Abstract

ABSTRACTSugarcane cultivars tolerant to drought stress are needed for specific location. In vitro selection can be used to obtain plants tolerant to drought stress through regeneration of callus in the culture medium. The purpose of this study was to obtain regenerants from irradiated callus on the stress medium. The experimental design was a completely randomized design with two factors. The first factor was the regeneration medium (RG) added with PEG 6000, i.e., 0%, 5%, 10% and 15%, while the second factor was the dose of gamma ray irradiation i.e. 0 krad, 5 and 10 krad, and 15 krad. There were 12 treatments, each treatment was repeated 18 times and each repetition was a culture bottles planted with three clums of callus 10 mm diameter. RG medium was the MS medium added with 0.5 mg L-1 BAP, Kinetin 0.1 mg L-1 and IBA 1.0 mg L-1. The higher the concentration of PEG, the less regenerants were produced. Regenerants could be generated from the selection medium PEG up to 15%. Several shoots still produced from callus irradiated with 10 krad. Selection medium with PEG up to 10% decreased the precentage of albino regenerants. The gamma ray irradiation dose and the concentration of PEG significanly affected the number of  roots produced.Keywords: drought tolerant, in vitro, polyethylene glycol, plantlets, albino

Abstract

Several bacterial pathogens can cause diseases of potato. Ralstonia solanacearum and Erwinia carotovora are those of the world’s most important diseases of potato, especially in tropical climates. This experiment was aimed to obtain putative potato cultivars having good tolerance to bacterial wilt caused by R. solanacearum and soft rot caused by E. carotovora. Two adopted potato cultivars in Indonesia, cv. Atlantic (2n=4X=48) and cv. Granola (2n=4X=48), were used as parents. The experiment was arranged in a Randomized Complete Design. Test for tolerance was perfomed in vitro using BF15 as susceptible control and Solanum stenotonum as tolerance control. The result showed that there was high diversity of tolerance level to both R. solanacearum and E. carotovora. Some clones showed good tolerance level as compared to cv. Atlantic and cv. Granola, but none showed that as compared to S. stenotonum.

Abstract

Endosperm quantity in kopyor coconut determines the quality of kopyor coconut. However, data of the best parental based on their progeny test is unavailable yet. In this study, we employed polycross approach and SSR molecular marker to estimate general combining ability of the best pollen contributor for the breeding purpose of this trait. Totally 151 kopyor coconuts were collected from the naturally open-pollinated population of kopyor Tall coconut Agom Lampung. The harvested nuts were rated for their endosperm quantity (scored 1-9). The analysis identified 31 paternal trees of the 53 progenies. The TJ-185 with the highest GCA has been considered as a promising paternal tree for development of high endosperm quantity of kopyor coconut.Keywords: combining ability, molecular marker, paternal trees, polycross, tall coconut

Abstract

Penelitian ini bertujuan untuk memperoleh protokol yang tepat dalam menginduksi kalus dan embrio somatik tanaman jambu biji merah.  Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan, Departemen Agronomi dan Hortikultura, Fakultas Pertanian, Institut Pertanian Bogor pada bulan September 2012 sampai dengan bulan Maret 2013. Penelitian tersusun atas dua percobaan yaitu percobaan pertama merupakan induksi kalus dan percobaan kedua merupakan induksi embrio somatik. Hasil penelitian menunjukkan bahwa media dengan perlakuan zat pengatur tumbuh R1 (5 mg l-1 2.4-D), R3 (5 mg l-1 2.4-D + 1 mg l-1 BAP) dan R6 (2.5 mg l-1 2.4-D + 2 mg l-1 2-iP) merupakan perlakuan terbaik untuk menginduksi kalus kompak pada 4 minggu setelah tanam (MST), sedangkan untuk menginduksi kalus remah didapatkan media dengan perlakuan zat pengatur tumbuh terbaik yaitu R5 (5 mg l-1 2.4-D + 2 mg l-1 2-iP). Embrio somatik primer terbentuk pada perlakuan R2E4 (2.5 mg l-1 2.4-D dipindahkan ke 1 mg l-1 2.4-D + 2 mg l-1 2-iP) dan perlakuan R5E4 (5 mg l-1 2.4-D + 2 mg l-1 2-iP dipindahkan ke 1 mg l-1 2.4-D + 2 mg l-1 2-iP) masing-masing pada 12 dan 16 MST.

