Abstract

Grain quality and aroma are important characteristics of rice that affect consumer acceptance. This research was conducted to study the grain quality and aroma of aromatic new plant type (NPT) promising rice lines. Thirty five lines as well as Ciherang and Sintanur varieties were planted at Bogor and Pusakanagara, West Java in the dry season (DS) 2009 and wet season (WS) 2009. Three methods, i.e. leaf aroma tested with KOH, rice aroma tested in the test tube, and cooked rice aroma test, were used to evaluate the aroma of the lines. The results showed that line B11742-RS*2-3-MR-34-1-2-1 was aromatic identified using different methods. The line had long, slender, and small chalkiness grains, high percentage of head rice, high amylose, and hard texture. Lines IPB 140-F-6, B11249-9C-PN-3-3-2-2-MR-1, and B11955-MR-84-1-4 also had a high aroma score and grain yield. Testing leaf aroma with KOH can be used as early selection method in breeding program for aromatic lines. Lines derived from aromatic parents from highlands of South Sulawesi did not show consistent aroma under three testing methods. Those tested lines had good grain quality, both physical and cooked rice quality.

Abstract

Late blight resistance gene (RB gene) isolated from Solanum bulbocastanum, is a broad resistance gene against all races of Phytophthora infestans. The gene was transformed into Katah-din event SP904 and SP951 using Agrobacterium tumefaciens and these transgenic plants have been crossed with susceptible potato cultivars Atlantic and Granola. Populations of the crosses have been molecularly characterized for the integration of the RB transgene. The study aimed to evaluate the resistance of the populations of crosses between transgenic Katahdin RB  and susceptible non-transgenic parents (Atlantic and Granola) to late blight in a confined field trial at Pasir Sarongge, Cianjur, West Java. A total of 84 clones originated from four popula-tions were evaluated for resistance to late blight. These included 22 clones of Atlantic x transgenic Katahdin SP904, 16 clones of Atlantic x transgenic Katahdin SP951, 19 clones of Granola x transgenic Katahdin SP904, and 27 clones of Granola x transgenic Katahdin SP951. Observations of the late blight infection were conducted when late blight symptoms were detected, i.e. at 56, 60, 63, 70, and 77 days after planting (DAP). The result showed there were high variations in the resistance level of all the 84 clones tested. Clones of crosses between susceptible parents (Atlantic or Granola) and resistant parents (transgenic Katahdin SP904 or Katahdin SP951) showed a similar pattern based on the area under disease progress curve (AUDPC) value, i.e. 377.2 greater than the AUDPC of the resistant parents (180.1), but smaller than that of the susceptible parents (670.7). Observation at 77 DAP resulted four resistant potato clones having resistance score of 7.0-7.6, higher than the transgenic parents Katahdin SP904 (4.6) and Katahdin SP951 (6.8), i.e. clone B8 (Atlantic x transgenic Katahdin SP951) with resistance score of 7.6 and clones B26 (Atlantic x transgenic Katahdin SP951), C183 (Granola x transgenic Katahdin SP904), and D89 (Granola x transgenic Katahdin SP951) with resistance score of 7. These four transgenic potato resistant clones need to be further developed as promising potato clones to late blight.

Abstract

Infection of Begomovirus has caused leaf curl disease in tomato. This infection has significantly impact on yield losses of tomato production. Recently, in Indonesia there was no effectively way to control this disease.  The use of resistant tomato variety is one of strategies to control this virus. Genetic engineering technology gives an opportunity to develop the transgenic tomato resistant to Begomovirus through pathogen derived resistance (PDR) approach. The objectives of this study were to construct the  Begomovirus AV1 candidate gene in the pBI121 and to introduce the construct into tobacco plant genome through Agrobacterium tumefaciens vector. A series activites in gene construct have been conducted include PCR amplification of AV1 gene using a pair of specific primer, cloning the gene into pGEM-T easy, transformation of the clone into Escherichia coli DH5α competent cell, construct the gene into pBI121, and transform the construct into A. tumefaciens. Leaf segments of in vitro tobacco plant were transformed by co-cultivation with A. tumefaciens containing ToLCV-AV1 construct. In the research activitiy, Indonesian  Begomovirus  AV1 gene  was  successfully amplified and inserted in expression vector plasmid pBI121. Tobacco transformants carrying kanamycin-resistant gene (nptII gene) were regenerated and established in the glasshouse. Those transformant plants are expected containing the AV1 gene.

