Abstract

Pasak bumi (Eurycoma longifolia) has the potential to be developed as antihypertensive, antipyretic, aphrodisiacs and health supplements. The use of E. longifolia as a traditional medicine needs to be pursued in the form of more effective and appropriate formulation. The capsule preparations are easy to make and cancover the bitter taste of E. longifolia. Clinical trials in this study use design pre-post treatment in healthy humans. Subjects used were male - healthy men and healthy women who met inclusion criteria and were subjected with formulated capsule for 14 days. The study resulted capsule formula comprising of the ethanolic extract of E. longifolia 300 mg, vivapur 101 300 mg, 58 mg maydis starch, aerosil 3%, talc 2%, and Mg stearate 1%. The results showed that the capsule of E. longifolia did not affect the value of heart rate, respiration rate, body temperature and weight (P > 0.05), based on paired t-test, but they causes a decrease in blood pressure of healthy human. The ethanol extract of E. longifolia caused vasodilation of blood vessels that can be used in antihypertensive therapy.

Abstract

Research of radical scavenging activity of sambiloto (Andrographis paniculata Nees) and patikan kebo (Euphorbia hirta L) ethanolic extracts and their combination toward 2,2-Diphenyl-1-Picrylhidrazyl (DPPH) has conducted. Subjects of this experiment comprise sambiloto ethanolic extract, patikan kebo ethanolic extract, dechlorophyllated extract of sambiloto and combinations among those three. Dechlorophyllation by electrocoagulation method is done to reduce chlorophyll content thereby increasing the level of active compound in the sambiloto extract. The results of dechlorophyllation showed that only total flavonoid content in the extract of sambiloto increased, meanwhile the radical scavenging activity results showed that patikan kebo ethanolic extract has the lowest IC50 value (22,36 µg/mL) compared to sambiloto ethanolic extract (IC50 value 499,03 µg/mL). Extract combination may increase DPPH radical scavenging activity of the sambiloto extract (IC50 value 76,06 µg/mL) but not higher than that of patikan kebo single extract.

Abstract

Rhizome of bangle (Zingiber cassumunar Roxb.) is one of the materials that are traditionally used in the traditional preparation to maintain skin from redness. The content of chemical compound in bangle rhizomes has many benefits, including inhibition of the inflammatory processes. This study aims to determine the topical anti-inflammatory activity of ethanolic extract of bangle rhizomes in mice induced by carrageenan. Thirty five female Swiss mice were used in this study. Animals were divided into 7 group of five: normal group, carrageenan 2% group, carrageenan 2% group plus control of basis (Biocream®), carrageenan 2% group plus 2.5% hydrocortisone cream, and carrageenan 2% group plus cream of ethanolic extract of bangle rhizomes with the concentration of 1%, 2.5% and 5% w/w. Carrageenan 2% was injected subcutaneously in all treatment group except normal group. The skinfold thickness of mice was measured every 1 hour for 6 hours after carrageenan injection. Results of this study shows that there is a significant differences between treatment groups (p <0.05) compared to carrageenan 2% treated group. To conclude, ethanolic extract of bangle rhizomes has topical anti-inflammatory in mice induced by carrageenan.

Abstract

The roots of Eurycoma longifolia, Jack has a lot of beneficial activity on human health. The use of Eurycoma longifolia, Jack roots as a traditional medicine should be made in the form of a dosage that is more effective, safe, and had no side effects, especially on hematology. Easy preparations are capsule. This study aimed at identifying the effect of ethanol extract capsule of Eurycoma longifolia, Jack roots on hematology of healthy volunteers. Healthy volunteers were assigned in the study, each of which were 10 male and 10 female who met the inclusion criteria. Both groups were given ethanol extract capsule of Eurycoma longifolia, Jack roots for 14 days, with the dose of 300 mg extract once a day, after meal, at night. Hematology examination was performed on day 0; 14 and 42. Results were compared statistically using Repeated ANOVA and Friedman tests. The result of the study indicated that the average value of hematology at the examination day of 0; 14; and 42, there were significant differences in MCV and leukocyte parameters in healthy male volunteers (p<0.05). MCH, LED and monocyte parameters in healthy female volunteers had a significant difference of (p<0.05). However, the parameter values were still within the normal range. Monitoring results showed that some of the side effects were frequent urinary, dizziness, nausea, sweating, insomnia, and increasing appetite on healthy male volunteers. Meanwhile, on healthy female volunteers, there were frequent urinary, insomnia, constipation. Having ethanol extract capsule of the Eurycoma longifolia, Jack roots did not affect the hematology parameters of healthy male and female volunteers, but it caused some side effects.