Abstract

Terung adalah tanaman yang mudah terserang oleh banyak jenis penyakit. Salah satu usaha yang dapat dilakukan untuk meningkatkan ketahanan terhadap penyakit tersebutadalah dengan mengintroduksi sifat ketahanandari kerabat liarnya melalui fusi protopas. Penelitian ini dilakukan untuk mempelajari keragaan in vitro dan in vivo hibrida somatik hasil fusi protoplas Solanum melongena cv. Dourga dengan Solanum torvum Sw.  Penelitian ini merupakan penelitian faktor tunggal dengan tujuh taraf klon, yaitu Solanum melongena cv. Dourga (SM), Solanum torvum Sw. (ST), dan 5 klon hibrida somatik (SMST1,  SMST2, SMST3, SMST4, and SMST5) yang disusun menggunakan Rancangan Acak Lengkap (RAL). Peubah yang diamati antara lain tinggi tanaman, jumlah daun, panjang daun, panjang tangkai daun, jumlah akar, panjang akar, jumlah buku, warna batang, warna tangkai daun, jumlah duri pada permukaan daun, dan bentuk lobus daun. Pada kondisi in vitro, pertumbuhan Solanum torvum Sw. berbeda sangat nyata dengan kelima klon hibrida somatik. Klon SMST1, SMST2, dan SMST4 menunjukkan pertumbuhan yang lebih baik  dari klon SMST3 dan SMST5. Pada kondisi in vivo, klon-klon yang diuji memiliki kekerabatan yang lebih dekat dengan Solanum torvum Sw.

Abstract

Cultivars Granola  dan Atlantic are the best cultivars so far for farmers, due to it adaptation in Indonesia. Cv Granola is resistant to several important diseases, but the quality is low, thus only produce for vegetable purposes. Cv Atlantic is the best cultivar, in term of productivity and quality. Cv Atlantic is suitable for industry purposes, susceptible to several important diseases. Crossing between both cultivars expected to produce high yielding cultivars and adapted to Indonesian environment. The method used was crossing and germinated the seed then selected in vitro. Each germinated seed was propagated in vitro and considered as clones. In vitro selection was performed through vigour, bacterial wilt and soft rot test, as well as micro tuber production. The result of selectionwas then verified in the field. From thousands of seed germinated producing 12 selected clones, 7 clones of them were showed better then their parent in term of tuberization and micro tuber production. The clones were Atnola 1, Atnola 5, Atnola 3, Atnola 10, Atnola 12, Atnola 24, and Atnola 26. Four clones showed level of resistance better than the parent, namely Atnola 3, Atnola 5, Atnola 8, and Atnola 10. The result of field test showed that the seven clones produced better tuber than the parent Atlantic dan Granola, which were Atnola 5 dan Atnola 10 showed vigour, tuber weight, and better level of resistance, thus will be the candidate of high yielding cultivars.

Abstract

The objective of this research was to study shallot bulb formation on few concentrations of growth retardant succinic acid daminozid hydrazide (SAOH). Completely Randomized Design with 2 factors were used in this experiment. The first factor was four concentrations of SAOH (0, 30, 60 and 90 ppm) and second was two cultivars of shallot (Bima Juna and Kuning Tablet). The cultivars did not give significant effect to total number of leaf, shoot, root, number and weight of bulb, diameter of bulb, and height of plantlets. While SADH concentrations gave very significant effect to number of leaf, but not significant to number of root, number and weight of bulb, diameter of bulb and height of plantlets. Combinations of the two factors only gave significant effect to number of leaf and shoot but not significant to number of root, number and weight of bulb and height of plantlets.   Keywords: bulb formation, SAOH, shallot

Abstract

In supporting the replacement of potato varieties with the varieties that are adaptive to drought, effective and fast methods are needed. One of which is through in vitro screening of drought-tolerant genotypes. The aims of this research were to determine the appropriate concentration of sorbitol as a critical limit in the in vitro screening process of drought-tolerant genotypes, as well as to determine vegetative characteristics that could be used as indicators of in vitro selection and also to obtain in vitro drought-tolerant genotypes. To achieve these objectives, we used single-nodal explants of seven genotypes cultured for six weeks on MS media added with four levels of sorbitol concentration. Of the four treatments, sorbitol level that could distinguish susceptible and tolerant plants was 0.2 M sorbitol and at this concentration, the characteristics that showed significant interactions were plant height and root length, so that both characteristics could be used as selection indicators for the analysis of tolerance tests for the seven genotypes. The results of tolerance test using the analysis of relative decline and stress-susceptibility index (SSI) showed that PKHT4 and PKHT6 were the genotypes that were tolerant in vitro. Abstract