Abstract

Potato late blight, caused by Phytophthora infestans is one of the most devastating plant disease. Potato yield losses due to this disease ranged from 47-100%. A major late blight resistance gene, called RB, previously was identified in the wild potato species Solanum bulbocastanum. RB gene has been integrated into cultivated potato Katahdin using Agrobacterium-mediated transfor-mation, and showed durable and broad spectrum resistance either in laboratory assay or in confined field trial. Evaluation of transgenic Katahdin SP904 and SP951 was conducted to verify whether the RB gene with broad spectrum to all known races of P. infestans in the United States and in Toluca, Mexico was also effective against P. infestans isolates in Indonesia. Efficacy of RB gene was evaluated for foliar and tuber resistance to West Java isolates. Transgenic Katahdin were more resistant in foliar than non transgenic plants, at 14 days after inoculation. Diseases intensity of transgenic Katahdin SP904 and SP951 were 19.8-43.8%, whereas non transgenic Katahdin, Granola, and Atlantic were 46.9-100%. In contrast to the foliar resistance phenotype, RB-containing tubers in transgenic Katahdin did not exhibit increased resistance to Lembang, Pangalengan and Galunggung isolates. Tubers of transgenic Katahdin SP904, SP951, and non transgenic Katahdin showed lesion volume of 0.93, 0.91, and 0.91 cm3, respectively. RB gene in transgenic Katahdin showed efficacy against late blight P. infestans in foliar, but did not showed efficacy in tuber. Transgenic Katahdin RB thus providing a potential source of resistance for breeding programs.