Abstract

The cytochrome P-450 (CYP) and glutathione S-transferase (GST) enzyme systems may influence the biological effects of carcinogens. As such, these enzymes may predict the developmental risk of breast cancer, as well as be potential targets for chemoprevention. The purpose of this study was to compare the expression of CYP1A1 and GST* between treatment groups. Expression of CYP1A1 and GST* was quantified in liver tissue from 18 female Sprague Dawley rats aged 40 days, which randomly divided into six treatment groups. Those are base line group (without DMBA and ethanolic extract treatment), DMBA induced cancer group, the other two groups was administrated by DMBA after treated by ethanolic extract in two different doses, 300 mg /kg BW and 750 mg/kg BW. The last two groups were given two doses of extract ethanolic only, without initiated of DMBA. The ingestion of the extract was carried for three weeks and the ingestion of DMBA was performed twice in the third week. The expression CYP1A1 and GST was quantified by immunohistochemistry. CYP1A1 expression was significantly higher (P < 0.05) in cancer group (DMBA) as compared with other groups. On the other hand, GST expression was lower in cancer group (P < 0.05). Result of this study demonstrated that ethanolic extract leaves of G. procumbens in 300 mg/kg BW dose could inhibit CYP1A1 stronger than are other and induced GST* level. Has effect on ethanolic extract leaves of G. procumbens has ability in played role of as blocking agent in preventing initiation stage of carsinogenesis, therefore it should be taken into account for chemopreventing agent in mammary carsinogenesis.Key words : Gynura procumbens, breast cancer, Chemopreventive, CYP1A1, GSTμ

Abstract

ABSTRACT Indonesia is a country with large plant biodiversity with medicinal properties, such as “sukun” (Artocarpus altilis) or known as “breadfruit”. Breadfruit is a woody evergreen plant that has been used traditionally for various purposes, including medication. The fruit is rich in carbohydrates and fibers as a food source. The leaf and cortex are the most widely used for treating various diseases and other health benefits. This article aimed to present a comprehensive review on the potency of breadfruit from the perspective of ethnobotany, phytochemistry, pharmacology, and toxicology. The data in this narrative review was obtained from the scientific journals in the databases of Google Scholar, PubMed, Scopus, and ScienceDirect. Other credible sources, such as textbooks, student thesis, and patents were also used to support the main data. Based on the literature study, breadfruit has been used empirically in Indonesia as a medicinal herb. The scientific data of breadfruit showed antiinflammatory, antiplatelet, antioxidant, antiatherosclerosis, antihyperlipidemic, antimalaria, antidiabetic, cardioprotective, and anticancer activities. Breadfruit contains terpenoids, flavonoids, alkaloids, and phenolics as bioactive compounds. However, the unique compounds are geranylated and prenylated flavonoids such as cycloartenol, artonin V, and cyclomulberin. These compounds are distributed in the leaf, cortex, wood, and fruit. Limited data is available regarding the toxicology profile of breadfruit. Breadfruit leaves ethanol extract did not show any significant toxic effects in the animal experiments. However, the toxicity of the water extract is unclear, and thus, needs to be investigated to ensure its safety. Keywords: Artocarpus communis, bioactivity, chemical constituents, ethnopharmacology ABSTRAK Indonesia dikenal sebagai negara yang kaya akan keragaman tumbuhan berpotensi obat, diantaranya sukun (Artocarpus altilis). Sukun merupakan tanaman berkayu yang secara tradisional dimanfaatkan untuk berbagai keperluan termasuk pengobatan. Buah sukun mengandung karbohidrat dan serat sebagai sumber pangan. Daun dan batang sukun merupakan bagian yang paling banyak dimanfaatkan dalam pengobatan dan kesehatan. Reviu artikel ini bertujuan untuk mengkaji secara komprehensif potensi sukun dari sudut pandang etnobotani, fitokimia, farmakologi, dan toksikologi. Artikel narrative review ini ditulis berdasarkan data yang diperoleh dari kajian literatur hasil penelitian yang ada di basis data Google Scholar, PubMed, Scopus, dan ScienceDirect. Beberapa sumber pustaka lain seperti buku, naskah tugas akhir dan paten juga digunakan untuk memperkaya penulisan. Hasil kajian literatur sukun menunjukkan bahwa tanaman ini memiliki riwayat empiris digunakan sebagai obat tradisional di Indonesia. Hasil penelitian ilmiah menunjukkan sukun memiliki aktivitas antiinflamasi, antiplatelet, antioksidan, antiatherosklerosis, antihiperlipi-demia, antimalaria, antidiabetes, kardioprotektif, dan antikanker. Sukun mengandung senyawa terpenoid, flavonoid, alkaloid, dan senyawa fenolik. Senyawa khas dari tumbuhan genus Artocarpus ini adalah flavonoid dengan gugus geranil atau prenil, misalnya sikloartenol, artonin V, dan siklomulberin. Senyawa tersebut tersebar dalam daun, kulit kayu, batang, dan buah. Data terkait profil toksikologi sukun masih terbatas. Ekstrak etanol daun sukun tidak menunjukkan efek toksik pada hewan uji. Namun, ekstrak airnya belum memiliki profil toksikologi yang jelas sehingga perlu dilakukan pengujian untuk memastikan keamanannya. Abstract

