lesio dikategorikan sebagai grade I sampai VI dan dianalisis secara deskriptif. Area lesio ditentukan melalui panjang lamina elastik interna (PLEI), area intima (AI), serta ketebalan maksimum intima (KIM) dan dianalisis dengan ANOVA. Hasil penelitian menunjukkan 55,56% monyet membentuk lesio plak aterosklerosis (grade IV sampai VI) yang mana mereka berasal dari kelompok intermediet dan hiper-responsif. Kelompok hiper-responsif menunjukkan AI dan KMI paling tinggi (0,979 mm2; 0,352 mm) diikuti kelompok intermediet (0,372 mm2; 0,237 mm) dan kelompok hipo-responsif (0,049 mm2; 0,052 mm) (P<0,05). Hasil ini menunjukkan semakin tinggi kolesterol plasma, semakin luas area lesio yang terbentuk. Keseluruhan hasil mendukung bahwa monyet yang diberi pakan aterogenik IPB-1 selama 2 tahun mampu membentuk lesio plak aterosklerosis pada arteri common iliak, dan berpotensi sebagai hewan model PAD, kecuali kelompok hipo-responsif.