Abstract

Tolerance of Transgenic Tobacco Expressing P5CS Gene Against Drought Stress. Drought is majorosmotic stress that dramatically limit plant growth and productivity. Proline accumulation has been correlatedwith tolerance to drought stress in plants. Therefore, overproduction of proline in plants may lead to increasedtolerance against these abiotic stresses. The objectives of this experiment were to determine the effects of droughtstress at the period of 15 – 90 days after planting (DAP) on growth of T1 plants derived from transgenic GStobacco, to evaluate their tolerance against drought stress, and to determine their leaf proline content. One groupof the tobacco plants were grown in plastic pots and subjected to stress condition during the period of 15 – 90DAP. The other group was grown optimally in plastic pot up to harvest period. All tobacco plants were harvestedat 91 DAP. Leaf proline content was determined at 63 DAP (after six periods of stress). The results indicatedreduced plant height, shoot diameter, leaf number, leaf dry weight and leaf area of all tobacco plants. Stresssensitivity index calculated using leaf dry weight character grouped T1 plants derived from P5CS transgenicGS tobacco into tolerance, medium tolerance and sensitive against drought stress while that of non-transgenicGS tobacco were only medium tolerance and sensitive against drought stress. Higher leaf proline content underdrought stress was observed in all T1 plants derived from P5CS transgenic tobacco than that of non-transgenicGS tobacco. These data demonstrated that proline accumulation as an osmoprotectant and that over-expressionP5CS gene results in the increased tolerance to osmotic stress in T1 plants derived from P5CS transgenic tobacco.Key words: Proline biosynthesis, proline accumulation, sensitivity index, biomass yield Stres kekeringan merupakan masalah utama stres osmotik yang dapat menjadi faktor pembataspertumbuhan dan produktivitas tanaman. Akumulasi prolina berkorelasi dengan tingkat toleransi tanamanterhadap stres kekeringan. Oleh karena itu over-produksi prolina diduga dapat meningkatkan toleransi tanamanterhadap stres kekeringan. Tujuan penelitian ini adalah (1) untuk mengevaluasi pengaruh stres kekeringanmelalui pengurangan air terhadap pertumbuhan dan hasil tanaman TI zuriat dari tembakau GS transgenik P5CSgenerasi TO, (2) menganalisis akumulasi prolina daun tanaman TI zurlat dari tembakau GS transgenik P5CSgenerasi TO kondisi stres dan non-stres, serta (3) menganalisis hubungan antara akumulasi prolina daun padakondisi stres kekeringan dengan pertumbuhan dan hasil tanaman. Percobaan dua faktor (tembakau transgenikstres kekeringan) disusun dengan rancangan acak kelompok. Sebagian tanaman yang diuji disiram setiap hari hingga mencapai kondisi kapasitas lapang dari awal tanam sampai dengan 90 HST dan digunakan sebagaiperlakuan non-stres. Sedangkan kelompok tanaman yang lain dipelihara dalam kondisi kapasitas lapanghingga 14 HST dan diberi perlakuan stres kekeringan dari umur 15 HST hingga panen (90 HST). Kandunganprolina diukur pada umur 63 HST (setelah 6x periode stres). Hasil penelitian menunjukkan semua tanamantembakau yang diuji mempunyai tinggi tanaman, diameter batang, jumlah, berat kering dan luas daun yanglebih rendah akibat perlakuan stres kekeringan yang diberikan dibandingkan dengan kondisi non-stres.Berdasarkan hasil perhitungan indeks sensitivitas terhadap stres kekeringan menggunakan peubah bobotdaun kering per tanaman maka tanaman TI zurlat dari tembakau GS transgenik P5CS generasi TO yang diujibersegregasi untuk kategori toleran, medium toleran dan peka, sedangkan tembakau GS non-transgenikdikategorikan sebagai medium toleran dan peka terhadap stres kekeringan. Tanaman TI zurlat dari tembakau GStransgenik P5CS generasi TO menunjukkan kandungan prolina yang lebih tinggi dalam kondisi stres kekeringandibandingkan dengan tembakau GS non-transgenik. Peningkatan akumulasi prolina yang cukup tinggi akibatover-ekspresi dari gen P5CS diduga berkorelasi dengan peningkatan toleransi tanaman terhadap stres kekeringan.Kata kunci : Biosintesis prolina, akumulasi prolina, indeks sensitivitas terhadap stres, produksi biomasa

Abstract

The F1 hybrid sterility in indica/japonica crosses is the major barrier in developing hybrid rice varieties between these two diverse germplasm. The sterility problem in japonica/indica hybrids can be overcome by using wide compatibility genes. The objective of this study was to identify wide compatibility varieties (WCVs) in some tropical japonica rice. Twenty five tropical japonica varieties as male parents were crossed with indica (IR64)and japonica (Akitakomachi) testers as female parents. The crosses were planted following a randomized complete block design with three replications. Varieties having average spikelet fertility of more than 70% with both the indica and japonica testers were rated as WCVs. Result from this study showed that six tropical japonica varieties were classified as WCVs, i.e., Cabacu, Grogol, Kencana Bali, Klemas, Lampung Lawer, and Napa. Hybrid sterility is caused by partial sterility of male and female gametes. The WCVs from the present study can be used in hybrid rice breeding program to solve hybrid sterility in indica/japonica hybrids.