The use of co-chemotherapy agent was needed since there were some toxicities on the normal tissues caused by the use of doxorubicin. The root of Eurycoma longifolia Jack, a part of plant, has a potential activity as co-chemotherapy. This study aimed to determine the effect of co-chemotherapy of ethyl acetate fraction of E. longifolia Jack roots and doxorubicin against cell proliferation activity by AgNOR method and determine expression of Bax protein of breast tissue in rats induced by DMBA. The rats were divided into five groups: I (baseline), II (DMBA 20 mg/kgBW), III (DMBA, doxorubicin 1.12 mg/kg), IV (DMBA, fraction of 100 mg/kg), and V (DMBA, doxorubicin, fractions). All the rats were sacrificed at week 16. Their breast tissue was evacuated. Cell proliferation and expression was observed using AgNOR method and immunohistochemistry, respectively. Results showed the percentage of p-AgNOR obtained by the healthy group, DMBA group, DMBA+Doxorubicin group, DMBA+ethyl acetate fraction of E. longifolia Jack root group, and DMBA+Doxorubicin+Ethyl acetate fraction of E. longifolia group were 00%, 14.672.11%, 1.831.21%, 6.832.03%, and 4.080.95%, respectively. The percentage of Bax expression obtained by the healthy group, DMBA group, DMBA+Doxorubicin group, DMBA+ethyl acetate fraction of E. longifolia Jack root group, and DMBA+Doxorubicin+Ethyl acetate fraction of E. longifolia group were 68.821.52%, 26.863.25%, 44.492.06%, 80.923.27%, and 78.704.87%, respectively. Based on the results, it was concluded that co-chemotherapy agent of ethyl acetate fraction of E. longifolia Jack roots and Doxorubicin could inhibit proliferation and trigger apoptosis through Bax expression in breast tissue of rats induced by DMBA.

Abstract

Lipid metabolism disorders can lead to hyperlipidemia that triggers atherosclerosis. This study aimed to identify the potential of standardized ethanolic extract of red spinach (Amaranthus tricolor L.) to prevent hyperlipidemia by referring to the reduction of triglyceride level and total cholesterols in male Sprague-Dawley rats. A total of 30 experimental animals was put into 6 groups, including normal, positive control (0.9 mg/kgBW of simvastatin), negative control, treatment I (200 mg/kgBW of extract), treatment II (400 mg/kgBW of extract), and treatment III (800 mg/kgBW of extract). Preventive therapy and positive control were administered from day 1 to day 67. Hyperlipidemia was induced by feeding pure lard and duck yolk to the rats twice daily from day 8 to day 67. Determination of triglyceride level and total cholesterols was conducted on day 0 and day 67. Abstract

Plants can be sources of immunomodulatory agents. Piper crocatum Ruiz & Pav. (red betel) leaves have been used to heal many diseases, especially to increase endurance. The aim of this research was to investigate the imunomodulatory effect of methanolic leaf extracts of Piper crocatum Ruiz & Pav. and its fractions by in vitro phagocytic macrophage activity test. The result showed that methanolic extract of Piper crocatum Ruiz & Pav. 75μg/mL could stimulate phagocytic macrophage activity equal to 100μg/mL product-X® that contained Echinacea extract. Separation of the extract by vacuum liquid chromatography using gradient solvent n-hexane - ethyl acetate yielded 5 fractions. The second fraction showed the highest activity. It contained alkaloid and terpenoid, while the extract also contained flavonoid and essential oil. Alkaloid and/or terpenoid could be the compound in the leaves of Piper crocatum Ruiz & Pav. that responsible increasing phagocytic macrophage activity.

Abstract

The root of pasak bumi (Eurycoma longifolia, Jack) is one of plant from Indonesia known as aphrodisiac. This study was conducted to evaluate the influence of pasak bumi standardized extract to testosterone level and histopathological changes of the testes in male Wistar rats. Sample were 50 adult male rats aged 3 4 months were divided into 5 groups. Group I (control) was given distilled water, group II (positive control), was given testosterone (Andriol ®), group III, IV, and V were each given a standardized extract of the roots of pasak bumi doses of 50, 100, and 200 mg/kg of body weight, respectively. The extract was given orally twice a day for six days and forty nine days and then testes was taken out on 7th and 50th day. Testosterone level was assayed on 7th and 50th day by the ELISA methods. The results were analyzed using one way ANOVA, and post hoc Dunnet (2-sided) (p < 0.05). The data of this study shows that treatment with water extract of pasak bumi root influence the number of Leydig cells that would also increase the concentration of testosterone in Wistar male rats positively at a dose of 100 mg/kg and 200 mg/kg body weight.