ABSTRACTLilium is usually propagated vegetatively by using bulb scales  or bulbs. Based on the totipotency ability  of  every  parts  of  plant,  it  is  possible  to  regenerate  them  into  plantlets.  The  objective  of  the experiment was to find out micropropagation technique of lily  by  using petal  of  three cultivars of liliumas explants. Different stages  of developing flower buds was used in this research. The best medium for callus induction was Murashige & Skoog (MS)  + 0.3  mg  l-1TDZ. The stage of flower bud  development D2  (diameter flower bud was 2.15 cm and 7 cm  in  length) of  ‘Frutty Pink’  cultivar  was  more responsiveon callus induction media than other cultivars (‘Sorbon’ and ‘Purple Diamond’).Key words: TDZ (thidiazuron),lilium, callus induction, flower, explant.ABSTRAKLilium  biasanya  diperbanyak  secara  vegetatif  dengan  umbi  sisik  atau bulb.  Berdasarkan  pada kemampuan totipotensi dari setiap bagian tanaman, dimungkinkan untuk meregenerasikan tanaman dari berbagai bagian tanaman menjadi planlet. Tujuan percobaan adalah menemukan teknik mikropropagasi lili  menggunakan  petal  tiga  kultivar  lili  sebagai  eksplan.  Stadia  perkembangan  bunga  yang  berbeda digunakan  dalam  penelitian  ini.  Media  yang  terbaik  untuk  induksi  kalus  adalah  media  Murashige  & Skoog (MS) +0.3 mg  l-1TDZ. Stadia perkembangan tunas bunga D2  dengan diametertunas bunga 2.15 cm  dan panjang bunga 7 cm  dari kultivar  ‘Frutty Pink’  lebih responsif dalam induksi   kalus dibanding kultivar lainnya (‘Sorbon’ dan ‘Purple Diamond’).Kata kunci: TDZ (thidiazuron), Lilium, induksi kalus, bunga, eksplan.

Abstract

ABSTRACTIn order to enhance the multiplication of  Protocorm Like Bodies (PLBs)  of Spathoglottis  plicataorchid  in vitro, several combinations of vitamin, sucrose concentration, and cytokinin were tested.   Thisexperiment was arranged in Completely Randomized Design (CRD) with two factors. The first factor was combination  of  vitamin  composition  and  sugar  concentrations  in  the medium  which  consisted  of  four different  combinations  (J1 = vitamin  MS + sucrose 30 g  L-1, J2 = vitamin B5 + sucrose 30 g  L-1, J3= vitamin  MS  + sucrose  40  g  L-1,  J4  =  vitamin  B5  +  sucrose  40  g  L-1).  The  second factor  was  seven combinations  of  cytokinin types and its  concentrations (S0 = without cytokinin (control), S1 =    20  μM BA, S2 = 40  μM BA, S3 = 20 μM kinetin,   S4 = 40  μM  kinetin, S5 = 75  ml L-1 coconut milk, and S6 = 150 ml L-1 coconut milk).  PLBs  were used as  explant  and were  grown on MS  solid medium containing various vitamin, sucrose, and cytokinin combinations  as mentioned above.  Growth and multiplication of PLBs were based on  the number of PLBs per explant, number of plantle ts per explant, number of roots formed, plantlet height and visual appearance and performa nce of the observations at 6  MST.  The best growth  and  multiplication  of  PLBs  orchid S.  plicata  was  produced  on  MS  medium  modified  with  B5 vitamins  and 30 g  L-1 sucrose, followed by those and  on MS medium enriched  with  75 ml  L-1 coconut milk and on MS medium supported by 20 μM BA.Key words : B5, benzyl adenine, coconut water, in vitro , orchid, sugar concentration,  vitamin compositionABSTRAKPercobaan ini bertujuan untuk mendapatkan formulasi kom posisi vitamin dan konsentrasi gula medium,serta jenis dan  konsentrasi sitokinin terbaik dalam menginduksi pertumbuhan dan  multiplikasi  lini klon PLBsanggrek Spathoglottis plicata dalam jumlah yang besar  secara  in vitro. Percobaan ini menggunakan Rancangan Acak Lengkap (RAL) Faktorial.  Eksplan yang digunakan adalah PLBs  yang ditanam pada  medium Murashige dan  Skoog  (MS)  yang  telah   dilakukan  modifikasi  sesuai  dengan  perlakuan.  Faktor  pertama  adalah  formulasi komposisi vitamin dan konsentrasi gula yang terdiri dari empat macam formulasi yaitu J1 = vitamin  MS + gula 30 g L-1, J2 = vitamin B5 + gula 30 g  L-1, J3= vitamin MS + gula 40 g L-1, J4 = vitamin B5 + gula 40 g  L-1. Faktor kedua adalah penambahan sitokinin (3 jenis dengan 2 taraf konsentrasi) yang terdiri dari 7 kombinasi perlakuan yaitu S0 = tanpa sitokinin (kontrol), S1 = BA 20 μM, S2 = BA 40 μM, S3 = kinetin 20 μM,  S4 = kinetin 40 μM, S5 = air kelapa 75 ml L- 1, dan S6 = air kelapa 150 ml  L-1. Pertumbuhan dan multiplikasi PLBs anggrek S. plicataterbaik  dihasilkan   pada medium  MS  dengan  modifikasi  vitamin  B5  dan  konsentrasi  gula  30  g  L-1dan  pada medium MS dengan penambahan air kelapa 75 ml L- 1 serta pada medium MS dengan penambahan BA 20  μMdengan  kriteria  jumlah  PLBs akhir dan  jumlah  planlet  akhir  tertinggi,  jumlah  akar  dan  tinggi  tanaman serta penampilan visual hasil pengamatan pada 6 minggu setelah tanam.Kata kunci : air kelapa, anggrek, B5, benzyl adenin, in vitro, komposisi vitamin, konsentrasi gula