Abstract

ABSTRACTPreliminary study: Gene transfer from transgenic potato Katahdin RB to non transgenicpotato. One of the concerns associated with the release of transgenic crops, is the possibilityof the gene flow from transgenic crops to neighboring crops of the same species or to relatedspecies. In plants, gene flow is a routine process occur through the natural hybridization. Theopportunity for gene flow occur depends principally on two factors, the degree of sexualcompatibility between donor and recipient species, and the physical distance between thetwo. The experiment was conducted to determine whether the gene flow from transgenicpotato Katahdin RB to non transgenic was occurred, based on selection using a 50 mg/lkanamycin, and to estimate gene flow mediated by natural hybridization at different isolationdistances. Preliminary result indicated that a rapid and simple method using MS0 liquid mediawith kanamycin 50 mg/l was effective for screening the seeds. There was a gene flow fromtransgenic potato Katahdin RB to non transgenic, based on a rapid and simple selectionmethod using 50 mg/l of kanamycin as selectable marker. The isolation distance used in thestudy were 0.8, 1.6, 2.4, 3.2, 4.0, 4.8, 5.6, 6.4, 7.2, 8.0, 8.8, 9.6, 10.4, and 11.2 m from the row oftransgenic potato Katahdin RB. The gene flow through natural hybridization at a isolationdistances of (0.8 - 1.6 m), (2.4 – 4 m), and (4.8 – 6.4 m) from transgenic to non transgenic plantswere 13.78, 10.92, and 3.82%, respectively. At a distance of 7.2 – 8 m, the frequency of gene flowwas declined to 0%. The frequency of gene flow from transgenic potatoes to non transgenicpotatoes markedly decreased by increasing the isolation distance, and was negligible at 7.2 m.Key words : natural hybridization, transgenic potato RB, kanamycin selection

Abstract

First generation (TO) transgenic plants do not always segregate their transgenes in a Mendellian segregation pattern. Moreover,instability of heterologous gene expression was often observed in transgenic plants. This phenomenon is often called gene silencing. Gene silencing could happen on different level of gene expression, notably at transcriptional or post-transcriptional level.The purpose of this research was to identify the transgene segregation pattern of a marker gene (hpt) as well as the introduced- regulator gene (OsLEA-oshox6) in second generation (Tl) transgenic rice plants.Gene segregation (hpt) analysis was carried out using PCR method. Gene expression analysis was done by hygromycin antibiotic resistant test of leaf samples.Analysis was carried out on 17 lines of Tl transgenic rice plants from Batutegi and Kasalath cultivars.Mendellian segregation pattern of 3:1 was revealed for all lines based on PCR analysis. Gene expression analysis showed almost all lines was segregated in a Mendellian fashion except for Tl-BT III 2C line. Less transgenic plants that expressed the hpt gene were suggested due to gene silencing effects.It was suggested to happen at transcriptional level.

Abstract

Information on genetic variability and heritability of plant characters is important to support the rice selection program. A study was conducted to determine the genetic variability of characters and grain yields of 35 New Plant Type Rice Lines (NPTR) under two different environments, with varieties Ciherang and Sintanur as checks. The trials were conducted at two locations (Bogor and Pusakanagara, West Java) during dry season (DS) 2009 and wet season (WS) of 2009/2010. Each trial was arranged in a randomized block design (RBD) with three replications. Twenty one day-old rice seedlings of each entry were planted in 2 m x 5 m plots at a 20 cm x 20 cm plant spacing, one seedling per hill. Results of the combined analysis showed that strains, environments, and its interaction was significantly affecting the whole characters of NPTR. High variabilities were observed on genetic, heritability, and genetic advance characters among the new rice type lines, particularly on the number of productive tillers, total number of grains per panicle, and total grain yield. These three characters were positively correlated with rice grain yield, and could be used as indicators for superior lines.

Abstract

Good male sterile lines should have high and stable sterility, as well as have good characters of flowers that enhances the outcrossing ability. Research was conducted at Sukamandi field station during DS of 2010 to study the character and flowering behaviour and their genetic variability among the new CMS lines. Five new CMS lines were tested in the field in three replicates of randomized complete block design. The results showed that CMS derived from the Wild Abortive, Kalinga and Gambiaca all were early flowering. The new CMS lines have better flower characters than that of IR58025A, including the following: bigger stigma, higher stigma exsertion percentage, wider degree of glume opening, and longer duration of glume opening. Better flowering behaviour enhanced the ability of outcrossing, and resulted in seed set obtained from CMS lines, up to 25.90%; the traditional IR58025A CMS line produced seed set of only 2.98%. The significant positive correlation was found between seed set with stigma width (r = 0.44*), with stigma exsertion (r = 0.54*); and with degree of glume opening of male sterile lines (r = 0.42*); also with filament length (r = 0.47*) and degree of glume opening of maintainer line (r = 0.57**). Those characters are important for the seed set of CMS lines in the hybrid seed production.
